    Retinal nerve fiber layer thickness measured by optical coherence tomography in Chinese teenagers aged from 13 years old to 18 years old

    AIM: To establish a reference range of retinal nerve fiber layer(RNFL)thickness by optical coherence tomography(OCT)in Chinese teenagers aged from 13 to 18 years old, and investigate its relationship with age, eye side, gender, and ethnic group.<p>METHODS: A total of 402 eyes from 201 normal Chinese aged from 13 to 18 years old were recruited for this study. Optic disk with 3.4mm diameter circle in different global average, quadrant and part-time bit retinal nerve fiber layer thickness(RNFLT)was measured by RNFL thickness average analysis program. Their RNFLT at different part-time bit, quadrant and global average RNFLT around the disc were measured by OCT with 3.4mm diameter circle, using the RNFL thickness average analysis program. The data was analyzed with SPSS statistical 19.0. The influences of several factors(such as age, eye side, gender, and ethnic group)on RNFLT were also analyzed.<p>RESULTS: The global average RNFLT at 13, 14, 15, 16, 17, 18 years old was 108.32±9.42μm, 109.23±9.67μm, 110.36±11.14μm, 111.27±10.21μm, 109.23±9.67μm, 112.11±8.83μm respectively. RNFLT of right eyes was 109.82±8.93μm and of left eyes was 110.33±9.89μm. All of the male's average RNFLT was 110.14±10.02μm, and all of the female's average RNFLT was 109.96±11.22μm. The average RNFLT of Han nationality was 110.22±9.31μm and of non-Han nationality was 109.87±8.65μm. The average RNFLT of all was 110.02±9.87μm, the RNFLT at the superior, nasal, inferior and temporal quadrant was 146.56 ±18.88μm, 76.49±13.28μm, 136.64±16.29μm, 82.01±12.55μm respectively. There was no significant difference in gender, eye side, and ethnic group(all <i>P</i><0.05), but the difference was significant compared with adult's data of the database. <p>CONCLUSION: This study has established a normal standard reference of RNFLT and its related indexes by OCT in Chinese teenagers aged 13-18 years old. Gender, age, eye side, and ethnic group have no effect on their RNFLT, which has significant difference with adult's data. And for the diagnosis and follow-up of diseases involving in RNFL changes, the adult's data can not be used

    Therapeutic effects of Saikosapoin D on bleomycininduced pulmonary fibrosis in mice via regulation of IL- 33/ST2 pathway

    Purpose: To investigate the therapeutic effects of saikosapoin D (SSD) on bleomycin (BLM)-induced pulmonary fibrosis (PF) in mice and its probable mechanisms.Methods: PF mice were prepared by intraperitoneal (i.p.) injection of BLM (5 mg/kg). Twenty-four hours later, 72 mice in SSD group were administered SSD (1.8 mg/kg, ip). After 3, 7, 14 and 28 days of injection, the mice were sacrificed. Blood samples and lung tissues were collected from 6 mice in each group. The lung tissues were subjected to histological examination. In addition, expressions of MyD88, TRAF6, IL-33 and ST2 in lung tissue were determined by western blotting assay. Serum levels of hydroxyproline (HYP), interleukin (IL)-4, IL-13 and interferon (IFN)-γ were measured by enzyme-linked immunosorbent assay (ELISA).Results: Pathological results showed that SSD treatment alleviated alveolitis and lung fibrosis (p &lt; 0.05) in lung tissues of PF mice at 14 and 28 days post-BLM injection. HYP and IL-13 levels of mice in SSD group were significantly lower than that in BLM group at days 14 and 28 post-BLM injection (p &lt; 0.05). Levels of IL-4 and IFN-γ were significantly lower when compared with values in BLM group on day 28 (p &lt; 0.05). Western blotting results revealed that expressions of MyD88, TRAF6, IL-33 and ST2 proteins were significantly decreased by SSD treatment (p &lt; 0.05).Conclusion: SSD exerts therapeutic effects on BLM-induced experimental PF in mice via regulation ofIL-33/ST2 pathway.Keywords: Saikosapoin D, Idiopathic pulmonary fibrosis, Myeloid differentiation factor, Hydroxyproline, Interleukin, Interferon, IL-33/ST2 pathwa

    Enhanced antitumor efficacy and reduced systemic toxicity of sulfatide containing nanoliposomal doxorubicin in a xenograft model of colorectal cancer

