    BACKGROUND: It has been hypothesised that an upregulation of the neuropeptide substance P (SP) and its preferred receptor, the neurokinin-1 receptor (NK-1 R), is a causative factor in inducing tenocyte hypercellularity, a characteristic of tendinosis, through both proliferative and antiapoptotic stimuli. We have demonstrated earlier that SP stimulates proliferation of human tenocytes in culture. AIM: The aim of this study was to investigate whether SP can mediate an antiapoptotic effect in tumour necrosis factor-α (TNF-α)-induced apoptosis of human tenocytes in vitro. RESULTS: A majority (approximately 75%) of tenocytes in culture were immunopositive for TNF Receptor-1 and TNF Receptor-2. Exposure of the cells to TNF-α significantly decreased cell viability, as shown with crystal violet staining. TNF-α furthermore significantly increased the amount of caspase-10 and caspase-3 mRNA, as well as both BID and cleaved-poly ADP ribosome polymerase (c-PARP) protein. Incubation of SP together with TNF-α resulted in a decreased amount of BID and c-PARP, and in a reduced lactate dehydrogenase release, as compared to incubation with TNF-α alone. The SP effect was blocked with a NK-1 R inhibitor. DISCUSSION: This study shows that SP, through stimulation of the NK-1 R, has the ability to reduce TNF-α-induced apoptosis of human tenocytes. Considering that SP has previously been shown to stimulate tenocyte proliferation, the study confirms SP as a potent regulator of cell-turnover in tendon tissue, capable of stimulating hypercellularity through different mechanisms. This gives further support for the theory that the upregulated amount of SP seen in tendinosis could contribute to hypercellularity

    THE ECONOMIC VALUE OF HIKING: FURTHER CONSIDERATIONS OF OPPORTUNITY COST OF TIME IN RECREATIONAL DEMAND MODELS

    The paper tests two alternative specifications for the opportunity cost of time in travel cost models. The standard travel cost survey design is enriched to include a contingent valuation type question about peoples' willingness to accept compensation to forgo a precisely defined recreational experience. It is hypothesized that individually revealed value of time more appropriately reflects the opportunity costs of time associated with a particular aspect of recreation than the wage rate which measures the trade-off between work and leisure generally. The results seem to indicate a better overall fit for the models with the elicited value of individual consumer's time than for the models with the more traditional hourly earnings (wage rates). The importance of the corrected measurement of the opportunity cost time is illustrated by showing that estimated consumer surpluses based on two different value of time measurements differ significantly.Recreation demand, Travel cost model, Value of time, Resource /Energy Economics and Policy,

    SCHEDULING AMPHIBIOUS CONNECTORS TO DELIVER MULTIPLE COMMODITIES

    Across the amphibious warfighting community, planners need to quickly develop schedules to deliver multiple supply commodities during ship-to-shore operations. We extend the single-commodity models underpinning the Marine Expeditionary Unit (MEU) Amphibious Connector Scheduler (MACS) tool to allow amphibious connectors to deliver multiple commodities, such as fuel, food, water, and ammunition. This extension provides amphibious planners with a flexible and versatile decision tool, which is much more operationally relevant compared to the one-commodity variant. Our primary contribution is a temporal network flow model that computes the optimal number of round trips for each connector configuration from the seabase to the shore to achieve both fast delivery and few connector trips. We formulate the optimization problem as a mixed integer linear program, and develop a linear program approximation to solve large-sized problems. Our approach generates several different schedules with different strengths, so the decision maker can choose the one that best meets mission requirements. This study is prompted by the current debate on whether the California Legislature should enact a moratorium on the death penalty during the 1963 General Session. In making their judgments on that grave question the members of the Legislature need to, consider and evaluate all available factual information. Unfortunately we have observed that, except for the moral question, the discussion on all sides has been noted chiefly for an abundance of undocumented opinions and conclusions, and by an almost total absence of relevant and material facts.
