    Schwann cells and mesenchymal stem cells in laminin- or fibronectin-aligned matrices and regeneration across a critical size defect of 15 mm in the rat sciatic nerve

    OBJECTIVE Artificial nerve guides are being developed to substitute for autograft repair after peripheral nerve injuries. However, the use of conduits is limited by the length of the gap that needs to be bridged, with the success of regeneration highly compromised in long gaps. Addition of aligned proregenerative cells and extracellular matrix (ECM) components inside the conduit can be a good strategy to achieve artificial grafts that recreate the natural environment offered by a nerve graft. The purpose of this study was to functionalize chitosan devices with different cell types to support regeneration in limiting gaps in the rat peripheral nerve. METHODS The authors used chitosan devices combined with proteins of the ECM and cells in a rat model of sciatic nerve injury. Combinations of fibronectin and laminin with mesenchymal stem cells (MSCs) or Schwann cells (SCs) were aligned within tethered collagen-based gels, which were placed inside chitosan tubes that were then used to repair a critical-size gap of 15 mm in the rat sciatic nerve. Electrophysiology and algesimetry tests were performed to analyze functional recovery during the 4 months after injury and repair. Histological analysis was performed at the midlevel and distal level of the tubes to assess the number of regenerated myelinated fibers. RESULTS Functional analysis demonstrated that SC-aligned scaffolds resulted in 100% regeneration success in a 15-mm nerve defect in this rat model. In contrast, animals that underwent repair with MSC-aligned constructs had only 90% regeneration success, and those implanted with acellular bridges had only 75% regeneration success. CONCLUSIONS These results indicate that the combination of chitosan conduits with ECM-enriched cellular gels represents a good alternative to the use of autografts for repairing long nerve gaps

    A Semantic-Based Approach to Attain Reproducibility of Computational Environments in Scientic Work ows: A Case Study

    Reproducible research in scientic work ows is often addressed by tracking the provenance of the produced results. While this approach allows inspecting intermediate and nal results, improves understanding, and permits replaying a work ow execution, it does not ensure that the computational environment is available for subsequent executions to reproduce the experiment. In this work, we propose describing the resources involved in the execution of an experiment using a set of semantic vocabularies, so as to conserve the computational environment. We dene a process for documenting the work ow application, management system, and their dependencies based on 4 domain ontologies. We then conduct an experimental evaluation sing a real work ow application on an academic and a public Cloud platform. Results show that our approach can reproduce an equivalent execution environment of a predened virtual machine image on both computing platforms

    Population Response of Three Quail Species to Habitat Restoration in South Texas

    Maintaining and increasing usable space is paramount for maintaining and increasing wild quail. Aside from weather and other factors that can temporarily reduce densities, range-wide, no factor has as much influence on quail populations as the amount of habitat present across the landscape. In the field of quail management, ‘‘bad news’’ is the norm, as many articles begin by explaining how a select species has declined. Here we provide good news and use 4 empirical examples of population increases for 3 quail species following creation of usable space and restoration of patch connectivity. From 2008–2014, a suite of independent projects aimed at increasing usable space for quail was initiated across South Texas. These projects included 3 focused on northern bobwhites (Colinus virginianus), 1 focused on scaled quail (Callipepla squamata), and 1 landowner-executed project focused on Montezuma quail (Cyrtonyx montezumae). Through the correction of attributes limiting habitat, bobwhite numbers increased 22–378% across 2 studies. On one particular study site, native grassland restoration resulted in the colonization of bobwhites from adjacent areas to 1 bobwhite/1.2 ha from nearly 0. For scaled quail in South Texas, reducing buffelgrass standing crop via grazing from about 2,240 kg/ha to 1,008 kg/ha resulted in the recolonization of a previously unoccupied habitat patch to a density of 1 scaled quail/6 ha. Finally, clearing monotypic stands of the invasive native plant, ash juniper (Juniperus ashei) in the Edwards Plateau of Texas, resulted in the reestablishment of native grasses and forbs and thus recolonization by Montezuma quail from nearby areas. Although habitat restoration and management can be a painstaking and lengthy process, addressing limiting factors to quail occupancy is the only known way to increase wild quail populations. We hope that highlighting these particular studies will provide inspiration to those interested in restoring and increasing quail across the US

