    Why should DNA topoisomerase i have a scaffold activity?

    Since the early 1990s, in vitro studies have demonstrated that DNA topoisomerase I pro-motes RNA polymerase II transcription, acting as a cofactor, regardless of its catalytic activity. Recent studies, carried in vivo, using yeast as a model system, also demonstrate that DNA topoisomerase I is able to recruit, without the involvement of its catalytic activity, the Sir2p deacetylase on ribosomal genes thus contributes to achieve their silencing. In this review, the DNA topoisomerase I capability, acting as a scaffold protein, as well as its involvement and role in several macromolecular complexes, will be discussed, in light of several observations reported in the literature, pointing out how its role goes far beyond its well-known ability to relax DNA

    We report the inclusion of electron-electron correlation in the calculation of transport properties within an ab initio scheme. A key step is the reformulation of Landauer's approach in terms of an effective transmittance for the interacting electron system. We apply this framework to analyze the effect of short range interactions on Pt atomic wires and discuss the coherent and incoherent correction to the mean-field approach.Comment: 5 pages, 3 figure

    A multi-layer model for double-skin Pd-based membranes:Layer-by-layer parameter fitting

    The Pd-based double-skin (DS) membrane has high potential to be implemented in various industrial processes. However, its multi-layer structure makes it difficult to model and understand the limiting transport mechanisms through each of these layers. In this work an extensive set of experiments on several membranes with different configurations was performed with the purpose of deriving a detailed model for the double-skin Pd-based membrane. This is important for the general understanding of the mass-transport taking place within each of the membrane layers, namely the support, the selective and the protective layer. Characterization, permeation tests and data-fitting led to the construction and validation of a multi-layer model for the DS-membrane. The experimental results also pointed out that the protective layer of HT or YSZ/Al2O3 has only a minor or negligible effect on the overall transport resistance of the membrane. The influence of the support was found to be small but not negligible compared to the dominating resistance of the dense selective PdAg layer. Experiments and simulations of the membrane permeation in multi-component mixtures also highlighted the large importance of concentration polarization and H2 depletion, which were successfully described in the model using a film-layer model together with a Sherwood correlation.</p

    Habitat Patch Diversity Evaluation for Sustainability: a Case Study of a Rural Area in Central Italy

    Landscape analysis is regarded as a new tool for monitoring and judging land use patterns in terms of sustainability of human activity systems at local level. A case study of evaluation for sustainability based on habitat patch diversity in an ecoregion of Central Italy is presented. In this region, ongoing land use patterns reflect both historical adaptation to local environmental constraints and positive, social-oriented management. More protective land use patterns are mostly widespread in fragile physiographic conditions like those of the mountain areas, where woodland, shrub, and grassland patches are larger and cover more than 90% of the land. This situation is regarded as a positive outcome of the traditional public ownership regime, because public lands amount to more than 70% in the mountain areas. The hilly areas, where public property drops to 28%, presents landscape metrics showing a well balanced situation between agricultural land use and protective native woods and grasslands, which provides a fine-grained and harmonious Mediterranean landscape. In the low-land areas, with anthropic pressure and more favourable conditions for crop productivity, there is much more agricultural land, even if some mitigation in terms of biodiversity maintenance is offered by the presence of hedgerow ecotones. In these areas, landscape analysis is not able to supply meaningful information about cropping system design and practices which can maintain a sustainable level of soil fertility and quality of natural resources and processes, and further analysis at cropping system level should be carried out

    The need to account for all currently available experimental observations concerning the gene nature, has reshaped the concept of gene turning it from the essentially mechanistic unit, predominant during the '70s, into a quite abstract open and generalized entity, whose contour appears less defined as compared to the past. Here we propose the essence of the gene to be considered double faced. In this respect genotypic and phenotypic entities of a gene would coexist and mix reciprocally. This harmonizes present knowledge with current definitions and predisposes for remodelling of our thinking as a consequence of future discoveries. A two sided view of the gene also allows to combine the genetic and epigenetic aspects in a unique solution, being structural and functional at the same time and simultaneously able to include the different levels in an overlapping unicum

    The decline of skeletal muscle mass and strength that leads to sarcopenia is a pathology that might represent an emergency healthcare issue in future years. Decreased muscle mass is also a condition that mainly affects master athletes involved in endurance physical activities. Skeletal muscles respond to exercise by reshaping the biochemical, morphological, and physiological state of myofibrils. Adaptive responses involve the activation of intracellular signaling pathways and genetic reprogramming, causing alterations in contractile properties, metabolic status, and muscle mass. One of the mechanisms leading to sarcopenia is an increase in reactive oxygen and nitrogen species levels and a reduction in enzymatic antioxidant protection. The present review shows the recent experimental models of sarcopenia that explore molecular mechanisms. Furthermore, the clinical aspect of sport sarcopenia will be highlighted, and new strategies based on nutritional supplements, which may contribute to reducing indices of oxidative stress by reinforcing natural endogenous protection, will be suggested

    Effect of Exercise on Fatty Acid Metabolism and Adipokine Secretion in Adipose Tissue

    Increased physical activity is an optimal way to maintain a good health. During exercise, triacylglycerols, an energy reservoir in adipose tissue, are hydrolyzed to free fatty acids (FAs) which are then released to the circulation, providing a fuel for working muscles. Thus, regular physical activity leads to a reduction of adipose tissue mass and improves metabolism. However, the reduction of lipid reservoir is also associated with many other interesting changes in adipose tissue FA metabolism. For example, a prolonged exercise contributes to a decrease in lipoprotein lipase activity and resultant reduction of FA uptake. This results in the improvement of mitochondrial function and upregulation of enzymes involved in the metabolism of polyunsaturated fatty acids. The exercise-induced changes in adipocyte metabolism are associated with modifications of FA composition. The modifications are adipose tissue depot-specific and follow different patterns in visceral and subcutaneous adipose tissue. Moreover, exercise affects adipokine release from adipose tissue, and thus, may mitigate inflammation and improve insulin sensitivity. Another consequence of exercise is the recently described phenomenon of adipose tissue “beiging,” i.e., a switch from energy-storing white adipocyte phenotype to thermogenic FA oxidizing beige adipocytes. This process is regulated by myokines released during the exercise. The heterogeneity of IoT use cases usually makes the development of digital twins extremely application-specific as well as prone to interoperability issues.     A Survey on the Web of Things

    The Web of Things (WoT) paradigm was proposed first in the late 2000s, with the idea of leveraging Web standards to interconnect all types of embedded devices. More than ten years later, the fragmentation of the IoT landscape has dramatically increased as a consequence of the exponential growth of connected devices, making interoperability one of the key issues for most IoT deployments. Contextually, many studies have demonstrated the applicability of Web technologies on IoT scenarios, while the joint efforts from the academia and the industry have led to the proposals of standard specifications for developing WoT systems. Through a systematic review of the literature, we provide a detailed illustration of the WoT paradigm for both researchers and newcomers, by reconstructing the temporal evolution of key concepts and the historical trends, providing an in-depth taxonomy of software architectures and enabling technologies of WoT deployments and, finally, discussing the maturity of WoT vertical markets. Moreover, we identify some future research directions that may open the way to further innovation on WoT systems
