Transcriptional, epigenetic and pharmacological control of JAK/STAT pathway in NK cells
Differentiation of Natural Killer (NK) cells is a stepwise process having its origin in the bone marrow and proceeding in the periphery, where these cells follow organ specific trajectories. Several soluble factors and cytokines regulate the distinct stages of NK cell differentiation, and ultimately, their functional properties. Cytokines activating the Janus kinases (JAKs) and members of the signal transducer and activator of transcription (STAT) pathway control distinct aspects of NK cell biology, ranging from development, terminal differentiation, activation, and generation of cells with adaptive properties. Here, we discuss how the recent advances of next generation sequencing (NGS) technology have led to unravel novel molecular aspects of gene regulation, with the aim to provide genomic views of how STATs regulate transcriptional and epigenetic features of NK cells during the different functional stages
The prevalence of pancreatic cystic lesions in patients with liver cirrhosis is double that in controls
Background and aims: Pancreatic cystic lesions (PCLs) are commonly diagnosed incidentally and are often preneoplastic. Their presence may influence the management of patients with chronic diseases such as liver cirrhosis (LC). This study evaluated the prevalence and nature of PCLs in LC patients. Methods: The images of 192 LC patients and 192 controls who underwent either computed tomography scan or nuclear magnetic resonance were reviewed for the presence and nature of PCLs. The prevalence of PCLs in both groups and differences between LC patients with and without PCLs were analysed. Multiple logistic regression analysis was used to investigate factors associated with PCLs. Results: Thirty-five of 192 LC patients (18%) and 19/192 controls (10%) had PCLs (p =.027). The prevalence of presumptive intraductal pancreatic mucinous neoplasm (IPMN) was double in LC patients compared with controls (14% vs 7%; p =.065). In multivariate analysis, age, LC and having undergone a magnetic resonance cholangiopancreatography were factors associated with PCLs and IPMNs. LC patients with PCLs were older at the time of imaging and had a longer history of liver disease (67 vs. 43 months; p = 0.039) compared with LC patients without PCLs. Conclusions: PCLs are more common in LC patients than in controls, and most are IPMNs. The occurrence of PCLs in LC patients seems to be related to age and disease duration
SARS-CoV-2 diagnostics in the virology laboratory of a University Hospital in Rome during the lockdown period
Italy was one of the most affected nations by coronavirus disease 2019 outside China. The infections, initially limited to Northern Italy, spread to all other Italian regions. This study aims to provide a snapshot of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) epidemiology based on a single-center laboratory experience in Rome. The study retrospectively included 6565 subjects tested for SARS-CoV-2 at the Laboratory of Virology of Sapienza University Hospital in Rome from 6 March to 4 May. A total of 9995 clinical specimens were analyzed, including nasopharyngeal swabs, bronchoalveolar lavage fluids, gargle lavages, stools, pleural fluids, and cerebrospinal fluids. Positivity to SARS-CoV-2 was detected in 8% (527/6565) of individuals, increased with age, and was higher in male patients (P <.001). The number of new confirmed cases reached a peak on 18 March and then decreased. The virus was detected in respiratory samples, in stool and in pleural fluids, while none of gargle lavage or cerebrospinal fluid samples gave a positive result. This analysis allowed to gather comprehensive information on SARS-CoV-2 epidemiology in our area, highlighting positivity variations over time and in different sex and age group and the need for a continuous surveillance of the infection, mostly because the pandemic evolution remains unknown
Beyond appearance: An unusual manifestation of isolated oral secondary syphilis
Syphilis is a sexually acquired chronic infection caused by Treponema pallidum and is characterized by a variety of clinical manifestations. The secondary stage of the disease results from the hematogenous and lymphatic dissemination of treponemes after a few weeks or months, and it is characterized by recurrent activity of the disease, with muco-cutaneous as well as systemic manifestations. Mucosal lesions range from small, superficial ulcers that resemble painless aphthae to large gray plaques, and they are generally associated with systemic manifestations of the disease. The exclusive asymptomatic oral localization not associated with general manifestations is uncommon but may actually be unrecognized and under-reported. We report a case of isolated oral manifestation as the unique presentation of secondary syphilis
Demonstration and Comparison of Operation of Photomultiplier Tubes at Liquid Argon Temperature
Liquified noble gases are widely used as a target in direct Dark Matter
searches. Signals from scintillation in the liquid, following energy deposition
from the recoil nuclei scattered by Dark Matter particles (e.g. WIMPs), should
be recorded down to very low energies by photosensors suitably designed to
operate at cryogenic temperatures. Liquid Argon based detectors for Dark Matter
searches currently implement photo multiplier tubes for signal read-out. In the
last few years PMTs with photocathodes operating down to liquid Argon
temperatures (87 K) have been specially developed with increasing Quantum
Efficiency characteristics. The most recent of these, Hamamatsu Photonics Mod.
