    Do Movement Patterns of GPS-Tracked Cattle on Extensive Rangelands Suggest Independence among Individuals?

    In behavioral studies, cattle within the same pasture are not considered as independent experimental units because of the potential confounding effects of the herd’s social interactions. However, evaluating cattle behavior on extensive rangelands is logistically challenging for researchers, and treating individual animals as independent experimental units may be beneficial for answering specific research questions. The objective of this study was to evaluate the association patterns among global positioning system (GPS)-tracked cattle at six different study sites in the western United States. A Half-Weight Index (HWI) association value was calculated for each pair of GPS-tracked cows (i.e., dyad) to determine the proportion of time that cattle were within 75 m and 500 m of each other. Cattle at two study sites exhibited relatively low mean HWI-association values (i.e., less than 0.23 HWI); whereas, cattle at other study sites tended to have greater mean HWI associations (i.e., greater than 0.35 HWI). Distinguishing features between study sites with low and high association values were the management of cattle prior to the study, herd size, pasture size, and the number of watering points. However, at all ranches except one, at least 75% of all dyadic associations had HWI values of less than 0.5 at 500 m, indicating that most of the GPS-tracked cows were greater than 500 m from each other for over 50% of tracking period. While interactions among cattle in the same pasture are often inevitable, our data suggests that under some situations, movement patterns of a sub-set of individual GPS-tracked cows may have levels of independence that are sufficient for analysis as individual experimental units. Understanding the level of independence among GPS-tracked cattle may provide options for analysis of grazing behavior for individual cattle within the same pasture

    Manipulation of the Spatial Grazing Behaviour of Livestock in Extensive Grassland Systems

    Spatial behaviour of livestock is a critical factor in grassland management. Recent and ongoing research suggests that new approaches can be used to manipulate where cattle graze. The combination of strategic supplement placement and low-stress herding can be used to target cattle grazing and potentially may be useful for managing fine fuels. A phenotype to genotype association study of cattle spatial behavior suggests that use of rugged terrain and areas far from water is inherited. Although more research is needed, selection for animals specifically adapted for mountainous terrain or extensive paddocks may be an alternative for managing grasslands in the near future

    Synthesis Paper: Targeted Livestock Grazing: Prescription for Healthy Rangelands

    Targeted livestock grazing is a proven tool for manipulating range land vegetation, and current knowledge about targeted livestock grazing is extensive and expanding rapidly. Targeted grazing prescriptions optimize the timing, frequency, intensity, and selectivity of grazing (or browsing) in combinations that purposely exert grazing/ browsing pressure on specific plant species or portions of the landscape. Targeted grazing differs from traditional grazing management in that the goal of targeted grazing is to apply defoliation or trampling to achieve specific vegetation management objectives,whereas the goal of traditional livestock grazing management is generally the production of livestock commodities. A shared aim of targeted livestock grazing and traditional grazing management is to sustain healthy soils, flora, fauna, and water resources that, in turn, can sustain natural ecological processes (e.g., nutrient cycle, water cycle, energy flow). Targeted grazing prescriptions integrate knowledge of plant ecology, livestock nutrition, and livestock foraging behavior. Livestock can be focused on target areas through fencing, herding, or supplement placement. Although practices can be developed to minimize the impact of toxins contained in target plants, the welfare of the animals used in targeted grazing must be a priority. Monitoring is needed to determine if targeted grazing is successful and to refine techniques to improve efficacy and efficiency. Examples of previous research studies and approaches are presented to highlight the ecological benefits that can be achieved when targeted grazing is applied properly. These cases include ways to suppress invasive plants and ways to enhance wildlife habitat and biodiversity. Future research should address the potential to select more adapted and effective livestock for targeted grazing and the associated animal welfare concerns with this practice. Targeted livestock grazing provides land managers a viable alternative to mechanical, chemical, and prescribed fire treatments to manipulate range land vegetation

