Sifat Fisiko-Kimia Dendeng Daging Giling Terkait Cara Pencucian (Leaching) Dan Jenis Daging Yang Berbeda
Dendeng is a typical Indonesian jerked meat commonly made from sliced or ground beef with 2 mm thickness. This research was conducted to study the effect of kind of meat and leaching method on pH, toughness, protein, fat content and peroxide value (PV) of ground jerked meat. The experiment was set up in a completely randomized factorial design with three replications. The first factor was three levels of leaching methods (no washing, washed in 1.5 x 1.5 x1.5 cm size and washed minced meat). The second factor was kind of meat namely horse meat, lamb and beef. The results indicated that neither interaction effect between leaching and kind of meat nor effect of leaching were significant on all characteristics observed. Kind of meat influenced significantly on the pH, toughness, fat content and PV of dendeng (P < 0.01)
A Novel Single Nucleotide Polymorphism in Exon 4 of Insulin-Like Growth Factor-1 Associated with Production Traits in Bali Cattle
Insulin-like growth factor-1 (IGF-1) is one of the gene candidates that can be used in selection strategy by using DNA markers (marker assisted selection). Gene candidate strategy is a molecular biology techniques to identify quantitative trait loci directly, with the assumption that genetic variation associated to quantitative trait variation. This study was designed to identify any new mutations in exon 4 that can cause the IGF-1 gene polymorphism and then affect the production traits on Bali cattle. Single nucleotide polymorphism (SNP) discovery was conducted by using the direct sequencing technique. Genetic variation of the genes candidate was identified by using PCR-RFLP technique. The results of this study indicate the presence of a new SNP in exon 4 of IGF-1 gene caused by the T/C transition, which can be identified using Rsa1 restriction enzyme. Genotypic polymorphism of IGF-1/Rsa1 has a significant influence on birth weight, weaning weight and average daily gain of Bali cattle. CC genotype had a birth weight rate, weaning weight and average daily gain of: 15.64±1.83; 83.15±9.00, and 0.439±0.07 respectively, higher than the TT and CT genotype. IGF-1/Rsa1 can be used as a genetic marker for selection of birth weight, weaning weight, and daily body weight gain

Polymorphism of Stearoyl-CoA Desaturase (SCD1) Gene in Indonesian Local Cattle
Stearoyl-Coa desaturase (SCD1) gene is one of genes that involves in converting saturated fatty acids to unsaturated fatty acids. SNP at exon 5 in SCD1 gene that changes amino acid valine to alanine (V293A) has an influence to meat fatty acid composition. The aim of this research was to analyze SCD1 gene polymorphisms based on SNP V293A at exon 5 of three Indonesian local cattle. The identification of SCD1 gene polymorphisms was done by using 98, 20 and 7 DNA sample from Ciamis, Bali/Banteng, and Ongole Grade (PO) cattle, respectively. PCR_RFLP method with AciI enzim was carried out to identify SNP Val293Ala. Allelic frequencies and heterozygosity value were analyzed by using POPGENE32. The result showed that SCD1 gene at Ciamis local cattle and PO cattle were polymorphic. Their frequencies were 74.5% and 71.4% for T and 25.5% and 28.6% for C, respectively. There were three genotypes on Ciamis local cattle i.e TT, CT and CC with their frequencies were 52%, 44.9% and 3.10%, respectively. There were two genotypes on PO cattle i.e TT and CT with their frequencies were 42.9% and 57.1%, respectively. Meanwhile, SCD1 gene in Bali cattle was monomorphic. Heterozygosity value of SCD1 gene in Ciamis and PO cattle were 0.38 and 0.44, respectively. Their heterozigosities were categorized as medium
Analisis Hubungan Keragaman Gen SCD1 (Stearoyl CoA Desaturase) dengan Komposisi Asam lemak Daging Sapi Lokal di Ciamis
