    Hainan sport tourism development—A SWOT analysis

    Hainan, as a popular tourism destination, is well-promoted by the Chinese central government. In particular, both central and local governments encourage Hainan’s sport tourism-related professionals to develop sport tourism as one of the most important tourist activities in Hainan. However, previous research has not reported on Hainan’s sport tourism strengths, weaknesses, opportunities, and threats as a tourism destination or a sports event host. This study uses SWOT analysis to identify the strengths, weaknesses, opportunities, and threats in the context of Hainan’s sport tourism development. A total of 12 dimensions, including branding, culture, finance, infrastructure, location, market, nature, policy, product, specialty, sustainability, and tourist were generated from our data analysis. In addition, a total of five future directions, including emphasizing event-oriented sport tourism, prioritizing sport motivation, identifying major sport tourism markets, making the rational use of sport tourism resources, and nurturing sport culture, are recommended as a result of this study

    Turner’s communitas and non-Buddhists who visit Buddhist temples

    Increasing numbers of people are participating in reflexive forms of spiritual travel. Rites of passage and intensification are becoming voluntary, and so is religion. Arguably, individuals travel to achieve mental and social escape. Their quests to religious sites are self-motivated, not obligated. Because of their mobility, Americans constantly explore new outlets for their spiritual growth, including different metaphysical movements and philosophies. This study focuses on what motivates Americans to visit Buddhist temples, including their desire to explore new ideas or life directions. The results of a survey study (N = 179) conducted in Los Angeles, California indicate that non-Buddhists visit Buddhist temples for stimulus-avoidance and intellectual purposes; to mentally relax and broaden themselves in the holy site. Furthermore, they may pursue a sense of communitas ‒ defined by social relations that are no longer normative, hierarchical and distant, but close and egalitarian. American Buddhist temple visitors are not necessarily Buddhists. As religious options grow and obligations decrease, religion has become secularized. As a consequence, non-religious temple-goers may be seeking Turner’s communitas ‒ a transition away from mundane structures toward a looser commonality of feeling with fellow visitors. They may also seek healing and renewal as well as a higher level of freedom. These individuals may desire to re-structure or re-orient their life-direction through contemplating at Buddhist temples. By examining these new phenomena, this study contributes to the field of religious tourism research; it reveals what motivates Americans to visit Buddhist temples and provides an anthropological explanation for these motivations

    Microvesicles secreted by macrophages shuttle invasion-potentiating microRNAs into breast cancer cells

    <p>Abstract</p> <p>Background</p> <p>Tumor-associated macrophages (TAMs) are alternatively activated cells induced by interleukin-4 (IL-4)-releasing CD4<sup>+ </sup>T cells. TAMs promote breast cancer invasion and metastasis; however, the mechanisms underlying these interactions between macrophages and tumor cells that lead to cancer metastasis remain elusive. Previous studies have found microRNAs (miRNAs) circulating in the peripheral blood and have identified microvesicles, or exosomes, as mediators of cell-cell communication. Therefore, one alternative mechanism for the promotion of breast cancer cell invasion by TAMs may be through macrophage-secreted exosomes, which would deliver invasion-potentiating miRNAs to breast cancer cells.</p> <p>Results</p> <p>We utilized a co-culture system with IL-4-activated macrophages and breast cancer cells to verify that miRNAs are transported from macrophages to breast cancer cells. The shuttling of fluorescently-labeled exogenous miRNAs from IL-4-activated macrophages to co-cultivated breast cancer cells without direct cell-cell contact was observed. miR-223, a miRNA specific for IL-4-activated macrophages, was detected within the exosomes released by macrophages and was significantly elevated in the co-cultivated SKBR3 and MDA-MB-231 cells. The invasiveness of the co-cultivated breast cancer cells decreased when the IL-4-activated macrophages were treated with a miR-223 antisense oligonucleotide (ASO) that would inhibit miR-223 expression. Furthermore, results from a functional assay revealed that miR-223 promoted the invasion of breast cancer cells via the Mef2c-β-catenin pathway.</p> <p>Conclusions</p> <p>We conclude that macrophages regulate the invasiveness of breast cancer cells through exosome-mediated delivery of oncogenic miRNAs. Our data provide insight into the mechanisms underlying the metastasis-promoting interactions between macrophages and breast cancer cells.</p

