    Chiral Koszul duality

    We extend the theory of chiral and factorization algebras, developed for curves by Beilinson and Drinfeld in \cite{bd}, to higher-dimensional varieties. This extension entails the development of the homotopy theory of chiral and factorization structures, in a sense analogous to Quillen's homotopy theory of differential graded Lie algebras. We prove the equivalence of higher-dimensional chiral and factorization algebras by embedding factorization algebras into a larger category of chiral commutative coalgebras, then realizing this interrelation as a chiral form of Koszul duality. We apply these techniques to rederive some fundamental results of \cite{bd} on chiral enveloping algebras of ⋆\star-Lie algebras

    Modeling DNA methylation dynamics with approaches from phylogenetics

    Methylation of CpG dinucleotides is a prevalent epigenetic modification that is required for proper development in vertebrates, and changes in CpG methylation are essential to cellular differentiation. Genome-wide DNA methylation assays have become increasingly common, and recently distinct stages across differentiating cellular lineages have been assayed. How- ever, current methods for modeling methylation dynamics do not account for the dependency structure between precursor and dependent cell types. We developed a continuous-time Markov chain approach, based on the observation that changes in methylation state over tissue differentiation can be modeled similarly to DNA nucleotide changes over evolutionary time. This model explicitly takes precursor to descendant relationships into account and enables inference of CpG methylation dynamics. To illustrate our method, we analyzed a high-resolution methylation map of the differentiation of mouse stem cells into several blood cell types. Our model can successfully infer unobserved CpG methylation states from observations at the same sites in related cell types (90% correct), and this approach more accurately reconstructs missing data than imputation based on neighboring CpGs (84% correct). Additionally, the single CpG resolution of our methylation dynamics estimates enabled us to show that DNA sequence context of CpG sites is informative about methylation dynamics across tissue differentiation. Finally, we identified genomic regions with clusters of highly dynamic CpGs and present a likely functional example. Our work establishes a framework for inference and modeling that is well-suited to DNA methylation data, and our success suggests that other methods for analyzing DNA nucleotide substitutions will also translate to the modeling of epigenetic phenomena.Comment: 8 pages, 5 figure

    Applied Technology Associates, Inc., (ATA) has demonstrated that inertial actuation can be employed effectively in digital, active vibration isolation systems. Inertial actuation involves the use of momentum exchange to produce corrective forces which act directly on the payload being actively isolated. In a typical active vibration isolation system, accelerometers are used to measure the inertial motion of the payload. The signals from the accelerometers are then used to calculate the corrective forces required to counteract, or 'cancel out' the payload motion. Active vibration isolation is common technology, but the use of inertial actuation in such systems is novel, and is the focus of the DAMPER project. A May 1991 report was completed which documented the successful demonstration of inertial actuation, employed in the control of vibration in a single axis. In the 1 degree-of-freedom (1DOF) experiment a set of air bearing rails was used to suspend the payload, simulating a microgravity environment in a single horizontal axis. Digital Signal Processor (DSP) technology was used to calculate in real time, the control law between the accelerometer signals and the inertial actuators. The data obtained from this experiment verified that as much as 20 dB of rejection could be realized by this type of system. A discussion is included of recent tests performed in which vibrations were actively controlled in three axes simultaneously. In the three degree-of-freedom (3DOF) system, the air bearings were designed in such a way that the payload is free to rotate about the azimuth axis, as well as translate in the two horizontal directions. The actuator developed for the DAMPER project has applications beyond payload isolation, including structural damping and source vibration isolation. This report includes a brief discussion of these applications, as well as a commercialization plan for the actuator

    A Robust Parsing Algorithm For Link Grammars

    In this paper we present a robust parsing algorithm based on the link grammar formalism for parsing natural languages. Our algorithm is a natural extension of the original dynamic programming recognition algorithm which recursively counts the number of linkages between two words in the input sentence. The modified algorithm uses the notion of a null link in order to allow a connection between any pair of adjacent words, regardless of their dictionary definitions. The algorithm proceeds by making three dynamic programming passes. In the first pass, the input is parsed using the original algorithm which enforces the constraints on links to ensure grammaticality. In the second pass, the total cost of each substring of words is computed, where cost is determined by the number of null links necessary to parse the substring. The final pass counts the total number of parses with minimal cost. All of the original pruning techniques have natural counterparts in the robust algorithm. When used together with memoization, these techniques enable the algorithm to run efficiently with cubic worst-case complexity. We have implemented these ideas and tested them by parsing the Switchboard corpus of conversational English. This corpus is comprised of approximately three million words of text, corresponding to more than 150 hours of transcribed speech collected from telephone conversations restricted to 70 different topics. Although only a small fraction of the sentences in this corpus are "grammatical" by standard criteria, the robust link grammar parser is able to extract relevant structure for a large portion of the sentences.     Evaluation of advanced lubricants for aircraft applications using gear surface fatigue tests

    Surface pitting fatigue life tests were conducted with five lubricants, using spur gears made from a single lot of consumable-electrode vacuum melted (CVM) AISI 9310 steel. The gears were case carbonized and hardened to a Rockwell c-60 and finish ground. The gear pitch diameter was 8.89 cm. The lot of gears was divided into five groups, each of which was tested with a different lubricant. The test lubricants can be classified as synthetic polyol-esters with various viscosities and additive packages. Test conditions included bulk gear temperature of 350 K, a maximum Hertz stress of 1.71 GPa (248 ksi) at the pitch line, and a speed of 10,000 RPM. The lubricant with a viscosity that provided a specific film thickness greater than one and with an additive package produced far greater gear surface fatigue lives than lubricants with a viscosity that provided specific film thickness less than one. A low viscosity lubricant with an additive package produced gear surface fatigue lives equivalent to a similar base stock lubricant with 30 percent higher viscosity, but without an additive package. Lubricants with the same viscosity and similar additive packages gave equivalent gear surface fatigue lives

    Effect of two synthetic lubricants on life of AISI 9310 spur gears

    Spur-gear fatigue tests were conducted with two lubricants using a single lot of consumable-electrode vacuum-melted (CVM) AISI 9310 spur gears. The gears were case carburized and hardened to Rockwell C60. The gear pitch diameter was 8.89 cm. The lot of gears was divided into two groups, each of which was tested with a different lubricant. The test lubricants can be classified as synthetic polyol-ester-based lubricants. One lubricant was 30 percent more viscous that the other. Both lubricants have similar pressure viscosity coefficients. Test conditions included a bulk gear temperature of 350 K, a maximum Hertz stress of 1.71 GPa at the pitch line, and a speed of 10,000 rpm. The surface fatigue life of gears tested with one lubricant was approximately 2.4 times that for gears tested with the other lubricant. The lubricant with the 30 percent higher viscosity gave a calculated elastohydrodynamic (EHD) film thickness that was 20 percent higher than the other lubricant. This increased EHD film thickness is the most probable reason for the improvement in surface fatigue life of gears tested with this lubricant over gears tested with the less viscous lubricant
