Authoritarian Recall: Mexico's Drug War and Subnational Patterns of Opposition to Democracy
In times of crisis, citizens' support for democracy can depend on how well they think their democracy can address that crisis compared to authoritarian alternatives. Mexico is in the midst of just such a crisis, as its war on drug trafficking organisations has brought an unprecedented rise in violence and, in some areas, posed a direct challenge to the state's capacity to govern. At the same time, its subnational political landscape ranges from vibrant, multi-party states to those with continued connections to a dominant one-party past. We leverage these variations in subnational political context and levels of drug-related violence to examine how the subnational political context mediates the relationship between a crisis and support for non-democratic alternatives. When faced with a violent shock to the system, public attitudes towards democracy depend in part on one's experiences with non-democratic alternatives and whether these authoritarian options appear to solve the crisis at hand more effectively
Targeted Insertion of the mPing Transposable Element
Class II DNA Transposable Elements (TEs) are moved from one location to another in the genome by the action of transposase proteins that bind to repeat sequences at the ends of the elements. Although the location TE insertion is mostly random, the addition of DNA binding domains to the transposase proteins has allowed for targeted insertion of some elements. In this study, the Gal4 binding domain was added to the transposase proteins, ORF1 and TPase, which mobilize the mPing element from rice. The Gal4:TPase construct was capable of increasing the number of mPing insertions into the Gal2 and Gal4 promoter sequences in yeast. While this confirms that mPing insertion preference can be manipulated, the target specificity is relatively low. Thus, the CRISPR/Cas9 system was tested for its ability to generate targeted insertion of mPing. A dCas9:TPase fusion protein had a low transposition rate suggesting that the addition of this large protein disrupts TPase function. Unfortunately, the use of a MS2 binding domain to localize the TPase to the MS2 hairpin containing gRNA failed to produce targeted insertion. Thus, our results suggest that the addition of small DNA binding domain to the N-terminal of TPase is the best strategy for targeted insertion of mPing
The Effects of Surface Texture on a Variety of Balance Assessments
Purpose: The purpose of this study was to observe whether there was a difference in balance scores between the utilization of different floor surfaces. The surfaces used to assess the balance scores within the individual were hardwood, traditional yoga mat, and NABOSO mat. The NABOSO technology was designed to increase mechanical proprioception in hands and feet.
Methods: The investigators recruited 18 participants between the ages of 19-25, all of them met eligibility criteria to partake in the study. They all completed a Physical Activity Readiness Questionnaire and informed consent document prior to the tests to ensure that there were no factors that would exclude them from the study. The subjects were randomly assigned with regards to how they would progress through the three floor surface conditions. The investigators then led the subjects in a brief warm-up, took their height, and leg height. All subjects started out on the Neurocom Balance Master performing a Unilateral Eyes Open & Eyes Closed test followed by the Y-Balance test.
Results: A repeated measures ANOVA revealed that there were no significant differences, p\u3e0.05, between the static and dynamic balance tests (Unilateral Stance and Y-Balance Test) among any of the three floor surface conditions (Control- hardwood, yoga mat, NABOSO mat).
Conclusion: The study conducted revealed no significant difference within the test scores of the participant among the three conditions. To provide further support or argument of the results collected in this study, future considerations should be taken into account to conduct the study on special populations such as geriatric patients or children/adolescents. Future studies should also consider prolonging the time of the study and increasing the sample size that might eliminate factors that will affect the accuracy of the results and observe whether the NABOSO mat has an increasing effect on balance and proprioception or experiences similar results to this study
Determining the Fundamental Failure Modes in Ni-rich Lithium Ion Battery Cathodes
Challenges associated with in-service mechanical degradation of Li-ion
battery cathodes has prompted a transition from polycrystalline to single
crystal cathode materials. Whilst for single crystal materials,
dislocation-assisted crack formation is assumed to be the dominating failure
mechanism throughout battery life, there is little direct information about
their mechanical behaviour, and mechanistic understanding remains elusive.
Here, we demonstrated, using in situ micromechanical testing, direct
measurement of local mechanical properties within LiNi0.8Mn0.1Co0.1O2 single
crystalline domains. We elucidated the dislocation slip systems, their critical
stresses, and how slip facilitate cracking. We then compared single crystal and
polycrystal deformation behaviour. Our findings answer two fundamental
questions critical to understanding cathode degradation: What dislocation slip
systems operate in Ni-rich cathode materials? And how does slip cause fracture?
This knowledge unlocks our ability to develop tools for lifetime prediction and
failure risk assessment, as well as in designing novel cathode materials with
increased toughness in-service

Amphotericin forms an extramembranous and fungicidal sterol sponge.
For over 50 years, amphotericin has remained the powerful but highly toxic last line of defense in treating life-threatening fungal infections in humans with minimal development of microbial resistance. Understanding how this small molecule kills yeast is thus critical for guiding development of derivatives with an improved therapeutic index and other resistance-refractory antimicrobial agents. In the widely accepted ion channel model for its mechanism of cytocidal action, amphotericin forms aggregates inside lipid bilayers that permeabilize and kill cells. In contrast, we report that amphotericin exists primarily in the form of large, extramembranous aggregates that kill yeast by extracting ergosterol from lipid bilayers. These findings reveal that extraction of a polyfunctional lipid underlies the resistance-refractory antimicrobial action of amphotericin and suggests a roadmap for separating its cytocidal and membrane-permeabilizing activities. This new mechanistic understanding is also guiding development of what are to our knowledge the first derivatives of amphotericin that kill yeast but not human cells
