    Energetics of Protein-DNA Interactions

    Protein-DNA interactions are vital for many processes in living cells, especially transcriptional regulation and DNA modification. To further our understanding of these important processes on the microscopic level, it is necessary that theoretical models describe the macromolecular interaction energetics accurately. While several methods have been proposed, there has not been a careful comparison of how well the different methods are able to predict biologically important quantities such as the correct DNA binding sequence, total binding free energy, and free energy changes caused by DNA mutation. In addition to carrying out the comparison, we present two important theoretical models developed initially in protein folding that have not yet been tried on protein-DNA interactions. In the process, we find that the results of these knowledge-based potentials show a strong dependence on the interaction distance and the derivation method. Finally, we present a knowledge-based potential that gives comparable or superior results to the best of the other methods, including the molecular mechanics force field AMBER99

    Subjective wellbeing among university students and recent graduates: Evidence from the United Kingdom

    This study compares studentsā€™ and recent graduatesā€™ perceptions of their subjective wellbeing and offers support mechanisms and resources to enhance wellbeing in higher education. Survey data were collected in September 2021 from 414 UK-based higher education students and recent graduates on their self-perceived subjective wellbeing in March 2020 (before COVID-19 regulations restrictions) and September 2021 (18 months later). Findings showed that subjective wellbeing scores fell for almost three-quarters of university students and recent graduates between March 2020 and September 2021. Interestingly, around one-fifth of participants reported increased subjective wellbeing scores whilst the remaining participants reported no impact. Positive impacts of the pandemic included opportunities for self-improvement with more free time for focusing on health and relationships. Adverse outcomes included feelings of isolation, reduced mental and physical health, difficulties undertaking degree studies and work, travel restrictions, and concerns for labour market competitiveness. The study advances the application of the conservation of resources theory and identifies strategies for higher education institutions to better support and improve their studentsā€™ and future graduatesā€™ subjective wellbeing. Strategies include access to counselling, mindfulness, opportunities for participation in hobbies, interaction with peers, flexible work and study options, and guidance on career and finances

    Conceptualisation and Operationalisation of the Personal Brand V.A.L.U.E. Career Development Tool

    The purpose of our manuscript is to conceptualise and operationalise the Personal Brand V.A.L.U.E. Career Development Tool. The V.A.L.U.E. acronym stands for Vision, Ability, Love, Understanding, and Ecosystem. The paper adopts a case study design based on qualitative insights via an in-depth and semi-structured interview with an individual who has used the Personal Brand V.A.L.U.E. Career Development Tool over five years, initially in their final year of university (2018-2019) and subsequently as a graduate in the labour market (2019-2023). The lived experience provided through the case study provides insights into the operationalisation of the Personal Brand V.A.L.U.E. Career Development Tool for (i) preparing and navigating the university-to-work transition, (ii) navigating an unplanned career transition, and (iii) navigating a planned career transition. The theoretical contribution comes from conceptualising the Personal Brand V.A.L.U.E. Career Development Tool. The practical contribution comes from offering implications for (a) students and universities and (b) workers and organisations to operationalise the tool. A future research agenda is also presented

    Radiological assessment for Space Station Freedom

    Circumstances have made it necessary to reassess the risks to Space Station Freedom crewmembers that arise from exposure to the space radiation environment. An option is being considered to place it in an orbit similar to that of the Russian Mir space station. This means it would be in a 51.6 deg inclination orbit instead of the previously planned 28.5 deg inclination orbit. A broad range of altitudes is still being considered, although the baseline is a 407 km orbit. In addition, recent data from the Japanese A-bomb survivors has made it necessary for NASA to have the exposure limits reviewed. Preliminary findings of the National Council on Radiation Protection and Measurements indicate that the limits must be significantly reduced. Finally, the Space Station will be a laboratory where effects of long-term zero gravity on human physiology will be studied in detail. It is possible that a few crewmembers will be assigned to as many as three 1-year missions. Thus, their accumulated exposure will exceed 1,000 days. Results of this radiation risk assessment for Space Station Freedom crewmembers finds that females less than 35 years old will be confined to mission assignments where the altitude is less than about 400 km. Slight restrictions may also need to be made for male crewmembers less than 35 years old

    [Excerpt] Harsh skepticism pervades current public debate about who deserves public support and on what basis, particularly regarding the claims of individuals with disabling illness and injury. Heretofore, these claims were accepted, even reservedly, and the needs of such individuals were considered to be legitimate even when they were monitored closely. The Supplemental Security Income (SSI) and Social Security Disability Insurance (SSDI) programs and their recipients have been among the most visible and vulnerable targets of increased scrutiny and shrinking public beneficence. In 1997, congressional legislation redefined SSI eligibility for children, sparked largely by concerns that children have been deployed to engage in a type of public begging by acting crazy in order to secure benefits for their families. Maladaptive behaviors was removed from the mental disorder listings, and the Social Security Administration (SSA) estimates that 135,000 children will lose their benefits after review. In March 1996, Congress eliminated SSI, SSDI, Medicare, and Medicaid benefits for persons whose drug addiction or alcoholism is a prominent cause of disability, and as a result 141,000 recipients have been terminated. The SSA also was ordered to begin another sweeping review of all recipients of disability income. SSA officials reportedly expect this process to produce a termination rate of 14 percent, resulting in an estimated 196,000 additional individuals who would cease to receive SSI and SSDI
