Classification and structure-based inference of transcriptional regulatory proteins
Dissertação de mestrado em BioinformáticaTranscription factors (TFs) are proteins that mediate the cellular response to the changes of the
surrounding environment. Studying their functional domains and protein structure is fundamental
in order to gain insight of the way they are triggered and how they shape genetic transcription.
The current work aimed for classifying both TFs and functional domains, understanding which
features can be related to the different functions of the TFs.
By using UniProtJAPI, a JAVA library that allows remote access to UniProt, the information
of 200 Escherichia coli’s (E. coli) TFs has been retrieved. This data was manually curated, in
order to remove domain duplicates and other excess information, and to add missing domains.
The obtained functional domains were classified according to their molecular function, while
the TFs were classified according to their regulatory function. TFs that exclusively induce gene
expression were classified as activators, while TFs that only perform gene repression were classified
as repressors. On the other hand, TFs that perform both the activation and repression of
transcription were classified as duals. The information was then analysed altogether in order
to understand what relationships between the TFs’ function and functional domains could exist.
Several analysis were performed, which include statistical tests and clustering methods. Along
with the analysis of the full list of TFs, TFs that are part of two-component signal transduction
systems and global TFs were given special focus, due to their important role in cellular function.
The results showed that there is a relationship between the functional domains and the regulatory
function of the different TFs. This may be related to the evolutionary relationships between
repressors and activators. It is also understandable that dual regulators are closely related to activators
and repressors than what activators and repressors are to each other. Moreover, TFs of
two-component signal transduction systems are similar to each other, given that they perform
similar functions. Their domain architectures are also predictable and do not vary from what
was expected of these TFs. However, in global TFs the results are opposite of the ones obtained
for two-component system TFs: their structures are very different from each other and each TF
is specific. The amount of different domains is high when comparing to the full sample of TFs,
since the number of domains exceeds the number of TFs. Domains of all classification types are
present in their structure and the domain architectures are varied, which reflects their different
activities within the cell.Os factores de transcrição (TFs) são proteínas que mediam resposta celular perante alterações do
meio em que se inserem. Estudar os seus domínios funcionais e estrutura proteica é fundamental
para compreender a forma como as suas funções são desencadeadas e como moldam a regulação
da transcrição. Este trabalho teve como objectivos a classificação dos TFs de acordo com a sua
função, assim como a classificação dos domínios funcionais.
Através do uso da UniProtJAPI, uma biblioteca de JAVA que permite o acesso remoto à
UniProt, foi recolhida informação de 200 TFs da Escherichia coli (E. coli). Estes dados foram
curados manualmente, com o objectivo de remover domínios duplicados e outra informação em
excesso, assim como de adicionar domínios em falta.
Os domínios funcionais obtidos foram classificados de acordo com a sua função molecular,
enquanto que os TFs foram classificados de acordo com a sua função regulatória. TFs que exclusivamente
induzem a expressão genética foram classificados como activadores, enquanto que
TFs que apenas reprimem a expressão genética foram classificados como repressores. Por usa
vez, TFs que tanto induzem como reprimem a expressão genética foram classificados como duais.
A informação dos domínios e dos TFs foi considerada como um todo de forma a compreender
quais as possíveis relações entre a função regulatória dos TFs e os domínios funcionais. Várias
análises foram efectuadas, das quais testes estatísticos e métodos de clustering. Para além da
análise de todos os TFs, foi também feita uma análise de TFs que fazem parte de two-component
transduction systems e TFs globais, devido à sua importância na actividade celular.
Os resultados demonstram que existe uma relação entre os domínios funcionais e a função
regulatória dos TFs. Esta pode ter a ver com as relações evolucionárias dos activadores e repressores.
