    <p>Abstract</p> <p>Background</p> <p>Contagious bovine pleuropneumonia (CBPP) is a mycoplasmal disease caused by <it>Mycoplasma mycoides </it>subsp. <it>mycoides </it>SC (<it>Mmm</it>SC). Since the disease is a serious problem that can affect cattle production in parts of Africa, there is a need for an effective and economical vaccine. Identifying which of the causative agent's proteins trigger potentially protective immune responses is an important step towards developing a subunit vaccine. Accordingly, the purpose of this study was to determine whether phage display combined with bioinformatics could be used to narrow the search for genes that code for potentially immunogenic proteins of <it>Mmm</it>SC. Since the production of IgG2 and IgA are associated with a Th<sub>1 </sub>cellular immune response which is implicated in protection against CBPP, antigens which elicit these immunoglobulin subclasses may be useful in developing a subunit vaccine.</p> <p>Results</p> <p>A filamentous phage library displaying a repertoire of peptides expressed by fragments of the genome of <it>MmmSC </it>was constructed. It was subjected to selection using antibodies from naturally- and experimentally-infected cattle. Mycoplasmal genes were identified by matching the nucleotide sequences of DNA from immunoselected phage particles with the mycoplasmal genome. This allowed a catalogue of genes coding for the proteins that elicited an immune response to be compiled. Using this method together with computer algorithms designed to score parameters that influence surface accessibility and hence potential antigenicity, five genes (<it>abc, gapN, glpO, lppB </it>and <it>ptsG</it>) were chosen to be expressed in <it>Escherichia coli</it>. After appropriate site-directed mutagenesis, polypeptides representing portions of each of these proteins were tested for immunoreactivity. Of these five, polypeptides representing expression products of <it>abc </it>and <it>lppB </it>were recognised on immunoblots by sera obtained from cattle during a natural outbreak of the disease.</p> <p>Conclusion</p> <p>Since phage display physically couples phenotype with genotype, it was used to compile a list of sequences that code for <it>Mmm</it>SC proteins bearing epitopes which were recognised by antibodies in the serum of infected animals. Together with the appropriate bioinformatic analyses, this approach provided several potentially useful vaccine or diagnostic leads. The phage display step empirically identified sequences by their interaction with antibodies which accordingly reduced the number of ORFs that had to be expressed for testing. This is a particular advantage when working with <it>Mmm</it>SC since the mycoplasmal codon for tryptophan needs to be mutated to prevent it from being translated as a stop in <it>E. coli</it>.</p

    Access to chloroquine in patients with rheumatic and musculoskeletal diseases attending rheumatology outpatient clinics during the COVID-19 pandemic

    Herbal medicines made from the bark of the Cinchona tree, and later quinine, have been widely used for centuries to treat medical conditions such as tropical malaria. More recently, chloroquine (CQ) and its synthetic derivatives have been used as antimalarials and to treat systemic lupus erythematosus, rheumatoid arthritis, and in the past 14 months or so, COVID-19 pneumonia. Anecdotal evidence and the erratic covering through social media of its potential efficacy in the treatment of COVID-19 pneumonia have resulted in the widespread off-label use of CQ in South Africa and worldwide. This study aimed to show that access to CQ as a chronic medication for rheumatic and musculoskeletal diseases was limited during the COVID-19 pandemic, and that this resulted in an increased incidence of flares in these patients, affecting their morbidity and potentially leading to mortality

    Assessment of the concentration of Cr, Mn and Fe in sediment using laser-induced breakdown spectroscopy

    In the present study, laser-induced breakdown spectroscopy (LIBS) has been applied for the determination of levels of Cr, Mn and Fe in sediment samples and the results have been compared with that of flame-atomic absorption spectroscopy (F-AAS). Fourteen sediment samples were collected from Tinishu Akaki River (TAR), Addis Ababa, Ethiopia. Pellets of 24 mm in diameter and 3-5 mm in thickness were prepared using a manual hydraulic press under a pressure of 8000 psi by mixing finely ground sediment samples with boric acid. The concentrations of Cr, Mn and Fe were successfully determined by Q-switched Nd:YAG laser at 1064 nm. Quantification of these metals was conducted against certified reference materials of sediments and soils. The LIBS results for the elements varied from 0.060–0.707 mg/g for Cr, 0.895–3.64 mg/g for Mn and 26.9–71.8 mg/g for Fe. The F-AAS results varied from 0.061–1.37 mg/g for Cr, 1.24–4.46 mg/g for Mn and 49.0–83.3 mg/g for Fe.     New age constraints on metamorphism, metasomatism and gold mineralisation at Plutonic Gold Mine, Marymia Inlier, Western Australia

