    Michelle Go was pushed in front of a subway car by a man suffering from schizophrenia that had fallen through the cracks of New York’s mental health care system. Michelle’s death was imminent because the severely ill man had every right to be on the streets under present law. This note will discuss the problems with New York’s mental hygiene laws that prevent courts from mandating treatment even when treatment is in the state’s best interest. Michelle’s death is not unique. Historically, New York has struggled to enact effective legislation governing the treatment of mentally ill individuals. As a result, hundreds of thousands of mentally ill New Yorkers evade treatment each year. While courts mandate treatment upon individuals adjudicated to be a threat to themselves or others, this standard creates a burden that has proven too difficult to establish in a court of law. The state’s mental hygiene statutes do not seek to prevent the deterioration of a person suffering from a severe mental illness. Therefore, courts lack the authority to mandate treatment even when treatment is in the patient’s best interest. Amending New York’s assisted outpatient statute to include a preventative standard for schizophrenia, manic depression, and bipolar disorder would allow courts to equitably protect the mentally ill population and the community at large

    Employee performance evaluation activities within a company or organization is an important activity that must be done in order to maintain the quality of human resources in the company or organization. Faculty and staff appraisal system that is currently performed by STT Musi still using the manual method, which uses a questionnaires that divided each semester and still using standard formulas for calculations. This method still holds many shortcomings and assessments made are subjective. One way that can be applied in providing solutions to today's problems is to apply a profile matching method for a more objective assessment. In making this DSS, using VB.Net programming language and SQLServer database programming language. The results of calculation of employee performance evaluation period from July to December 2013 by using a manual calculation is ST04 as the best employee with value 6,97 and manual calculations with the system results ST04 with value 6,9675 as the best employee

    Determining the Role of TLR Agonists in T Cell-Based Cancer Therapy

    One of the main mechanisms by which total body irradiation (TBI) enhances adoptive cell transfer (ACT) immunotherapy is by inducing systemic innate immune activation, triggered by microbial LPS released from the radiation-injured gut. Although microbial LPS is a critical mediator of TBI enhancement, it is unknown whether exogenous administration of LPS can augment therapeutic outcome. We report here that administration of LPS to non-irradiated animals did not replicate the ACT effectiveness seen in irradiated animals. In contrast, however, we found that LPS dramatically augmented ACT treatment in irradiated mice. Moreover, bacterial derived TLR agonists other than LPS (such as CpG ODN and Monophospholipid A) but not fungi- or virus-derived signals (such as PolyI:C, Zymosan, Loxoribine, and Imiquimod) improved antitumor immune responses in irradiated animals. Of clinical importance, we found that in vitro priming with LPS, CpG or MPL enhanced the anti-tumor activity of transferred CD8+ T cells. While it would be ideal to simply add TLR agonists to APCs to enhance their immunogenicity and in turn increase CD8+ T cell function, we must consider the potential negative feedback immune-regulatory mechanisms that result from TLR agonist priming, such as elevated IL-10 production. We found that transiently neutralizing IL-10 enhanced anti-tumor activity of CD8+ T-cells. However, priming CD8+ T cells with CpG ODN while neutralizing IL-10 did not augment the anti-tumor response in mice. Furthermore, we found that IL-10 was necessary to maintain IL-17 production and illicit a potent anti-tumor response in Th17 cells primed with CpG ODN

    The factors affecting CyA dosing and kinetics in LT patients are complex, and have been thoroughly investigated and reviewed. Plasma or WB CyA concentration monitoring remains the best method presently available for adjusting CyA dosage in LT patients in a timely manner. The availability of an FPIA assay for CyA has produced rapid drug analysis for transplant patient monitoring, but adds additional factors that must be considered in interpreting CyA concentrations. Liver dysfunction may disproportionately elevate CyA plasma or blood levels when analyzed by FPIA in relation to PC-RIA or HPLC, and adjustment of the therapeutic range or analysis by a more specific assay method may be necessary for dosage adjustment in these patients. The availability of a more specific antibody in an FPIA assay may avert these problems, as would the development of immunologic monitoring techniques that provide a global assessment of immune suppression produced by increasingly complex immunosuppressive regimens in LT patients

We argue that confidence in the European Union depends on an underlying trust in social and political institutions, and that confidence in the EU does not differ substantially from confidence in other international organizations.