É, também, perceptível que os reguladores duais relacionam-se com mais proximidade
dos activadores e dos repressores do que os activadores e os repressores se relacionam entre
si. Para além disso, TFs de two-component transduction systems têm estruturas semelhantes ,
uma vez que desempenham funções idênticas. As duas arquitecturas de domínios também são
previsíveis e não variam do que era esperado. Contudo, para os TFs globais, os resultados são
antagónicos: as suas estruturas são diferentes umas das outras e cada TF é específico. A quantidade
de domínios diferentes é elevada em comparação com a amostra completa de TFs, uma vez
que o número de domínios excede o número de TFs. Domínios de todas as classificações estão
presentes na estrutura dos TFs globais e as arquitecturas de domínios são variadas, o que reflecte
as suas actividades específicas na célula
Discriminação, preconceito e atitudes relativamente a lésbicas : estudo numa amostra de estudantes de enfermagem
Dissertação de mestrado integrado em Psicologia (área de especializacao em Psicologia da Saúde)As atitudes, perceções e crenças dos profissionais de saúde acerca da homossexualidade feminina têm
implicações práticas no acesso desta comunidade a cuidados de saúde. No entanto, a investigação
neste domínio ainda permanece limitada. Neste sentido, este estudo pretende compreender e analisar a
discriminação, o preconceito e as atitudes para com lésbicas de futuros enfermeiros, com base numa
amostra particular de 204 estudantes de enfermagem do sexo feminino, com idades compreendidas
entre os 18 e 31 anos (M=21). Na realização deste trabalho, foram aplicados os seguintes
instrumentos: a) um questionário com dados sociodemográficos; b) a Escala de Perceção de
Discriminação; c) uma versão reduzida do Questionário Multidimensional do Preconceito Polimorfo, e
d) a Escala de Discriminação na Saúde. Os resultados mostram que estes estudantes, em média,
consideram que a religião é a dimensão mais discriminatória, ao contrário da área da saúde, que
apontam como sendo aquela em que há menos discriminação. Adicionalmente, a maioria admite que a
homofobia varia com o contexto e localização geográfica e acredita que a orientação sexual pode ser
um fator de risco (79.2%), relativamente a episódios de mal-estar psicológico/psiquiátrico. A
heteronormatividade dos participantes destacou-se como um fator relacionado com o nível de
discriminação na área da saúde. As crenças e/ou atitudes mais negativas foram encontradas nos
estudantes mais novos, nos que habitam em meios rurais, nos que detêm orientação política
tendencialmente de direita, e nos que não conhecem nem têm amigas lésbicas. No entanto, estes dados
devem ser interpretados com precaução, tendo em consideração fatores externos que poderão
contribuir para as perceções dos participantes. Seria desejável que os estudantes tivessem acesso a
mais informação sobre a comunidade lésbica, nomeadamente, através da inclusão de material
apropriado nos programas curriculares, potenciando um maior conhecimento e atitudes mais positivas
para com as pessoas lésbicas. É necessária a continuidade desta linha de estudos, no sentido de dar voz
às necessidades das minorias sexuais e em particular às das lésbicas, invisíveis pela condição de
mulher e orientação sexual em simultâneo.The attitudes, perceptions and beliefs of healthcare professionals about feminin homosexuality draw
practical implications in the access of this community to healthcare services. Nevertheless, research in
this domain is still sparse. In this direction, this study proposes to comprehend and analyse the
discrimination, the prejudice and the attitudes toward lesbians of future nurses, based on a particular
sample of 204 female nursing students, with ages between 18 and 31 years old (M=21). In the
development of this work, the following instruments have been applied: a) a questionary containing
sociodemographic data; b) the Scale of Perception of Discrimination; c) a shortened version of the
Multidimensional Questionary of the Polymorphic Prejudice and d) the Scale of Discrimination in
Health. The results show that these students, in average, consider that religion is the most
discriminatory dimension. Additionally, the majority admits that the homophobia varies with the
context and geographical location and believes that the sexual orientation can be a risk fator (79.2%),
in what concerns episodes of psychological/psychiatric discomfort. The heteronormativity of the
participants has revealed to be as a fator associated with the level of discrimination in the healthcare
area. The more negative beliefs and/or attitudes were assumed by the younger students, by those that
live in the countryside, by those that possess a tendentially right-wing political ideology, and by those
that do not have lesbian friends. However, these statistics must be interpreted with the precaution,
having in consideration external factors that may possibly contribute to the participants’ perceptions. It
would be desirable for students to have access to more information about the lesbian community,
through the inclusion of particular material in their curriculum programmes, fostering a greater
knowledge and more positive attitudes toward lesbian people. It is essential to continue this line of
studies, to voice the needs of the sexual minorities, in particular the lesbians that are invisible due to

Nanopartículas lipídicas: aplicações cosméticas
Projeto de Pós-Graduação/Dissertação apresentado à Universidade Fernando Pessoa como parte dos requisitos para obtenção do grau de Mestre em Ciências FarmacêuticasTendo em conta a sua composição e as vantagens que apresentam, as nanopartículas lipídicas têm sido amplamente aplicadas em cosmetologia, existindo atualmente vários produtos comercializados. Deste modo, a análise das potencialidades destes sistemas para novas aplicações cosméticas é essencial.
