    Bioluminescence-based assays for assessing antibacterial properties of medicinal plants

    Extended research has been carried out to clarify the ecological role of plant secondary metabolites (SMs). Although their primary ecological function is self-defence, bioactive compounds have long been used in alternative medicine or in biological control of pests. One single plant may contain a wide variety of bioactive compounds, making analytics rather costly. The total bactericide capacity can be quantified by either microbiological or ecotoxicological methods. Here, the principle and possible applications of a specific bacterial bioluminescence inhibition based ecotoxicological assay are reviewed

    Trends in major lower limb amputations related to peripheral arterial disease in Hungary : A nationwide study (2004-2012)

    ObjectivesTo assess the trends of peripheral arterial disease associated major lower limb amputation in Hungary over a 9 year period (2004–2012) in the whole Hungarian population.MethodsThis was a retrospective cohort study employing administrative health care data. Major amputations were identified in the entire Hungarian population during a 9 year period (2004–2012) using the health care administrative data. Direct standardization was used to eliminate the potential bias induced by the different age and sex structure of the compared populations. For external direct standardization, the ESP 2013 was chosen as reference.Results76,798 lower limb amputations were performed. The number of major amputations was 38,200; these procedures affected 32,084 patients. According to case detection, 50.4% of the amputees were diabetic. The overall primary amputation rate was 71.5%. The annual crude and age adjusted major amputation rates exhibited no significant long-term pattern over the observation period. The major lower limb amputation incidence for the overall period was 42.3/105 in the total population and 317.9/105 in diabetic population.ConclusionAccording to this whole population based study from Hungary, the incidence of lower limb major amputation is high with no change over the past 9 years. An explanation for this remains to be determined, as the traditional risk factors in Hungary do not account for it. The characteristics of major amputation (the rate of primary amputation, the ratio of below to above knee amputation and the age of the affected population) underline the importance of screening, early detection, improved vascular care and an optimal revascularization policy.     Assessment of the credibility of public websites about medicinal herbs

    In recent decades, there has been a growing interest in the use of herbs and herbal medicinal products, both in developing and developed countries. While electronic medium has become a more and more important tool for presenting information about health-related issues, several studies demonstrated that the internet often contains inaccurate and/or misleading information. In our study we assessed 30 Hungarian websites and 2 cellphone applications intended for public use and evaluated the quality and credibility of the information presented about medicinal plants recommended. It was found that websites showed very diverse safety: most websites gave mixed information, that is, some medicinal herbs and their potential hazard were properly described, while others were not. There were, however, websites, which completely missed to give information about any potential hazard. As credibility of public websites can be in most cases questioned, it is strongly recommended for potential users to consult more than one source of information

    Vibrational spectra of C60C8H8 and C70C8H8 in the rotor-stator and polymer phases

    C60-C8H8 and C70-C8H8 are prototypes of rotor-stator cocrystals. We present infrared and Raman spectra of these materials and show how the rotor-stator nature is reflected in their vibrational properties. We measured the vibrational spectra of the polymer phases poly(C60C8H8) and poly(C70C8H8) resulting from a solid state reaction occurring on heating. Based on the spectra we propose a connection pattern for the fullerene in poly(C60C8H8), where the symmetry of the C60 is D2h.     The first biosimilar approved for the treatment of osteoporosis

    To demonstrate the clinical comparability between RGB-10 (a biosimilar teriparatide) and the originator, a comparative pharmacokinetic trial was conducted. The study was successful in establishing bioequivalence. Marketing authorisation for RGB-10 (Terrosa®) was granted by the European Medicines Agency in 2017.Teriparatide, the first bone anabolic agent, is the biologically active fragment of human parathyroid hormone. The imminent patent expiry of the originator will open the door for biosimilars to enter the osteology market, thereby improving access to a highly effective, yet prohibitively expensive therapy.Subsequent to establishing comparability on the quality and non-clinical levels between RGB-10, a biosimilar teriparatide, and its reference product (Forsteo®), a randomised, double-blind, 2-way cross-over comparative study (duration: four days) was conducted in 54 healthy women (ages: 18 to 55 years) to demonstrate the pharmacokinetic/pharmacodynamic (PK/PD) equivalence and comparable safety of these products. Extents of exposure (AUC0-tlast) and peak exposure (Cmax), as measured by means of ELISA, were evaluated as co-primary PK endpoints, and serum calcium levels, as measured using standard automated techniques, were assessed for PD effects. Safety was monitored throughout the study.The 94.12% CIs for the ratio of the test to the reference treatments, used due to the two-stage design (85.20-98.60% and 85.51-99.52% for AUC0-tlast and Cmax, respectively), fell within the 80.00-125.00% acceptance range. The calcium PD parameters were essentially identical with geometric mean ratios (GMRs) of 99.93% and 99.87% for AUC and Cmax, respectively. Analysis of the safety data did not reveal any differences between RGB-10 and its reference.Based on the high level of similarity in the preclinical data and the results of this clinical study, marketing authorisation for RGB-10 (Terrosa®) was granted by the European Medicines Agency (EMA) in 2017