    Altered spontaneous brain activity during dobutamine challenge in healthy young adults: A resting-state functional magnetic resonance imaging study

    IntroductionThere is a growing interest in exploring brain-heart interactions. However, few studies have investigated the brain-heart interactions in healthy populations, especially in healthy young adults. The aim of this study was to explore the association between cardiovascular and spontaneous brain activities during dobutamine infusion in healthy young adults.MethodsForty-eight right-handed healthy participants (43 males and 5 females, range: 22–34 years) underwent vital signs monitoring, cognitive function assessment and brain MRI scans. Cardiovascular function was evaluated using blood pressure and heart rate, while two resting-state functional magnetic resonance imaging (rs-fMRI) methods—regional homogeneity (ReHo) and amplitude of low-frequency fluctuation (ALFF)—were used together to reflect the local neural activity of the brain. Logistic regression was used to model the association between brain and heart.ResultsResults showed that blood pressure and heart rate significantly increased after dobutamine infusion, and the performance in brain functional activity was the decrease in ReHo in the left gyrus rectus and in ALFF in the left frontal superior orbital. The results of logistic regression showed that the difference of diastolic blood pressure (DBP) had significant positive relationship with the degree of change of ReHo, while the difference of systolic blood pressure (SBP) had significant negative impact on the degree of change in ALFF.DiscussionThese findings suggest that the brain-heart interactions exist in healthy young adults under acute cardiovascular alterations, and more attention should be paid to blood pressure changes in young adults and assessment of frontal lobe function to provide them with more effective health protection management

    Proto-oncogene Src links lipogenesis via lipin-1 to breast cancer malignancy

    Src基因是哺乳动物中发现的第一个原癌基因,其编码的蛋白是一个酪氨酸激酶,在促进乳腺癌、肺癌等诸多肿瘤的发生、进展和恶化中起着重要的作用。在研究中,研究团队发现Src能够承接生长因子和肥胖微环境相关的因子如胰岛素和瘦素的信号,通过直接磷酸化lipin-1,增强其催化合成甘油脂的活性,提高细胞摄入的脂肪酸向甘油脂尤其是磷脂转化。进一步实验表明,Src磷酸化lipin-1能够加速乳腺癌细胞生长,促进小鼠模型中肿瘤的进展和转移。这项研究不但做出了对脂肪合成途径的调控机制的又一重要发现,还揭示了原癌基因Src可以承接癌细胞内外的活化信号,通过lipin-1为媒介重塑癌细胞脂代谢,使得肿瘤细胞具有增殖和转移的优势。该论文揭示了臭名昭著的原癌基因Src通过直接结合并磷酸化lipin-1(一种磷脂酸磷酸化酶,在脂质代谢中具有重要作用),以增强其酶活性,从而加速甘油酯的合成速率,进而促进乳腺癌的发生发展。 该研究由厦门大学生命科学学院、广州医科大学第五附属医院、第四军医大学西京医院和中山大学孙逸仙纪念医院等单位合作完成,厦门大学生命科学学院博士后宋林涛和广州医科大学第五附属医院刘志华教授为该论文的共同第一作者。【Abstract】Increased lipogenesis has been linked to an increased cancer risk and poor prognosis; however, the underlying mechanisms remain obscure. Here we show that phosphatidic acid phosphatase (PAP) lipin-1, which generates diglyceride precursors necessary for the synthesis of glycerolipids, interacts with and is a direct substrate of the Src proto-oncogenic tyrosine kinase. Obesity-associated microenvironmental factors and other Src-activating growth factors, including the epidermal growth factor, activate Src and promote Src-mediated lipin-1 phosphorylation on Tyr398, Tyr413 and Tyr795 residues. The tyrosine phosphorylation of lipin-1 markedly increases its PAP activity, accelerating the synthesis of glycerophospholipids and triglyceride. Alteration of the three tyrosine residues to phenylalanine (3YF-lipin-1) disables lipin-1 from mediating Src-enhanced glycerolipid synthesis, cell proliferation and xenograft growth. Re-expression of 3YF-lipin-1 in PyVT;Lpin1−/− mice fails to promote progression and metastasis of mammary tumours. Human breast tumours exhibit increased p-Tyr-lipin-1 levels compared to the adjacent tissues. Importantly, statistical analyses show that levels of p-Tyr-lipin-1 correlate with tumour sizes, lymph node metastasis, time to recurrence and survival of the patients.     Segmenting the Rural Tourism Market: The Case of Potter County, Pennsylvania, USA