Neste trabalho é efetuada uma revisão bibliográfica relativa aos diferentes produtos cosméticos contendo nanopartículas lipídicas, quer estes se encontrem em fase de estudo ou já disponíveis no mercado.
Na primeira parte do trabalho é feita uma breve introdução acerca da estrutura da pele, da definição de produtos cosméticos e das suas principais caraterísticas e aplicações. Na segunda parte são descritos os sistemas de nanopartículas lipídicas, tendo em conta as suas caraterísticas estruturais, diferentes aplicações em cosmetologia e são dados exemplos dos produtos cosméticos existentes no mercado. According to their composition and advantages, lipid nanoparticles have been widely applied in cosmetology, and there are several products that can be found on the market nowadays. Therefore, the analysis of the potential of these new systems for cosmetic applications is essential.
In this work is performed a literature review about the diverse cosmetic products containing lipid nanoparticles, whether they are in clinical trials or already in the market.
In the first part, a brief introduction regarding the structure of the skin, the definition of cosmetic products, their main characteristics and applications are presented. In the second part are described the lipid nanoparticles systems, considering their structural characteristics, different applications in cosmetology and are given some examples of the cosmetic products that are already on the market
English as a communication tool in a global world: the case of Wall Street English
A globalização é um processo que dura há seculos e tem modificado as dinâmicas do mundo em relação a, por exemplo, negócios, pessoas e culturas. O seu impacto tem tido consequências como a expansão internacional das empresas, os fluxos elevados de migração e o incremento da cooperação mundial. Estes exemplos promovem encontros interculturais, particularmente em empresas multinacionais, que agora procuram potenciais colaboradores para lá das fronteiras de países, o que pode criar dilemas, especialmente devido a barreiras linguísticas e culturais. Contudo, o uso da língua inglesa como uma ferramenta de comunicação intercultural tem sido uma solução amplamente usada, sendo, por muitos, considerada uma “língua global” (Crystal, D. 2003) – uma língua que tem um status não-oficial a nível internacional e que é favorável em contraste com outras línguas quando barreiras de comunicação precisam de uma solução.
A necessidade de aprender Inglês tem, também por isso, vindo a crescer, sobretudo através de escolas e cursos de formação.
Wall Street English, criada nos anos setenta do século XX, é uma das empresas que contribui para o crescimento do número de falantes do inglês, em Portugal e no mundo, daí ter sido escolhida como a empresa de acolhimento do meu estágio curricular.
Este relatório dará, por isso, enfoque às atividades por mim desenvolvidas durante os seis meses de estágio na Wall Street English, ao perfil da organização e à importância da língua inglesa na sociedade global.Globalization is a centuries-long process that has shaped the world’s dynamic forces regarding, as examples, business, people, and culture. Its impact has had consequences such as companies’ international expansion, high flows of migration, and a vivid inclination to worldwide cooperation. These three examples by themselves have promoted intercultural encounters, particularly in multinational companies, which now seek potential collaborators beyond countries’ frontiers, which can lead to dilemmas, especially due to linguistic and cultural barriers. Yet, the use of English as an intercultural communication tool is being fairly used solution, being, by many, considered a “global language” (Crystal, D. 2003) – a language that has an unofficial status at an international level and which is favored against others when communication barriers need a solution.
The need to learn English has, therefore, been growing, mostly through schools and training courses.
