    Research in development: learning from the CGIAR Research Program on Aquatic Agricultural Systems

    This working paper aims to synthesize and share learning from the experience of adapting and operationalizing the Research in Development (RinD) approach to agricultural research in the five hubs under the The CGIAR Research Program on Aquatic Agricultural Systems. It seeks to share learning about how the approach is working in context and to explore the outcomes it is achieving through initial implementation over 3 ½ years. This learning can inform continuation of agricultural research in the second phase of the CGIAR research programs and will be useful to others aiming to implement research programs that seek to equitably build capacity to innovate in complex social-ecological systems. Each of the chapters in this working paper have shown that RinD has produced a range of outcomes that were often unexpected and broader in scope than might result from other approaches to agricultural research.     A modular digital twinning framework for safety assurance of collaborative robotics

    Digital twins offer a unique opportunity to design, test, deploy, monitor, and control real-world robotic processes. In this paper we present a novel, modular digital twinning framework developed for the investigation of safety within collaborative robotic manufacturing processes. The modular architecture supports scalable representations of user-defined cyber-physical environments, and tools for safety analysis and control. This versatile research tool facilitates the creation of mixed environments of Digital Models, Digital Shadows, and Digital Twins, whilst standardising communication and physical system representation across different hardware platforms. The framework is demonstrated as applied to an industrial case-study focused on the safety assurance of a collaborative robotic manufacturing process. We describe the creation of a digital twin scenario, consisting of individual digital twins of entities in the manufacturing case study, and the application of a synthesised safety controller from our wider work. We show how the framework is able to provide adequate evidence to virtually assess safety claims made against the safety controller using a supporting validation module and testing strategy. The implementation, evidence and safety investigation is presented and discussed, raising exciting possibilities for the use of digital twins in robotic safety assurance

    Re-thinking the Fiscal and Monetary Political Economy of the Green State

    Proponents of the Green State repudiate the historical antipathy to the state from many in the green movement and endorse the pragmatic usage of state capacity and legitimacy to realise environmental protection. This article offers a sympathetic critique of the Green State’s fiscal and monetary institutional design in order to refine the concept further. It will investigate an under-theorised contradiction in the political economy of the Green State; centring upon the operationalisation of an interventionist state, moving beyond economic growth, and deference to the ceteris paribus conventions of state financing. It is argued that the three cannot co-exist harmoniously, given the ramifications of moving beyond growth for the fiscal capacity of the state. Therefore, there is a need to go further than even Eckersley does in re-politicising and challenging capitalist conventions. Specifically, Eckersley’s own critical constructivist approach is invoked to interrogate the capitalist conventions that constitute the constraints surrounding state financing, such as the depoliticised production of money and the viability of debt relations

    Delayed mucosal anti-viral responses despite robust peripheral inflammation in fatal COVID-19

    Background While inflammatory and immune responses to SARS-CoV-2 infection in peripheral blood are extensively described, responses at the upper respiratory mucosal site of initial infection are relatively poorly defined. We sought to identify mucosal cytokine/chemokine signatures that distinguished COVID-19 severity categories, and relate these to disease progression and peripheral inflammation. Methods We measured 35 cytokines and chemokines in nasal samples from 274 patients hospitalised with COVID-19. Analysis considered the timing of sampling during disease, as either the early (0-5 days post-symptom onset) or late (6-20 days post-symptom onset). Results Patients that survived severe COVID-19 showed IFN-dominated mucosal immune responses (IFN-γ, CXCL10 and CXCL13) early in infection. These early mucosal responses were absent in patients that would progress to fatal disease despite equivalent SARS-CoV-2 viral load. Mucosal inflammation in later disease was dominated by IL-2, IL-10, IFN-γ, and IL-12p70, which scaled with severity but did not differentiate patients who would survive or succumb to disease. Cytokines and chemokines in the mucosa showed distinctions from responses evident in the peripheral blood, particularly during fatal disease. Conclusions Defective early mucosal anti-viral responses anticipate fatal COVID-19 but are not associated with viral load. Early mucosal immune responses may define the trajectory of severe COVID-19

