    Structural investigation of the Arabidopsis thaliana circadian clock

    Plants, like most organisms, have developed elaborate mechanisms for anticipating periodic environmental changes. The circadian clock allows an organism to adapt its metabolic, developmental and physiological processes to coincide with favourable environmental conditions. At the centre of the Arabidopsis thaliana clock, linking environmental inputs and driving the overt biological rhythm is a central oscillator that consists of multiple interlocked transcriptional/translational negative feedback loops. What is known about the structure of the central oscillator comes primarily from genetic analysis. Less clear, is how putative oscillator proteins perform their perceived functions in circadian rhythm maintenance. Described are the cloning, expression and purification of clock-associated proteins; TOCI-PRR, ELF4, LUX and LIPl. Purified ELF4 was subjected to unsuccessful crystallisation trials, probably due to its intrinsically unstructured nature. A truncated form of LIP 1 was shown to be an active OTPase, representing the first example of an active OTPase in the plant clock. In addition, a protocol for the production of ssDNA aptamers has been developed (against SRRl), which can be used to replace antibody-based experimentation. The work presented discusses the difficulty in obtaining the novel, plant-specific proteins in quantities required for crystallisation, and suggests alternative methods for structural and biochemical analysis of these proteins. Moreover, this thesis combines experimental data with a range of Bioinformatic tools to aid design for subsequent biochemical expression, purification and crystallisation trials

    Forest Focus Monitoring Database System - Technical Report 2005 Level II Data

    Forest Focus (Regulation (EC) No 2152/2003) is a Community scheme for harmonized, broad-based, comprehensive and long-term monitoring of European forest ecosystems. Under this scheme the monitoring of air pollution effects on forests is carried out by participating countries on the basis of the systematic network of observation points (Level I) and of the network of observation plots for intensive and continuous monitoring (Level II). According to Article 15(1) of the Forest Focus Regulation Member States shall annually, through the designated authorities and agencies, forward to the Commission geo-referenced data gathered under the scheme, together with a report on them by means of computer telecommunications and/or electronic technology. For managing the data JRC has implemented a Forest Focus Monitoring Database System. This Technical Report presents the results obtained from all processing stages (data reception, validation checks ¿ compliance, conformity, uniformity) for submitted data referring to the monitoring year 2005. This report presents the results at the end of the processing phase after data have been re-submitted in 2007. It presents in addition a brief comment on the data status for each NFC, for the reporting year, with respect to the parameter assessed and including analyses of spatial variability of data and temporal trends of parameters.JRC.DDG.H.7-Land management and natural hazard

    Forest Focus (Regulation (EC) No 2152/2003 ) is a Community scheme for harmonised, broad-based, comprehensive and long-term monitoring of European forest ecosystems. It concentrates in particular on protecting forests against air pollution and fire. To supplement the monitoring system, Forest Focus stipulates the development of new instruments relating to soil monitoring, carbon sequestration, biodiversity, climate change and protective functions of forests. Under this scheme the monitoring of air pollution effects on forests is carried out by participating countries on the basis of the systematic network of observation points (Level I) and of the network of observation plots for intensive and continuous monitoring (Level II). The monitoring activity continues from the network and plots established and implemented under Council Regulation (EEC) No 3528/86 .and Regulations (EEC) No 1696/87 and (EC) No 1091/94 . The monitoring programme of air pollution effects is linked to International Cooperative Programme on Assessment and Monitoring of Air Pollution Effects on Forest (ICP Forests). ICP Forests reports to the working Group on Effects of the Convention of the Long-Range Trans-boundary Air Pollution (CLRTAP) of the United Nations Economic Commission for Europe (UN-ECE). Forest Focus Article 15(1) stipulates that the Member States shall annually, through the designated authorities and agencies, forward to the Commission geo-referenced data gathered under the scheme, together with a report on them by means of computer telecommunications and/or electronic technology. For managing the data DG JRC has implemented a Forest Focus Monitoring Database System. The system was developed and realized under contract by a Consortium, coordinated by I-MAGE Consult with Nouvelles Solutions Informatiques s.a. (NSI) as consortium partner and the Bundesforschungsanstalt für Forst- und Holzwirtschaft (BFH) as sub-contractor. The designated authorities and agencies submitted annually to DG Joint Research Centre of the European Commission their observations made on the network of observation plots for intensive and continuous monitoring (Level II).     Abstract The northern white rhino (Ceratotherium simum cottoni) is functionally extinct with only two females left alive. However, cryopreserved material from a number of individuals represents the potential to produce additional individuals using advanced reproductive and genetic rescue technologies and perhaps eventually a population to return to their native range. If this could and were done, how should it be done responsibly and thoughtfully. What issues and questions of a technical, bioethical, and societal nature will it raise that need to be anticipated and addressed? Such issues are explored in this article by an interdisciplinary team assembled to provide context to the northern white rhino project of the San Diego Zoo Global

    The effects of foam rolling on ankle dorsiflexion range of motion in healthy adults: A systematic literature review

    Background: Reduced ankle dorsiflexion is associated with lower limb injury and dysfunction, with static stretching mostly used to increase ankle range of motion. Foam rolling is an alternative intervention, shown to immediately increase ankle range of motion, while the long-term application has conflicting evidence. Aims: To assess the effects of single and multiple foam rolling interventions on ankle dorsiflexion range of motion in healthy adults and appraise the methodological quality of the included studies. Design: Systematic literature review. Methods: Five electronic databases were systematically searched to identify randomised controlled trials reporting the effects of foam rolling on ankle dorsiflexion. Data was extracted from studies that met the inclusion criteria and independently appraised by each reviewer using the PEDro scale. Results: Thirty-two articles were identified; six studies included foam rolling compared to other interventions on ankle dorsiflexion range of motion. Five of the six studies reported a significant increase (p < 0.05) in ankle dorsiflexion within groups compared to baseline measurements, after a single foam rolling intervention. One study found a significant within group increase in long-term effects after foam rolling on ankle dorsiflexion over seven weeks. The mean PEDro score for all studies was 6/10 indicating a high-quality level of evidence. Conclusion: There is strong evidence suggesting that foam rolling may be effective in increasing range of motion in a healthy adult population in the short term up to 30 min; however, definitive conclusions on long-term effects cannot be drawn due to a lack of evidence, with further research recommended