    The Plutonic Well Greenstone Belt (PWGB) is located in the Marymia Inlier between the Yilgarn and Pilbara cratons in Western Australia, and hosts a series of major Au deposits. The main episode of Au mineralisation in the PWGB was previously interpreted to have either accompanied, or shortly followed, peak metamorphism in the late Archean at ca 2650 Ma with a later, minor, event associated with the Capricorn Orogeny. Here we present new Pb isotope model ages for sulfides and Rb–Sr ages for mica, as well as a new 207Pb–206Pb age for titanite for samples from the Plutonic Gold Mine (Plutonic) at the southern end of the PWGB. The majority of the sulfides record Proterozoic Pb isotope model ages (2300–2100 Ma), constraining a significant Au mineralising event at Plutonic that occurred >300 Myr later than previously thought. A Rb–Sr age of 2296 ± 99 Ma from muscovite in an Au-bearing sample records resetting or closure of the Rb–Sr system in muscovite at about the same time. A younger Rb–Sr age of 1779 ± 46 Ma from biotite from the same sample may record further cooling, or resetting during a late-stage episode of metasomatism in the PWGB. This could have been associated with the 1820–1770 Ma Capricorn Orogeny, or a late-stage hydrothermal event potentially constrained by a new 207Pb–206Pb age of 1725 ± 26 Ma for titanite in a chlorite–carbonate vein. This titanite age correlates with a pre-existing age for a metasomatic event dated at 1719 ± 14 Ma by U–Pb ages of zircon overgrowths in a sample from the Marymia Deposit. Based on the Pb-isotope data presented here, Au mineralising events in the PWGB are inferred to have occurred at ca 2630, 2300–2100 Ma, during the Glenburgh and Capricorn orogenies, and 1730–1660 Ma. The 2300–2100 Ma event, which appears to have been significant based on the amount of sulfide of this age, correlates with the inferred age for rifting of the Marymia Inlier from the northern margin of the Yilgarn Craton. The texturally-later visible Au may have been deposited during the Glenburgh and Capricorn orogenies

    First Measurements with NeXtRAD, a Polarimetric X/L Band Radar Network

    NeXtRAD is a fully polarimetric, X/L Band radar network. It is a development of the older NetRAD system and builds on the experience gained with extensive deployments of NetRAD for sea clutter and target measurements. In this paper we will report on the first measurements with NeXtRAD, looking primarily at sea clutter and some targets, as well as early attempts at calibration using corner reflectors, and an assessment of the polarimetric response of the system. We also highlight innovations allowing for efficient data manipulation post measurement campaigns, as well as the plans for the coming years with this system

It has been proposed that childhood trauma interferes with normal neurodevelopment, thereby establishing a biological vulnerability to schizophrenia. Poor premorbid adjustment is frequently a precursor to schizophrenia, and may be a manifestation of neurodevelopmental compromise. We investigated the relationship between childhood trauma and premorbid adjustment in 77 patients with first-episode schizophrenia spectrum disorders. We also investigated possible mediating roles for other selected risk factors in the relationship. We found several significant correlations between different trauma types and both social and academic premorbid adjustment from childhood to late adolescence. There were no significant moderating effects for family history of schizophrenia or family history of psychiatric disorder. History of obstetric complications, substance abuse and poor motor coordination weakened some of the associations between childhood trauma and premorbid adjustment, while poor sequencing of motor acts strengthened the association. Our results confirm previous studies indicating an association between childhood trauma and premorbid adjustment. Results indicate a general rather than specific association, apparent with different types of trauma, and affecting both social and academic components of premorbid adjustment across childhood, early and late adolescence.     Density functional study of the actinide nitrides

    The full potential all electron linearized augmented plane wave plus local orbitals (FP-LAPW + lo) method, as implemented in the suite of software WIEN2K, has been used to systematically investigate the structural, electronic, and magnetic properties of the actinide compounds AnN (An = Ac, Th, Pa, U, Np, Pu, Am). The theoretical formalism used is the generalized gradient approximation to density functional theory (GGA-DFT) with the Perdew-Burke-Ernzerhof (PBE) exchange-correlation functional. Each compound has been studied at six levels of theory: non-magnetic (NM), non-magnetic with spin-orbit coupling (NM+SOC), ferromagnetic (FM), ferromagnetic with spin-orbit coupling (FM+SOC), anti-ferromagnetic (AFM), and anti-ferromagnetic with spin-orbit coupling (AFM+SOC). The structural parameters, bulk moduli, densities of states, and charge distributions have been computed and compared to available experimental data and other theoretical calculations published in the literature. The total energy calculations indicate that the lowest energy structures of AcN, ThN, and PaN are degenerate at the NM+SOC, FM+SOC, and AFM+SOC levels of theory with vanishing total magnetic moments in the FM+SOC and AFM+SOC cases, making the ground states essentially non-magnetic with spin-orbit interaction. The ground states of UN, NpN, PuN, and AmN are found to be FM+SOC at the level of theory used in the present computations. The nature of the interactions between the actinide metals and nitrogen atom, and the implications on 5f electron delocalization and localization are discussed in detail.Comment: 5 tables, 12 figure