    G'3 as the logic of modal 3-valued Heyting algebras

    In 2001, W. Carnielli and Marcos considered a 3-valued logic in order to prove that the schema ϕ ∨ (ϕ → ψ) is not a theorem of da Costa’s logic Cω. In 2006, this logic was studied (and baptized) as G'3 by Osorio et al. as a tool to define semantics of logic programming. It is known that the truth-tables of G'3 have the same expressive power than the one of Łukasiewicz 3-valued logic as well as the one of Gödel 3-valued logic G3. From this, the three logics coincide up-to language, taking into acccount that 1 is the only designated truth-value in these logics. From the algebraic point of view, Canals-Frau and Figallo have studied the 3-valued modal implicative semilattices, where the modal operator is the well-known Moisil-Monteiro-Baaz Δ operator, and the supremum is definable from this. We prove that the subvariety obtained from this by adding a bottom element 0 is term-equivalent to the variety generated by the 3-valued algebra of G'3. The algebras of that variety are called G'3-algebras. From this result, we obtain the equations which axiomatize the variety of G'3-algebras. Moreover, we prove that this variety is semisimple, and the 3-element and the 2-element chains are the unique simple algebras of the variety. Finally an extension of G'3 to first-order languages is presented, with an algebraic semantics based on complete G'3-algebras. The corresponding soundness and completeness theorems are obtained

    Identification of the first structurally validated covalent ligands of the small GTPase RAB27A

    Rab27A is a small GTPase, which mediates transport and docking of secretory vesicles at the plasma membrane via protein–protein interactions (PPIs) with effector proteins. Rab27A promotes the growth and invasion of multiple cancer types such as breast, lung and pancreatic, by enhancing secretion of chemokines, metalloproteases and exosomes. The significant role of Rab27A in multiple cancer types and the minor role in adults suggest that Rab27A may be a suitable target to disrupt cancer metastasis. Similar to many GTPases, the flat topology of the Rab27A-effector PPI interface and the high affinity for GTP make it a challenging target for inhibition by small molecules. Reported co-crystal structures show that several effectors of Rab27A interact with the Rab27A SF4 pocket (‘WF-binding pocket’) via a conserved tryptophan–phenylalanine (WF) dipeptide motif. To obtain structural insight into the ligandability of this pocket, a novel construct was designed fusing Rab27A to part of an effector protein (fRab27A), allowing crystallisation of Rab27A in high throughput. The paradigm of KRas covalent inhibitor development highlights the challenge presented by GTPase proteins as targets. However, taking advantage of two cysteine residues, C123 and C188, that flank the WF pocket and are unique to Rab27A and Rab27B among the >60 Rab family proteins, we used the quantitative Irreversible Tethering (qIT) assay to identify the first covalent ligands for native Rab27A. The binding modes of two hits were elucidated by co-crystallisation with fRab27A, exemplifying a platform for identifying suitable lead fragments for future development of competitive inhibitors of the Rab27A-effector interaction interface, corroborating the use of covalent libraries to tackle challenging targets

    Vibrational properties of CdGa2S4 at high pressure

    [EN] Raman scattering measurements have been performed in cadmium digallium sulphide (CdGa2S4) with defect chalcopyrite structure up to 25 GPa in order to study its pressure-induced phase transitions. These measurements have been complemented and compared with latticedynamics ab initio calculations including the TO-LO splitting at high pressures in order to provide a better assignment of experimental Raman modes. In addition, experimental and theoretical Gruneisen parameters have been reported in order to calculate the molar heat capacity and thermal expansion coefficient of CdGa2S4. Our measurements provide evidence that CdGa2S4 undergoes an irreversible phase transition above 15 GPa to a Raman-inactive phase, likely with a disordered rock salt structure. Moreover, the Raman spectrum observed on downstroke from 25 GPa to 2 GPa has been attributed to a new phase, tentatively identified as a disordered zinc blende structure, that undergoes a reversible phase transition to the Raman-inactive phase above 10 GPa.
