Effect of the natural sweetener xylitol on gut hormone secretion and gastric emptying in humans:A pilot dose-ranging study
Sugar consumption is associated with a whole range of negative health effects and should be reduced and the natural sweetener xylitol might be helpful in achieving this goal. The present study was conducted as a randomized, placebo-controlled, double-blind, cross-over trial. Twelve healthy, lean volunteers received intragastric solutions with 7, 17 or 35 g xylitol or tap water on four separate days. We examined effects on: gut hormones, glucose, insulin, glucagon, uric acid, lipid profile, as well as gastric emptying rates, appetite-related sensations and gastrointestinal symptoms. We found: (i) a dose-dependent stimulation of cholecystokinin (CCK), active glucagon-like peptide-1 (aGLP-1), peptide tyrosine tyrosine (PYY)-release, and decelerated gastric emptying rates, (ii) a dose-dependent increase in blood glucose and insulin, (iii) no effect on motilin, glucagon, or glucose-dependent insulinotropic peptide (GIP)-release, (iv) no effect on blood lipids, but a rise in uric acid, and (v) increased bowel sounds as only side effects. In conclusion, low doses of xylitol stimulate the secretion of gut hormones and induce a deceleration in gastric emptying rates. There is no effect on blood lipids and only little effect on plasma glucose and insulin. This combination of properties (low-glycemic sweetener which stimulates satiation hormone release) makes xylitol an attractive candidate for sugar replacement
Monitoring international migration flows in Europe. Towards a statistical data base combining data from different sources
The paper reviews techniques developed in demography, geography and statistics that are useful for bridging the gap between available data on international migration flows and the information required for policy making and research. The basic idea of the paper is as follows: to establish a coherent and consistent data base that contains sufficiently detailed, up-to-date and accurate information, data from several sources should be combined. That raises issues of definition and measurement, and of how to combine data from different origins properly. The issues may be tackled more easily if the statistics that are being compiled are viewed as different outcomes or manifestations of underlying stochastic processes governing migration. The link between the processes and their outcomes is described by models, the parameters of which must be estimated from the available data. That may be done within the context of socio-demographic accounting. The paper discusses the experience of the U.S. Bureau of the Census in combining migration data from several sources. It also summarizes the many efforts in Europe to establish a coherent and consistent data base on international migration.

Decreased expression of breast cancer resistance protein in the duodenum in patients with obstructive cholestasis
Background/Aims: The expression of transporters involved in bile acid homeostasis is differentially regulated during obstructive cholestasis. Since the drug efflux transporter breast cancer resistance protein (BCRP) is known to transport bile acids, we investigated whether duodenal BCRP expression could be altered during cholestasis. Methods: Using real-time RT-PCR analysis we determined mRNA expression levels in duodenal tissue of 19 cholestatic patients. Expression levels were compared to 14 healthy subjects. BCRP protein staining was determined in biopsies of 6 cholestatic and 6 healthy subjects by immunohistochemistry. Results: We found that in patients with obstructive cholestasis mean duodenal BCRP mRNA levels were significantly reduced to 53% and mean protein staining was reduced to 57%. Conclusions: BCRP, a transporter for bile acids and numerous drugs, appears to be down-regulated in the human duodenum during cholestasis. Methods to study splicing from high-throughput RNA Sequencing data
The development of novel high-throughput sequencing (HTS) methods for RNA
(RNA-Seq) has provided a very powerful mean to study splicing under multiple
conditions at unprecedented depth. However, the complexity of the information
to be analyzed has turned this into a challenging task. In the last few years,
a plethora of tools have been developed, allowing researchers to process
RNA-Seq data to study the expression of isoforms and splicing events, and their
relative changes under different conditions. We provide an overview of the
methods available to study splicing from short RNA-Seq data. We group the
methods according to the different questions they address: 1) Assignment of the
sequencing reads to their likely gene of origin. This is addressed by methods
that map reads to the genome and/or to the available gene annotations. 2)
Recovering the sequence of splicing events and isoforms. This is addressed by
transcript reconstruction and de novo assembly methods. 3) Quantification of
events and isoforms. Either after reconstructing transcripts or using an
annotation, many methods estimate the expression level or the relative usage of
isoforms and/or events. 4) Providing an isoform or event view of differential
splicing or expression. These include methods that compare relative
event/isoform abundance or isoform expression across two or more conditions. 5)
Visualizing splicing regulation. Various tools facilitate the visualization of
the RNA-Seq data in the context of alternative splicing. In this review, we do
not describe the specific mathematical models behind each method. Our aim is