Wall Street English, created in the seventies, is one of the companies responsible for the growth of English speakers, not only in Portugal, but also in the world. Thus, it was the host company for my curriculum internship. This report will focus on the activities developed by me throughout the six-month internship at Wall Street English regarding the organization’s profile and the importance of the English language in the global society
The acute effect of maximal exercise on central and peripheral arterial stiffness indices and hemodynamics in children and adults

FEDER-001883)This study compared the effects of a bout of maximal running exercise on arterial stiffness in children and adults. Right carotid blood pressure and artery stiffness indices measured by pulse wave velocity (PWV), compliance and distensibility coefficients, stiffness index α and β (echo-tracking), contralateral carotid blood pressure, and upper and lower limb and central/aortic PWV (applanation tonometry) were taken at rest and 10 min after a bout of maximal treadmill running in 34 children (7.38 ± 0.38 years) and 45 young adults (25.22 ± 0.91 years) having similar aerobic potential. Two-by-two repeated measures analysis of variance and analysis of covariance were used to detect differences with exercise between groups. Carotid pulse pressure (PP; η(2) = 0.394) increased more in adults after exercise (p < 0.05). Compliance (η(2) = 0.385) decreased in particular in adults and in those with high changes in distending pressure, similarly to stiffness index α and β. Carotid PWV increased more in adults and was related to local changes in PP but not mean arterial pressure (MAP). Stiffness in the lower limbs decreased (η(2) = 0.115) but apparently only in those with small MAP changes (η(2) = 0.111). No significant exercise or group interaction effects were found when variables were adjusted to height. An acute bout of maximal exercise can alter arterial stiffness and hemodynamics in the carotid artery and within the active muscle beds. Arterial stiffness and hemodynamic response to metabolic demands during exercise in children simply reflect their smaller body size and may not indicate a particular physiological difference compared with adults.info:eu-repo/semantics/publishedVersio
USE OF ASSISTED PHOTOGRAMMETRY FOR INDOOR AND OUTDOOR NAVIGATION PURPOSES
Nowadays, devices and applications that require navigation solutions are continuously growing. For instance, consider the increasing demand of mapping information or the development of applications based on users’ location. In some case it could be sufficient an approximate solution (e.g. at room level), but in the large amount of cases a better solution is required.
The navigation problem has been solved from a long time using Global Navigation Satellite System (GNSS). However, it can be unless in obstructed areas, such as in urban areas or inside buildings. An interesting low cost solution is photogrammetry, assisted using additional information to scale the photogrammetric problem and recovering a solution also in critical situation for image-based methods (e.g. poor textured surfaces). In this paper, the use of assisted photogrammetry has been tested for both outdoor and indoor scenarios. Outdoor navigation problem has been faced developing a positioning system with Ground Control Points extracted from urban maps as constrain and tie points automatically extracted from the images acquired during the survey. The proposed approach has been tested under different scenarios, recovering the followed trajectory with an accuracy of 0.20 m.
For indoor navigation a solution has been thought to integrate the data delivered by Microsoft Kinect, by identifying interesting features on the RGB images and re-projecting them on the point clouds generated from the delivered depth maps. Then, these points have been used to estimate the rotation matrix between subsequent point clouds and, consequently, to recover the trajectory with few centimeters of error
Structuring wicked problems in transdisciplinary research using the Social-Ecological Systems framework: an application to the montado system, Alentejo, Portugal
The aspiration to establish an effective dialogue between science and society has inspired some ground breaking examples of transdisciplinarity (TD). The core idea of TD is that different academic disciplines work jointly with practitioners to solve common problems. The first step of TD implies a contextualization that requires holistic and systemic thinking. To achieve this contextualization, we applied the Social–Ecological Systems (SES) framework with the aim of developing TD to deal with the recorded decline in area and tree density of the montado land-use system located in the Alentejo region, Portugal. The study was based both on a literature review and on the analysis of qualitative and quantitative data collected in a number of research projects on the montado. The results show that the lack of consensus regarding the system boundaries, the diverse range of mental models, and the disconnection between policymaking and system singularities are some of the conditions that can hinder TD efforts. The framework allowed the identification of knowledge gaps that limit the understanding of the problem complexity to be dealt with by a TD research process. There is a need to gain a better understanding of the governance system, and to characterize the different types of agro-silvo-pastoral combinations that can be designated as montado. With this detailed understanding, a tailored TD process can be designed. This work argues for the active use of the SES framework in TD in environmental management. Future research could focus on the framework’s utility in developing tools to assess and monitor transdisciplinary research
Colombia: Sistema general de seguridad social en salud. Estrategia del BID 2011-2014
Esta nota se centra en aportar la información requerida para proponer una ruta que le permita al país enfrentar los principales retos actuales del Sistema general de seguridad social en Colombia reimpulsados por la Sentencia Corte Constitucional T-760 de 2008 (ST-760), la universalización de coberturas y la unificación del paquete básico de salud. Con ese propósito en mente, la nota se ha estructurado de la siguiente manera: una primera sección que describe el modelo de aseguramiento en salud en Colombia; esto es; el Sistema General de Seguridad Social en Salud (SGSSS). La segunda sección describe los logros alcanzados desde que empezó a operar el Sistema, en afiliación, acceso, protección financiera y estado de salud.