    Because of its importance in the sustainable development of rural communities, rural tourism has been frequently studied as an alternative form of economic development in rural areas. In the United States, many rural communities rely on tourism for economic growth to compensate for declines in manufacturing, agriculture, extraction of natural resources, and population. Although there is an increased interest in developing rural tourism in many countries, there is little information regarding the motivations of rural tourists in the United States. Therefore, this study aims to understand travel motivations and characteristics of tourists visiting a rural destination to provide a better understanding of rural tourism in the United States. Our study site, Potter County, Pennsylvania, represents a typical American rural area with a population of 18,080, which is currently struggling to pursue economic revitalization by attracting tourists. The findings of this study indicated that visitors to Potter County do not fit a homogenous rural tourist profile due to their broad travel preferences. The findings also suggest that tourism in rural areas may be a better descriptor than rural tourism in the context of Potter County, Pennsylvania

    Effect of Phase Clustering Bias on Phase-Amplitude Coupling for Emotional EEG

    Background: Emotions are thought to be related to distinct patterns of neural oscillations, but the interactions among multi-frequency neural oscillations during different emotional states lack full exploration. Phase-amplitude coupling is a promising tool for understanding the complexity of the neurophysiological system, thereby playing a crucial role in revealing the physiological mechanisms underlying emotional electroencephalogram (EEG). However, the non-sinusoidal characteristics of EEG lead to the non-uniform distribution of phase angles, which could potentially affect the analysis of phase-amplitude coupling. Removing phase clustering bias (PCB) can uniform the distribution of phase angles, but the effect of this approach is unknown on emotional EEG phase-amplitude coupling. This study aims to explore the effect of PCB on cross-frequency phase-amplitude coupling for emotional EEG. Methods: The technique of removing PCB was implemented on a publicly accessible emotional EEG dataset to calculate debiased phase-amplitude coupling. Statistical analysis and classification were conducted to compare the difference in emotional EEG phase-amplitude coupling prior to and post the removal of PCB. Results: Emotional EEG phase-amplitude coupling values are overestimated due to PCB. Removing PCB enhances the difference in coupling strength between fear and happy emotions in the frontal lobe. Comparable emotion recognition performance was achieved with fewer features after removing PCB. Conclusions: These findings suggest that removing PCB enhances the difference in emotional EEG phase-amplitude coupling patterns and generates features that contain more emotional information. Removing PCB may be advantageous for analyzing emotional EEG phase-amplitude coupling and recognizing human emotions

    Sustained Small Interfering RNA-Mediated Human Immunodeficiency Virus Type 1 Inhibition in Primary Macrophages

    Small interfering RNAs (siRNAs) can induce potent gene silencing by degradation of cognate mRNA. However, in dividing cells, the silencing lasts only 3 to 7 days, presumably because of siRNA dilution with cell division. Here, we investigated if sustained siRNA-mediated silencing of human immunodeficiency virus type 1 (HIV-1) is possible in terminally differentiated macrophages, which constitute an important reservoir of HIV in vivo. CCR5, the major HIV-1 coreceptor in macrophages, and the viral structural gene for p24 were targeted either singly or in combination. When transfected 2 days prior to infection, both CCR5 and p24 siRNAs effectively reduced HIV-1 infection for the entire 15-day period of observation, and combined targeting of both genes abolished infection. To investigate whether exogenously introduced siRNA is maintained stably in macrophages, we tested the kinetics of siRNA-mediated viral inhibition by initiating infections at various times (2 to 15 days) after transfection with CCR5 and p24 siRNAs. HIV suppression mediated by viral p24 siRNA progressively decreased and was lost by day 7 posttransfection. In contrast, viral inhibition by cellular CCR5 knockdown was sustained even when transfection preceded infection by 15 days, suggesting that the continued presence of target RNA may be needed for persistence of siRNA. The longer sustenance of CCR5 relative to p24 siRNA in uninfected macrophages was also confirmed by detection of internalized siRNA by modified Northern blot analysis. We also tested the potential of p24 siRNA to stably silence HIV in the setting of an established infection where the viral target gene is actively transcribed. Under these circumstances, long-term suppression of HIV replication could be achieved with p24 siRNA. Thus, siRNAs can induce potent and long-lasting HIV inhibition in nondividing cells such as macrophages