    Sulfatide is a glycosphingolipid known to interact with several extracellular matrix proteins, such as tenascin-C which is overexpressed in many types of cancer including that of the colon. In view of the limited success of chemotherapy in colorectal cancer and high toxicity of doxorubicin (DOX), a sulfatide-containing liposome (SCL) encapsulation approach was taken to overcome these barriers. This study assessed the in vitro cytotoxicity, biodistribution, therapeutic efficacy and systemic toxicity in vivo of sulfatide-containing liposomal doxorubicin (SCL-DOX) using human colonic adenocarcinoma HT-29 xenograft as the experimental model. In vitro, SCL-DOX was shown to be delivered into the nuclei and displayed prolonged retention compared with the free DOX. The use of this nanodrug delivery system to deliver DOX for treatment of tumor-bearing mice produced a much improved therapeutic efficacy in terms of tumor growth suppression and extended survival in contrast to the free drug. Furthermore, treatment of tumor-bearing mice with SCL-DOX resulted in a lower DOX uptake in the principal sites of toxicity of the free drug, namely the heart and skin, as well as reduced myelosuppression and diminished cardiotoxicity. Such natural lipid-guided nanodrug delivery systems may represent a new strategy for the development of effective anticancer chemotherapeutics targeting the tumor microenvironment for both primary tumor and micrometastases

    A Study of Ground Deformation in the Guangzhou Urban Area with Persistent Scatterer Interferometry

    The Interferometric Point Target Analysis (IPTA) technique and Advanced Synthetic Aperture Radar (ASAR) images acquired over Hong Kong from 2007–2008 were used to detect ground deformation in the urban area of Guangzhou city in South China. A ground deformation rate map with scattered distribution of point targets shows the maximum subsidence (rise) rate as high as -26 to -20 mma-1 (16–21 mma-1), implying that the study area is an active zone for ground deformation. Based on the point target map, a contour ground deformation rate map is generated. The map shows three major subsidence zones located in the middle-west, the east, and the southwest of the study area, respectively. All the six ground collapse accidents that occurred in 2007–2008 fall within the subsidence zones, qualitatively validating the IPTA results. Ground subsidence and geological conditions on Datansha Island are examined. The results indicate that the local geological conditions, such as limestone Karst geomorphology as well as silt layers characterized by high water content, high void ratio, high compressibility, low bearing capacity and low shear strength, and underground engineering projects are responsible for ground subsidence and ground collapse accidents occurred there

    Improved efficacy and reduced toxicity of doxorubicin encapsulated in sulfatide-containing nanoliposome in a glioma model

    As a glycosphingolipid that can bind to several extracellular matrix proteins, sulfatide has the potential to become an&nbsp;effective targeting agent for tumors overexpressing tenasin-C in their microenvironment. To overcome the dose-limiting&nbsp;toxicity of doxorubicin (DOX), a sulfatide-containing nanoliposome (SCN) encapsulation approach was employed to&nbsp;improve treatment efficacy and reduce side effects of free DOX. This study analysed in vitro characteristics of sulfatidecontaining&nbsp;nanoliposomal DOX (SCN-DOX) and assessed its cytotoxicity in vitro, as well as biodistribution, therapeutic&nbsp;efficacy, and systemic toxicity in a human glioblastoma U-118MG xenograft model. SCN-DOX was shown to achieve highest&nbsp;drug to lipid ratio (0.5:1) and a remarkable in vitro stability. Moreover, DOX encapsulated in SCN was shown to be delivered&nbsp;into the nuclei and displayed prolonged retention over free DOX in U-118MG cells. This simple two-lipid SCN- DOX nanodrug has favourable pharmacokinetic attributes in terms of prolonged circulation time, reduced volume of distribution and&nbsp;enhanced bioavailability in healthy rats. As a result of the improved biodistribution, an enhanced treatment efficacy of SCNDOX&nbsp;was found in glioma-bearing mice compared to the free drug. Finally, a reduction in the accumulation of DOX in the&nbsp;drug&rsquo;s principal toxicity organs achieved by SCN-DOX led to the diminished systemic toxicity as evident from the plasma&nbsp;biochemical analyses. Thus, SCN has the potential to be an effective and safer nano-carrier for targeted delivery of&nbsp;therapeutic agents to tumors with elevated expression of tenascin-C in their microenvironment

    Dorsalis Pedis Artery Blood Pressure is a Good indicator for Blood Pressure Monitoring during Shock and Recovery

    Abstract To compare Supine ankle dorsalis pedis artery and brachial artery blood pressure in patients during shock resuscitation and of different ages was compared, in order to evaluate the clinical value of ankle dorsalis pedis artery blood press monitoring. 80 hypovolemic shock (trauma, hemorrhagic shock) patients and 150 selected patients, including 50 children, 50 young and 50 old patients, were enrolled in this study. The left dorsalis pedis artery and brachial blood pressure were monitored by multi-function monitor in all patients. We observed a correlation between dorsalis pedis artery blood pressure and brachial blood pressure during the period of shock and the recovery with the use of vasoactive drugs. In addition, the values of dorsalis pedis artery blood pressure and brachial blood pressure were significantly correlated in three groups with different ages. There is a positive correlation between dorsalis pedis artery blood pressure and brachial blood pressure in the state of shock and recovery as well as in populations of different ages. Monitoring dorsalis pedis artery blood pressure can reflect the hemodynamic changes timely and accurately, and has important clinical value
