    Other State Circle Site records include: 18AP50, Bordley-Randall Site (area 11), http://hdl.handle.net/1903/11016 ; 18AP55, Donaldson House (area 2), http://hdl.handle.net/1903/11019 ; 18AP61, Public Well Site (area 18), http://hdl.handle.net/1903/11020Archaeological excavations were conducted at State Circle in Annapolis during the fall and winter of 1989-1990 by "Archaeology in Annapolis", a cooperative program between the University of Maryland, College Park and the Historic Annapolis Foundation. Excavation was conducted as part of the undergrounding of public utility wires within State Circle, Francis Street, and School Street. The work was undertaken to satisfy the conditions of compliance as set forth in the Annotated Code of Maryland, Article 83b, sections 5-617 and 5-18. Twenty excavation area were selected for excavation within the project area. Areas were selected based on data gathered during historical background research. Areas were also selected in an attempt to gather information concerning the Baroque town plan designed in 1695 by Royal Governor Francis Nicholson. Three sites previously identified in the project area (18AP22, 18AP28, and 18AP50) were tested. Nine additional sites were discovered during excavation (18AP54, 18AP55, 18AP56, 18AP57, 18AP58, 18AP59, 18AP60, 18AP61, and 18AP62). At least one hand dug 3 ft by 5 ft unit per site was excavated. In all, a total of 23 units were excavated

    This paper describes the development of neural model-based control strategies for the optimisation of an industrial aluminium substrate disk grinding process. The grindstone removal rate varies considerably over a stone life and is a highly nonlinear function of process variables. Using historical grindstone performance data, a NARX-based neural network model is developed. This model is then used to implement a direct inverse controller and an internal model controller based on the process settings and previous removal rates. Preliminary plant investigations show that thickness defects can be reduced by 50% or more, compared to other schemes employed

    Effect of Off-Body Laser Discharge on Drag Reduction of Hemisphere Cylinder in Supersonic Flow

    The interaction of an off-body laser discharge with a hemisphere cylinder in supersonic flow is investigated. The objectives are 1) experimental determination of the drag reduction and energetic efficiency of the laser discharge, and 2) assessment of the capability for accurate simulation of the interaction. The combined computational and experimental study comprises two phases. In the first phase, laser discharge in quiescent air was examined. The temporal behavior of the shock wave formed by the laser discharge was compared between experiment and simulation and good agreement is observed. In the second phase, the interaction of the laser discharge with a hemisphere cylinder was investigated numerically. Details of the pressure drag reduction and the physics of the interaction of the heated region with the bow shock are included. The drag reduction due to this interaction persisted for about five characteristic times where one characteristic time represents the time for the flow to move a distance equal to the hemisphere radius. The energetic efficiency of laser discharge for the case with 50 mJ energy absorbed by the gas is calculated as 3.22

    Nrf2 regulates mass accrual and the antioxidant endogenous response in bone differently depending on the sex and age

    Accumulation of reactive oxygen species (ROS) is an important pathogenic mechanism underling the loss of bone mass and strength with aging and other conditions leading to osteoporosis. The transcription factor erythroid 2-related factor2 (Nrf2) plays a central role in activating the cellular response to ROS. Here, we examined the endogenous response of bone regulated by Nrf2, and its relationship with bone mass and architecture in the male and female murine skeleton. Young (3 month-old) and old (15 month-old) Nrf2 knockout (KO) mice of either sex exhibited the expected reduction in Nrf2 mRNA expression compared to wild type (WT) littermates. Nrf2 deletion did not lead to compensatory increase in Nrf1 or Nrf3, other members of this transcription factor family; and instead, Nrf1 expression was lower in KO mice. Compared to the respective WT littermate controls, female KO mice, young and old, exhibited lower expression of both detoxifying and antioxidant enzymes; young male KO mice, displayed lower expression of detoxifying enzymes but not antioxidant enzymes; and old male KO mice showed no differences in either detoxifying or antioxidant enzymes. Moreover, old male WT mice exhibited lower Nrf2 levels, and consequently lower expression of both detoxifying and antioxidant enzymes, compared to old female WT mice. These endogenous antioxidant responses lead to delayed rate of bone acquisition in female KO mice and higher bone acquisition in male KO mice as quantified by DXA and ÎĽCT, demonstrating that Nrf2 is required for full bone accrual in the female skeleton but unnecessary and even detrimental in the male skeleton. Therefore, Nrf2 regulates the antioxidant endogenous response and bone accrual differently depending on sex and age. These findings suggest that therapeutic interventions that target Nrf2 could be developed to enhance the endogenous antioxidant response in a sex- and age-selective manner

    Isolation of a Highly Thermal Stable Lama Single Domain Antibody Specific for Staphylococcus aureus Enterotoxin B

    <p>Abstract</p> <p>Background</p> <p>Camelids and sharks possess a unique subclass of antibodies comprised of only heavy chains. The antigen binding fragments of these unique antibodies can be cloned and expressed as single domain antibodies (sdAbs). The ability of these small antigen-binding molecules to refold after heating to achieve their original structure, as well as their diminutive size, makes them attractive candidates for diagnostic assays.</p> <p>Results</p> <p>Here we describe the isolation of an sdAb against <it>Staphyloccocus aureus </it>enterotoxin B (SEB). The clone, A3, was found to have high affinity (Kd = 75 pM) and good specificity for SEB, showing no cross reactivity to related molecules such as Staphylococcal enterotoxin A (SEA), Staphylococcal enterotoxin D (SED), and Shiga toxin. Most remarkably, this anti-SEB sdAb had an extremely high Tm of 85°C and an ability to refold after heating to 95°C. The sharp Tm determined by circular dichroism, was found to contrast with the gradual decrease observed in intrinsic fluorescence. We demonstrated the utility of this sdAb as a capture and detector molecule in Luminex based assays providing limits of detection (LODs) of at least 64 pg/mL.</p> <p>Conclusion</p> <p>The anti-SEB sdAb A3 was found to have a high affinity and an extraordinarily high Tm and could still refold to recover activity after heat denaturation. This combination of heat resilience and strong, specific binding make this sdAb a good candidate for use in antibody-based toxin detection technologies.</p