    SARS-CoV-2-specific nasal IgA wanes 9 months after hospitalisation with COVID-19 and is not induced by subsequent vaccination

    BACKGROUND: Most studies of immunity to SARS-CoV-2 focus on circulating antibody, giving limited insights into mucosal defences that prevent viral replication and onward transmission. We studied nasal and plasma antibody responses one year after hospitalisation for COVID-19, including a period when SARS-CoV-2 vaccination was introduced. METHODS: In this follow up study, plasma and nasosorption samples were prospectively collected from 446 adults hospitalised for COVID-19 between February 2020 and March 2021 via the ISARIC4C and PHOSP-COVID consortia. IgA and IgG responses to NP and S of ancestral SARS-CoV-2, Delta and Omicron (BA.1) variants were measured by electrochemiluminescence and compared with plasma neutralisation data. FINDINGS: Strong and consistent nasal anti-NP and anti-S IgA responses were demonstrated, which remained elevated for nine months (p < 0.0001). Nasal and plasma anti-S IgG remained elevated for at least 12 months (p < 0.0001) with plasma neutralising titres that were raised against all variants compared to controls (p < 0.0001). Of 323 with complete data, 307 were vaccinated between 6 and 12 months; coinciding with rises in nasal and plasma IgA and IgG anti-S titres for all SARS-CoV-2 variants, although the change in nasal IgA was minimal (1.46-fold change after 10 months, p = 0.011) and the median remained below the positive threshold determined by pre-pandemic controls. Samples 12 months after admission showed no association between nasal IgA and plasma IgG anti-S responses (R = 0.05, p = 0.18), indicating that nasal IgA responses are distinct from those in plasma and minimally boosted by vaccination. INTERPRETATION: The decline in nasal IgA responses 9 months after infection and minimal impact of subsequent vaccination may explain the lack of long-lasting nasal defence against reinfection and the limited effects of vaccination on transmission. These findings highlight the need to develop vaccines that enhance nasal immunity. FUNDING: This study has been supported by ISARIC4C and PHOSP-COVID consortia.     ROSIE: A ROS adapter for a modular digital twinning framework

    As robotic systems become more interactive and complex, there is a need to standardise interfaces and simplify development processes. This is particularly pertinent in the field of manufacturing, where human-robot collaboration is on the increase, but where standards and proprietary software are key barriers to deployment and adoption.In this article we present the ROSIE Adapter, a general-purpose, modular adapter developed in ROS designed to support the creation and connection of industry-ready digital twins. Together with our previous work on the modular CSI digital-twin framework, we demonstrate how the ROSIE Adapter creates a versatile "plug-and-play"interface that simplifies the development of new robotic processes, and improves accessibility to novice users. Furthermore, the adaptor supports integration of intuitive interface devices, such as speech and augmented reality interfaces, which enable more natural collaboration. We describe the adaptor and its use in two real-world applications, demonstrate the ease of use via a three-day hackathon event, and provide results showing the faithfulness of the arising digital twins to their connected physical systems

    A new professionalism for agricultural research for development

    There have been repeated calls for a ‘new professionalism’ for carrying out agricultural research for development since the 1990s. At the centre of these calls is a recognition that for agricultural research to support the capacities required to face global patterns of change and their implications on rural livelihoods, requires a more systemic, learning focused and reflexive practice that bridges epistemologies and methodologies. In this paper, we share learning from efforts to mainstream such an approach through a large, multi-partner CGIAR research program working in aquatic agricultural systems. We reflect on four years of implementing research in development (RinD), the program’s approach to the new professionalism. We highlight successes and challenges and describe the key characteristics that define the approach. We conclude it is possible to build a program on a broader approach that embraces multidisciplinarity and engages with stakeholders in social-ecological systems. Our experience also suggests caution is required to ensure there is the time, space and appropriate evaluation methodologies in place to appreciate outcomes different to those to which conventional agricultural research aspires
