As a result of immunological and nucleic-acid screening of plasma donations for transfusion-transmissible viruses, and the incorporation of viral reduction processes during plasma fractionation, coagulation-factor concentrates (CFC) are now judged safe in terms of many known infectious agents, including hepatitis B and C viruses, HIV, and human T-cell lymphotropic virus. However, emerging pathogens could pose future threats, particularly those with blood-borne stages that are resistant to viral-inactivation steps in the manufacturing process, such as non-lipid-coated viruses. As outlined in this Review, better understanding of infectious diseases allows challenges from newly described agents of potential concern in the future to be anticipated, but the processes of zoonotic transmission and genetic selection or modification ensure that plasma-derived products will continue to be subject to infectious concerns. Manufacturers of plasma-derived CFC have addressed the issue of emerging infectious agents by developing recombinant products that limit the need for human plasma during production. Such recombinant products have extended the safety profile of their predecessors by ensuring that all reagents used for cell culture, purification steps, and stabilisation and storage buffers are completely independent of human plasma
Increased frequency of Tim-3 expressing T cells is associated with symptomatic West Nile virus infection
More than a decade after West Nile virus (WNV) entered North America, and despite a significant increase in reported cases during the 2012 and 2013 seasons, no treatment or vaccine for humans is available. Although antiviral T cells contribute to the control of WNV, little is known about their regulation during acute infection. We analyzed the expression of Tim-3 and PD-1, two recently identified T cell negative immune checkpoint receptors, over the course of WNV infection. Symptomatic WNV+ donors exhibited higher frequencies of Tim-3+ cells than asymptomatic subjects within naĂŻve/early differentiated CD28+/-CD57-CD4+ and differentiated CD28-CD57-CD8+ T cells. Our study links Tim-3-expression on T cells during acute WNV infection with the development of symptomatic disease, suggesting Tim-3 and its ligands could be targeted therapeutically to alter anti-WNV immunity and improve disease outcome
Carnegie Supernova Project-II: The Near-infrared Spectroscopy Program
Shifting the focus of Type Ia supernova (SN Ia) cosmology to the
near-infrared (NIR) is a promising way to significantly reduce the systematic
errors, as the strategy minimizes our reliance on the empirical
width-luminosity relation and uncertain dust laws. Observations in the NIR are
also crucial for our understanding of the origins and evolution of these
events, further improving their cosmological utility. Any future experiments in
the rest-frame NIR will require knowledge of the SN Ia NIR spectroscopic
diversity, which is currently based on a small sample of observed spectra.
Along with the accompanying paper, Phillips et al. (2018), we introduce the
Carnegie Supernova Project-II (CSP-II), to follow up nearby SNe Ia in both the
optical and the NIR. In particular, this paper focuses on the CSP-II NIR
spectroscopy program, describing the survey strategy, instrumental setups, data
reduction, sample characteristics, and future analyses on the data set. In
collaboration with the Harvard-Smithsonian Center for Astrophysics (CfA)
Supernova Group, we obtained 661 NIR spectra of 157 SNe Ia. Within this sample,
451 NIR spectra of 90 SNe Ia have corresponding CSP-II follow-up light curves.
Such a sample will allow detailed studies of the NIR spectroscopic properties
of SNe Ia, providing a different perspective on the properties of the unburned
material, radioactive and stable nickel produced, progenitor magnetic fields,
and searches for possible signatures of companion stars.Comment: 20 pages, 7 figures, accepted for publication in PAS