    Macro- and micro-structural insights into primary dystonia: a UK Biobank study

    BACKGROUND: Dystonia is a hyperkinetic movement disorder with key motor network dysfunction implicated in pathophysiology. The UK Biobank encompasses > 500,000 participants, of whom 42,565 underwent brain MRI scanning. This study applied an optimized pre-processing pipeline, aimed at better accounting for artifact and improving data reliability, to assess for grey and white matter structural MRI changes between individuals diagnosed with primary dystonia and an unaffected control cohort. METHODS: Individuals with dystonia (n = 76) were identified from the UK Biobank using published algorithms, alongside an age- and sex-matched unaffected control cohort (n = 311). Grey matter morphometric and diffusion measures were assessed, together with white matter diffusion tensor and diffusion kurtosis metrics using tractography and tractometry. Post-hoc Neurite Orientation and Density Distribution Imaging (NODDI) was also undertaken for tracts in which significant differences were observed. RESULTS: Grey matter tremor-specific striatal differences were observed, with higher radial kurtosis. Tractography identified no white matter differences, however segmental tractometry identified localised differences, particularly in the superior cerebellar peduncles and anterior thalamic radiations, including higher fractional anisotropy and lower orientation distribution index in dystonia, compared to controls. Additional tremor-specific changes included lower neurite density index in the anterior thalamic radiations. CONCLUSIONS: Analysis of imaging data from one of the largest dystonia cohorts to date demonstrates microstructural differences in cerebellar and thalamic white matter connections, with architectural differences such as less orientation dispersion potentially being a component of the morphological structural changes implicated in dystonia. Distinct tremor-related imaging features are also implicated in both grey and white matter

    In Search of a Trade Mark: Search Practices and Bureaucratic Poetics

    Trade marks have been understood as quintessential ‘bureaucratic properties’. This article suggests that the making of trade marks has been historically influenced by bureaucratic practices of search and classification, which in turn were affected by the possibilities and limits of spatial organisation and technological means of access and storage. It shows how the organisation of access and retrieval did not only condition the possibility of conceiving new trade marks, but also served to delineate their intangible proprietary boundaries. Thereby they framed the very meaning of a trade mark. By advancing a historical analysis that is sensitive to shifts, both in actual materiality and in the administrative routines of trade mark law, the article highlights the legal form of trade mark as inherently social and materially shaped. We propose a historical understanding of trade mark law that regards legal practice and bureaucratic routines as being co-constitutive of the very legal object itself

    Endogenous Epoxygenases Are Modulators of Monocyte/Macrophage Activity

    Background: Arachidonic acid is metabolized through three major metabolic pathways, the cyclooxygenase, lipoxygenase and CYP450 enzyme systems. Unlike cyclooxygenase and lipoxygenases, the role of CYP450 epoxygenases in monocyte/macrophage-mediated responses is not known.Methodology/Principal Findings: When transfected in vitro, CYP2J2 is an efficient activator of anti-inflammatory pathways through the nuclear receptor peroxisome proliferator-activated receptor (PPAR) alpha. Human monocytes and macrophages contain PPAR alpha and here we show they express the epoxygenases CYP2J2 and CYP2C8. Inhibition of constitutive monocyte epoxygenases using the epoxygenase inhibitor SKF525A induces cyclooxygenase (COX)-2 expression and activity, and the release of TNF alpha, and can be reversed by either add back of the endogenous epoxygenase products and PPAR alpha ligand 11,12-epoxyeicosatrienoic acid (EET) or the addition of the selective synthetic PPAR alpha ligand GW7647. In alternatively activated (IL-4-treated) monocytes, in contrast to classically activated cells, epoxygenase inhibition decreased TNF alpha release. Epoxygenases can be pro-inflammatory via superoxide anion production. The suppression of TNF alpha by SKF525A in the presence of IL-4 was associated with a reduction in superoxide anion generation and reproduced by the superoxide dismutase MnCl2. Similar to these acute activation studies, in monocyte derived macrophages, epoxygenase inhibition elevates M1 macrophage TNF alpha mRNA and further decreases M2 macrophage TNF alpha.Conclusions/Significance: In conclusion, epoxygenase activity represents an important endogenous pathway which limits monocyte activation. Moreover endogenous epoxygenases are immuno-modulators regulating monocyte/macrophage activation depending on the underlying activation state
