    Candida albicans Induces Selective Development of Macrophages and Monocyte Derived Dendritic Cells by a TLR2 Dependent Signalling

    As TLRs are expressed by haematopoietic stem and progenitor cells (HSPCs), these receptors may play a role in haematopoiesis in response to pathogens during infection. We have previously demonstrated that in in vitro defined conditions inactivated yeasts and hyphae of Candida albicans induce HSPCs proliferation and differentiation towards the myeloid lineage by a TLR2/MyD88 dependent pathway. In this work, we showed that C. albicans invasive infection with a low virulence strain results in a rapid expansion of HSPCs (identified as LKS cells: Lin− c-Kit+ Sca-1+ IL-7Rα−), that reach the maximum at day 3 post-infection. This in vivo expansion of LKS cells in TLR2−/− mice was delayed until day 7 post- infection. Candidiasis was, as expected, accompanied by an increase in granulopoiesis and decreased lymphopoiesis in the bone marrow. These changes were more pronounced in TLR2−/− mice correlating with their higher fungal burden. Accordingly, emigration of Ly6Chigh monocytes and neutrophils to spleen was increased in TLR2−/− mice, although the increase in macrophages and inflammatory macrophages was completely dependent on TLR2. Similarly, we detected for the first time, in the spleen of C. albicans infected control mice, a newly generated population of dendritic cells that have the phenotype of monocyte derived dendritic cells (moDCs) that were not generated in TLR2−/− infected mice. In addition, C. albicans signalling through TLR2/MyD88 and Dectin-1 promotes in vitro the differentiation of Lin− cells towards moDCs that secrete TNF-α and are able to kill the microorganism. Therefore, our results indicate that during infection C. albicans can directly stimulate progenitor cells through TLR2 and Dectin-1 to generate newly formed inflammatory macrophages and moDCs that may fulfill an essential role in defense mechanisms against the pathogen

Conclusion Surface radiotherapy is a treatment modality that should be taken into account in patients with breast cancer who present bleeding as a consequence of local tumor growth, given that this is a treatment comfortable for the patient, non invasive and increases the quality of patient’s life

    O objetivo do presente texto foi apresentar o estado da arte dos estudos sobre as modalidades esportivas de combate (MEC). Inicialmente, é destacada a relevância destas modalidades, tanto do ponto de vista histórico, quanto em relação à sua representatividade em competições internacionais, como os Jogos Olímpicos. Também são apresentadas as áreas mais comuns de atuação do profissional de Esporte nas MEC, bem como as iniciativas de organização de eventos, publicações, grupos de estudos e instituições científicas direcionadas às MEC. Posteriormente, estudos com possibilidade de aplicação em diferentes áreas de intervenção - preparação física, técnica e tática, gestão e organização - por parte do profissional do Esporte foram destacados. Finalmente, perspectivas de novos estudos e aspectos relacionados à preparação profissional são evidenciados.The objective of the present text was to present the state of art of studies related to combat sports (CS). Initially, it is presented the relevancy of these sports, from a historical perspective to its representativeness in international competitions, as the Olympic Games. The common areas of Sport's professional intervention, the initiatives of events, publications, research groups and scientific organizations related to the CS are also presented. Additionally, studies applied to different areas of Sport's professional intervention - physical training, technical and tactical preparation, management and organization - were also illustrated. Finally, perspectives of new studies and aspects related to professional preparation were reported

    Comparative study of MLPA-FISH to determine DNA copy number alterations in neuroblastic tumors

    Neuroblastoma tumor cells show complex combinations of genetic aberrations, and to date many different methods have been used for their detection. To apply genome-wide techniques, such as Multiplex Ligation–dependent Probe Amplification (MLPA), in routine diagnosis their validation is appropriate and necessary. DNA copy number alterations in 129 cases of neuroblastic tumors were detected using MPLA, and the results validated by Fluorescence In Situ Hybridization (FISH) (MYCN gene, 1p36, 11q and 17q). Kappa index values showed very good concordance between the two techniques in detecting homogeneous MYCN amplification (1); 11q deletion (0.908) and 17q gain (0.922). The validation results showed that MLPA is a highly efficient technique for diagnosis based on the genetic aberrations in relevant regions in neuroblastoma, showing a high concordance with FISH

    Incidence and prognostic impact of U2AF1 mutations and other gene alterations in myelodysplastic neoplasms with isolated 20q deletion

    This work was supported by 2019 3rd Grant of the Spanish Group of Myelodysplastic Syndromes (GESMD).Background: In myelodysplastic neoplasms (MDS), the 20q deletion [del(20q)] is a recurrent chromosomal abnormality that it has a high co-occurrence with U2AF1 mutations. Nevertheless, the prognostic impact of U2AF1 in these MDS patients is uncertain and the possible clinical and/or prognostic differences between the mutation type and the mutational burden are also unknown. Methods: Our study analyzes different molecular variables in 100 MDS patients with isolated del(20q). Results & Conclusions: We describe the high incidence and negative prognostic impact of U2AF1 mutations and other alterations such as in ASXL1 gene to identify prognostic markers that would benefit patients to receive earlier treatment
