    New protocol for rapid cassava multiplication in field conditions: a perspective on speed breeding

    Despite the economic and social importance, high-yielding cassava cultivars are only released after extensive research, mainly due to the low multiplication rate. This study aimed to assess the impact of using smaller-sized seed cuttings treated with agrochemicals (8MP) compared to the conventional planting size (16 cm) on genetic parameters, agronomic performance, and the ranking of cassava clones based on yield and growth attributes. The evaluation was carried out in clonal evaluation trial (CET), preliminary yield trial (PYT), and uniform yield trials (UYT). Additionally, a new selection scheme for cassava breeding programs was proposed. A total of 169 clones were evaluated, including 154 improved clones at different stages of selection and 15 local varieties used as checks. Field trials were conducted using both sizes of propagative material (8MP and 16 cm) in each phase of the breeding program. The data were analyzed using mixed models, considering the random effects of genotype and genotype-environment interaction (G×E) to determine variances and heritabilities. Bland-Altman concordance and correlation analysis of selection indices were employed to examine the consistency in the ranking of cassava clones using different seed cutting sizes. The distribution of variance components, heritabilities, means, and range of the 8MP and 16 cm trials in different phases of the cassava breeding program exhibited remarkable similarity, thereby enabling a comparative assessment of similar genetic effects. With a selection intensity of 30%, the concordance in clone ranking was 0.41, 0.57, and 0.85 in CET, PYT, and UYT trials, respectively, when comparing the selection based on 8MP and 16 cm trials. It is worth noting that the ranking of the top 15% remained largely unchanged. Based on the findings, proposed changes in the cassava selection scheme involve increasing the number of trials starting from the CET phase, early incorporation of G×E interaction, elimination of the PYT trial, reduction of the breeding cycle from 5 to 3 years, and a decrease in the time required for variety development from 11 to 9 years. These modifications are expected to lead to cost reduction and enhance the effectiveness of cassava breeding programs

    Model-assisted phenotyping by digital images in papaya breeding program

    Manual phenotyping for papaya Carica papaya (L) breeding purposes limits the evaluation of a great number of plants and hampers selection of superior genotypes. This study aimed to validate two methodologies for the phenotyping of morpho-agronomic plant traits using image analysis and fruit traits through image processing. In plants of the ‘THB’ variety and ‘UENF/Caliman-01’ hybrid two images (A and B) were analyzed to estimate commercial and irregularly shaped fruits. Image A was also used in the estimation of plant height, stem diameter and the first fruit insertion height. In ‘THB’ fruits, largest and smallest diameters, length, and volume were estimated by using a caliper and image processing (IP). Volume was obtained by water column displacement (WCD) and by the expression of ellipsoid approximation (EA). Correlations above 0.85 between manual and image measurements were obtained for all traits. The averages of the morpho-agronomic traits, estimated by using images, were similar when compared to the averages measured manually. In addition, the errors of the proposed methodologies were low compared to manual phenotyping. Bland-Altman's approach indicated agreement between the volume estimated by WCD and EA using caliper and IP. The strong association obtained between volume and fruit weight suggests the use of regression to estimate this trait. Thus, the expectation is that image-based phenotyping can be used to expand the experiments, thereby maintaining accuracy and providing greater genetic gains in the selection of superior genotypes

    Combining ability of recombined F4 papaya lines: a strategy to select hybrid combination

    The growing demand for quality of papaya by the domestic and international markets requires the development of genotypes that are capable of incorporating traits, such as high yield and fruit quality. This study estimated the genetic parameters and the specific combining ability of F4 papaya lines crossed to the SS-72/12 tester to identify genotypes with higher genetic value for hybrid production. We evaluated 169 treatments consisting of 62 topcross hybrid combinations, 97 F5 lines, and 10 controls in a 13 × 13 lattice design with five replicates and two plants per plot. The following traits were evaluated: plant height (PH), stem diameter (SD), first-fruit insertion height (FFIH), number of marketable fruits (NMF), number of deformed fruits (NDF), number of fruitless nodes (NFN), average fruit weight (FW), and yield (YLD). Differences p < 0.05 were found in most traits evaluated. The specific combining ability estimates indicate that hybrids formed by the cross between SS-72/12 tester and the following lines are promising, considering the multiple traits: UCLA08-088, UCLA08-101, UCLA08-071, UCLA08-014, UCLA08-025, UCLA08-028, UCLA08-122, UCLA08-055, UCLA08-026, and UCLA08-092. These hybrids meet the demands for domestic and international markets and may be available to producers as new papaya cultivars

    A novel seed treatment-based multiplication approach for cassava planting material.