R11065 with peak QE up to about 35%, has been extensively tested within the R&D
program of the WArP Collaboration. During these testes the Hamamatsu PMTs
showed superb performance and allowed obtaining a light yield around 7
phel/keVee in a Liquid Argon detector with a photocathodic coverage in the 12%
range, sufficient for detection of events down to few keVee of energy
deposition. This shows that this new type of PMT is suited for experimental
applications, in particular for new direct Dark Matter searches with LAr-based
experiments
Measurement of Through-Going Particle Momentum By Means Of Multiple Scattering With The ICARUS T600 TPC
The ICARUS collaboration has demonstrated, following the operation of a 600
ton (T600) detector at shallow depth, that the technique based on liquid Argon
TPCs is now mature. The study of rare events, not contemplated in the Standard
Model, can greatly benefit from the use of this kind of detectors. In
particular, a deeper understanding of atmospheric neutrino properties will be
obtained thanks to the unprecedented quality of the data ICARUS provides.
However if we concentrate on the T600 performance, most of the
charged current sample will be partially contained, due to the reduced
dimensions of the detector. In this article, we address the problem of how well
we can determine the kinematics of events having partially contained tracks.
The analysis of a large sample of atmospheric muons collected during the T600
test run demonstrate that, in case the recorded track is at least one meter
long, the muon momentum can be reconstructed by an algorithm that measures the
Multiple Coulomb Scattering along the particle's path. Moreover, we show that
momentum resolution can be improved by a factor two using an algorithm based on
the Kalman Filtering technique
Performance Of A Liquid Argon Time Projection Chamber Exposed To The WANF Neutrino Beam
We present the results of the first exposure of a Liquid Argon TPC to a
multi-GeV neutrino beam. The data have been collected with a 50 liters
ICARUS-like chamber located between the CHORUS and NOMAD experiments at the
CERN West Area Neutrino Facility (WANF). We discuss both the instrumental
performance of the detector and its capability to identify and reconstruct low
multiplicity neutrino interactions.Comment: 14 pages, 12 figures. Submitted for publication to Physical Review
Discovery of underground argon with low level of radioactive 39Ar and possible applications to WIMP dark matter detectors
We report on the first measurement of 39Ar in argon from underground natural
gas reservoirs. The gas stored in the US National Helium Reserve was found to
contain a low level of 39Ar. The ratio of 39Ar to stable argon was found to be
<=4x10-17 (84% C.L.), less than 5% the value in atmospheric argon
(39Ar/Ar=8x10-16). The total quantity of argon currently stored in the National
Helium Reserve is estimated at 1000 tons. 39Ar represents one of the most
important backgrounds in argon detectors for WIMP dark matter searches. The
findings reported demonstrate the possibility of constructing large multi-ton
argon detectors with low radioactivity suitable for WIMP dark matter searches.Comment: 6 pages, 2 figures, 2 table
Search for anomalies in the neutrino sector with muon spectrometers and large LArTPC imaging detectors at CERN
A new experiment with an intense ~2 GeV neutrino beam at CERN SPS is proposed
in order to definitely clarify the possible existence of additional neutrino
states, as pointed out by neutrino calibration source experiments, reactor and
accelerator experiments and measure the corresponding oscillation parameters.
The experiment is based on two identical LAr-TPCs complemented by magnetized
spectrometers detecting electron and muon neutrino events at Far and Near
positions, 1600 m and 300 m from the proton target, respectively. The ICARUS
T600 detector, the largest LAr-TPC ever built with a size of about 600 ton of
imaging mass, now running in the LNGS underground laboratory, will be moved at
the CERN Far position. An additional 1/4 of the T600 detector (T150) will be
constructed and located in the Near position. Two large area spectrometers will
be placed downstream of the two LAr-TPC detectors to perform charge
identification and muon momentum measurements from sub-GeV to several GeV
energy range, greatly complementing the physics capabilities. This experiment
will offer remarkable discovery potentialities, collecting a very large number
of unbiased events both in the neutrino and antineutrino channels, largely
adequate to definitely settle the origin of the observed neutrino-related