rather to provide an overview that could serve as an entry point for users who
need to decide on a suitable tool for a specific analysis. We also attempt to
propose a classification of the tools according to the operations they do, to

Executive functions in preschool children with aggressive behavior: impairments in inhibitory control
The question whether executive function (EF)
deficits in children are associated with conduct problems
remains controversial. Although the origins of aggressive
behavior are to be found in early childhood, findings from
EF studies in preschool children with aggressive behavior
are inconsistent. The current study aimed to investigate
whether preschool children with aggressive behavior show
impairments in EF. From a population-based sample, 82
preschool children who were showing aggressive behavior
as indicated by scores at or above the 93rd percentile on the
Aggressive Behavior Scale of the CBCL 1 1/2-5 were
selected. These children with aggressive behavior were
matched on IQ to a group of typically developing control
children (N=99). Six neuropsychological tasks were administered
to assess set shifting, inhibition, working memory
and verbal fluency. A factor analysis was conducted which
yielded one clear factor: inhibition. Aggressive preschool children showed poorer performance on this inhibition
factor than control children and boys performed worse on
this factor than girls. This association between aggressive
behavior and inhibition deficits was maintained after controlling
for attention problems. In addition, gender differences
in all EFs measured were found with boys exhibiting
more impairment in EF than girls. These findings demonstrate
that preschool children with aggressive behavior show
impairments in inhibition, irrespective of attention problems
Default Network Deactivations Are Correlated with Psychopathic Personality Traits
Background: The posteromedial cortex (PMC) and medial prefrontal cortex (mPFC) are part of a network of brain regions that has been found to exhibit decreased activity during goal-oriented tasks. This network is thought to support a baseline of brain activity, and is commonly referred to as the ‘‘default network’’. Although recent reports suggest that the PMC and mPFC are associated with affective, social, and self-referential processes, the relationship between these default network components and personality traits, especially those pertaining to social context, is poorly understood. Methodology/Principal Findings: In the current investigation, we assessed the relationship between PMC and mPFC deactivations and psychopathic personality traits using fMRI and a self-report measure. We found that PMC deactivations predicted traits related to egocentricity and mPFC deactivations predicted traits related to decision-making. Conclusions/Significance: These results suggest that the PMC and mPFC are associated with processes involving selfrelevancy and affective decision-making, consistent with previous reports. More generally, these findings suggest a link between default network activity and personality traits
Predation Risk Shapes Social Networks in Fission-Fusion Populations
Predation risk is often associated with group formation in prey, but recent advances in methods for analysing the social structure of animal societies make it possible to quantify the effects of risk on the complex dynamics of spatial and temporal organisation. In this paper we use social network analysis to investigate the impact of variation in predation risk on the social structure of guppy shoals and the frequency and duration of shoal splitting (fission) and merging (fusion) events. Our analyses revealed that variation in the level of predation risk was associated with divergent social dynamics, with fish in high-risk populations displaying a greater number of associations with overall greater strength and connectedness than those from low-risk sites. Temporal patterns of organisation also differed according to predation risk, with fission events more likely to occur over two short time periods (5 minutes and 20 minutes) in low-predation fish and over longer time scales (>1.5 hours) in high-predation fish. Our findings suggest that predation risk influences the fine-scale social structure of prey populations and that the temporal aspects of organisation play a key role in defining social systems
A systematic review on integration mechanisms in human and animal health surveillance systems with a view to addressing global health security threats
Lymphatic filariasis and onchocerciasis are neglected tropical diseases (NTDs) targeted for elimination by mass (antifilarial) drug administration. These drugs are predominantly active against the microfilarial progeny of adult worms. New drugs or combinations are needed to improve patient therapy and to enhance the effectiveness of interventions in persistent hotspots of transmission. Several therapies and regimens are currently in (pre-)clinical testing. Clinical trial simulators (CTSs) project patient outcomes to inform the design of clinical trials but have not been widely applied to NTDs, where their resource-saving payoffs could be highly beneficial. We demonstrate the utility of CTSs using our individual-based onchocerciasis transmission model (EPIONCHO-IBM) that projects trial outcomes of a hypothetical macrofilaricidal drug. We identify key design decisions that influence the power of clinical trials, including participant eligibility criteria and post-treatment follow-up times for measuring infection indicators.