    Cassava (Manihot esculenta Crantz) is an important food security crop in many parts of the developing world. The crop?s high yield potential and multitude of uses?both for nutrition and processing?render cassava a promising driver for the development of rural value chains. It is traditionally propagated from stem cuttings of up to 30 cm in length, giving a multiplication rate as low as 1:10. Propagating cassava traditionally is very inefficient, which leads to challenges in the production and distribution of quality planting material and improved cultivars, greatly limiting the impact of investments in crop breeding. The work described in the present study aimed to develop a seed treatment approach to facilitate the use of shorter seed pieces, increasing the multiplication rate of cassava and thus making the crop?s seed systems more efficient. After several tests, formulation was identified, consisting of thiamethoxam 21 g ha-1, mefenoxam 1.0 g ha-1, fludioxonil 1.3 g ha-1, thiabendazole 7.5 g ha-1 and Latex 2% as a binder. Plant growing from seed pieces treated with this formulation displayed increased crop establishment and early crop vigor, leading to an improved productivity throughout a full growing cycle. This allowed to reduce the cassava seed piece size to 8 cm with no negative effects on germination and crop establishment, leading to yields comparable to those from untreated 16 cm pieces. This, in turn, will allow to increase the multiplication ratio of cassava by a factor of up to 3. Notably, this was possible under regular field conditions and independently of any specialised treatment facilities. Compared with existing seed production protocols, the increased multiplication rates allowed for efficiency gains of between 1 to 1.9 years compared to conventional five-year cycles. We believe that the technology described here holds considerable promise for developing more reliable and remunerative delivery channels for quality cassava planting material and improved genetics

    Papaya recombinant inbred lines selection by image-based phenotyping

    The selection of superior Carica papaya (L) genotypes depends on the availability of genetic variability and on the favorable and simultaneous response of the genotypes to those traits of most interest. However, manual phenotyping (MP) demands intensive labor, is time-consuming and expensive. The aim of the current study is to access the efficiency of image-based phenotyping (IBP) in estimating genetic parameters and in selecting F4 recombinant inbred lines. The genetic parameters and values were estimated in accordance with the REML/BLUB procedure and combined selection using the selection index based on standardized genetic values. The majority of traits accessed through IBP showed experimental coefficients of variation similar to those found through MP. Both methodologies showed genetic parameters of similar magnitude, indicating expressive genetic variability between lines in the traits accessed in this study. The same superior lines were indicated in both methodologies and expressive genetic gains obtained through the lines were selected for all traits. IBP performance was similar to that of MP with respect to the estimates of breeding-relevant traits such as commercial fruits and yield. Thus, IBP showed efficient phenotypic assessment, as well as selective accuracy in accessing genetic variability and genetic gains, when it was compared to MP. Since IBP is far less dependent on labor, it is expected to be incorporated into the routine of papaya breeding programs as a way of increasing the number of accessed lines and, consequently, increasing genetic gains

    Quantification of f loral abnormalities in a population generated from sexual polymorphism aiming at recurrent selection in papaya

    ABSTRACT The aim of this study was to estimate the percentage of deformed fruits (carpelloid, pentandric, and bananoid), summer sterility, repeatability coefficients, and correlations among the variables for floral abnormalities; and select genotypes withthe lowest incidence of deformed fruits and summer sterility. In theexperiment, which involved 254 plants, traits were evaluated using the digital-phenotypic method and the obtained images were processed and analysed using ImageJ v1.48 software with the Cell Counter plugin. Summer sterility had the highest percentage of occurrence in the evaluated periods. Based on the quantificationof total abnormalities, the 30 genotypes with the lowest incidence offloral abnormalities were selected. Summer sterility is the main problem of the papaya crop in terms of floral abnormalities. The non-significant correlations between the traits: NDF/TNF+NFN (number of deformed fruits/number of fruits + number of fruitless nodes) and NFS/TNF+NFN (number of fruitless nodes/number of fruits + number of fruitless nodes); NDF/TNF+NFN and NFN/SUM(number of fruitless nodes/sum of abnormalities); andNFS/TNF+NFN and NDF/SUM (number of deformed fruits/sum of abnormalities) indicate that the variables are independent of each other. Of the 254 evaluated progenies, at least 30 were identified as having potential for selection, with lower occurrence of total floral abnormalities