SCD1 merupakan kandidat gen yang berperan dalam mengkonversi asam lemak jenuhmenjadi tidak jenuh, dan rasio keduanya menentukan kualitas daging. Single NucleotidePolymorphism (SNP) V293A pada gen SCD1 yang mengubah asam amino Valina menjadiAlaninaberpengaruh terhadap komposisi asam lemak daging. Penelitian ini bertujuan mengidentifikasi keragaman gen SCD1 berdasarkan SNP V293A, dan hubungannya dengan kandungan asam lemak pada sapi lokal di Ciamis. Penelitian ini menggunakan 14 sampel daging dan DNA dengan genotipe yang berbeda. Metode PCR-RFLPdengan enzim Aci1digunakan untuk mendeteksi SNP V293A pada exon 5. Hubungan keragaman gen SCD1 dengan komposisi asam lemak daging dianalisis dengan uji T. Hasil penelitian ini menunjukkan gen SCD1 pada sapi lokal Ciamis polimorfik, terdapat dua alel, yaitu alel T dan C dengan tiga genotipe, yaitu TT, CT dan CC. SNP V293A pada gen SCD1 tidak berpengaruh terhadap komposisi asam lemak daging sapi lokal di Ciamis Jawa Barat.Kata kunci : asam lemak daging, PCR-RFLP, Sapi lokal Ciamis, SCD1
Hubungan Keragaman Gen Leptin dengan Kualitas Fisik Daging Sapi Lokal Di Ciamis (Relationship between Leptin Gene Diversity with Physical Quality of Local Beef In Ciamis)
Leptin terlibat dalam berbagai proses fisiologispembentukan lemak. Keragaman gen Leptin karena adanya SNP Arg25Cys pada exon 2, mempunyai hubungan dengan deposisi lemak, yang dapat mempengaruhi kualitas daging. Penelitian ini bertujuan untuk mengetahui keragaman gen leptin dan hubungannya dengan kualitas fisik daging sapi lokal di Ciamis Jawa Barat. Penelitian ini menggunakan 14 sampel daging bagian udamaru yang diperoleh dari peternakan rakyat.Sampel DNA diisolasi dari daging. Amplifikasi DNA menggunakan PCR, dan penentuan genotipe dianalisis dari hasil sequencing produk PCR. Hasil penelitian menunjukkan Gen Leptin pada sapi lokal Ciamis bersifat polimorfik, terdapat tiga alel, yaitu C, T (memiliki mutasi Arg25Cys) dan H (memiliki mutasi Arg25His) serta terdapat tiga genotipe CC, CT dan CH. Perbedaan genotipe gen Leptin tidak berpengaruh terhadap kualitas fisik daging sapi lokal di Ciamis.Kata kunci: Kualitas daging, Lepti
Production and physical composition of Priangan male sheep carcass supplemented by Pasak Bumi peptide (Eurycoma longifolia Jack)
Pasak Bumi (Eurycoma longifolia Jack/ELJ) is frequently claimmed to increase expression of masculinity, including inducing the lean growth. This study was aimed to determine the effect of ELJ peptide supplementation on yield and physical composition of lamb carcasses. Sixteen Priangan rams about 1 year old with average weight of 30.43±1.41 kg were maintained in individual cages. The experimental rams were divided into 4 treatments i.e. T0, T1, T2, and T3. In T0, T1 and T2 treatments, the experimental rams were administered: 0, 1.5, and 3 mg ELJ peptide/kg BW, respectively. In the T3 treatment, the experimental ram were administered 1 mg/kg commercial product eurypeptide®. The ELJ peptide and commercial product were administered first in the morning prior to feeding. The experiment was conducted in a completely randomized design of four treatments and four replications. Data were covariance analyzed with an empty body weight as covariate for carcass weight and the right carcass weight as covariate for carcasses physical composition. The results showed that the weights of dissection body fat of T2 and T3 were lower than T0 (P < 0.05) and T1 (P < 0.01). Fat weight reduction was found in all parts of the carcass and more apparent in the fore saddle. Lean weights were higher ( P < 0.05) in T3 and T2 as compared to T1 and T0. The pattern of lean weight differed among treatments in the wholesale cuts that follow the total lean weight eventhough it was not significant. ELJ peptide treatment did not significantly affect either bone weight or wholesale cuts weight. In conclusion, T2 and T3 can improve carcass composition with equal ability
