Mémoire en recherche-création.Le matin du 28 avril, la narratrice se présente au centre où est hébergée sa grand-mère, Aline Martin, mais celle-ci a disparu. Sous le choc, la jeune femme refuse de quitter la résidence tant que sa grand-mère n’aura pas été retrouvée. Le récit de Sa disparition se construit alors autour de fragments de discours puisés dans le champ du réel - entrevues d’aînés en résidence, forums en ligne de soutien aux familles, politiques ministérielles, rapports d’organismes, reportages journalistiques - et prend les aspects d’une enquête dont l’objectif ne sera pas tant de retrouver Aline, mais d’éclairer les discours qui se sont construits dans l’espace politique afin de justifier la disparition des personnes âgées de notre radar social. Ce récit fragmentaire s’apparente au procédé du sampling chez Olivier Cadiot par lequel il rassemble, agence et bouscule des objets oraux et textuels pour tenter de désarticuler les discours ambiants invasifs. L’essai examine cette pratique du sampling chez Cadiot en analysant trois aspects du roman Le colonel des zouaves : la subversion des citations, la structure en boucles et la fragmentation du sujet.On the morning of April 28th, the narrator arrives at the center where her grandmother, Aline Martin, has been living, only to find that she has disappeared. Deeply shaken, the young woman refuses to leave the residence until her grandmother has been found. The story of Sa disparition builds itself around fragments of discourses drawn from various sources – interviews with residents in care centres, on-line family support groups, ministry policies, reports from NGOs, journalistic coverage – and becomes an investigation whose goal is not so much to find Aline as to shed light on the discourses that have been constructed in our political space in order to justify the disappearance of the elderlies from our social radar. This fragment-based writing is related to Olivier Cadiot’s sampling method, by which he collects, organizes and shakes up oral and textual objects in an attempt to disarticulate prevailing discourses. The essay examines Cadiot’s sampling method by analysing three important aspects of his novel Le colonel des zouaves: the subversion of citations, the loop structure and the fragmentation of the subject

Rapport II(2).2. Etude de l’alimentation en métal liquide d’un poste de coulée semi-continue

The semi-continuous aluminium casting installations at the Issoire plant are provided with a flow regulator controlled by the liquid metal level in the ingot mould. The original regulators were unsatisfactory and liable to produce faulty ingots unless kept constantly under supervision. In order to remedy this state of affairs, the operation of the regulators at Issoire was studied and analysed on a water analogy model with a view to finding a means of preventing flow fluctuation and uncontrolled float movements. An ovoid valve plug was devised, which was then tried out at Issoire and was found to give a constant rate of flow with a perfectly steady float, in addition to simplifying the layout and reducing the risk of faulty operation. As the resistance of the valve plug material still remains a problem, however, only one casting station has been fitted with the ovoid version ; all the others have the same float regulating system, except that the valves have a flat shutter.Les dispositifs de coulée semi-continue d’aluminium de l’usine d’Issoire comportent un régulateur de débit commandé par le niveau de métal liquide dans la lingotière. Le mauvais fonctionnement des régulateurs anciens imposait une surveillance constante car il provoquait des défauts dans les lingots. Pour remédier à cet inconvénient, une étude sur modèle analogique à l’eau a été entreprise. Les défauts constatés à Issoire, bien reproduits sur le modèle, ont été analysés. Puis, on a procédé à la recherche d’un nouveau dispositif permettant d’éviter les fluctuations de débit et les mouvements intempestifs du flotteur. Les solutions proposées (obturateur ovoïde) ont été testées sur un banc de coulée. Les objectifs ont été atteints : débit constant et flotteur parfaitement immobile. La conduite de l’installation est simplifiée, les risques de défauts réduits. Il subsiste néanmoins un problème de tenue du matériau constituant l’obturateur. De ce fait il n’est utilisé que sur un poste de coulée. Sur les autres postes de coulée les flotteurs diffèrent simplement par l’obturateur qui est plan.Delachanal M., Bulmanski H. Rapport II(2).2. Evidence of a functional role for interaction between ICAM-1 and nonmuscle alpha-actinins in leukocyte diapedesis.

International audienceICAM-1 is involved in both adhesion and extravasation of leukocytes to endothelium during inflammation. It has been shown that the ICAM-1 cytoplasmic domain is important for transendothelial migration of leukocytes but the precise molecular mechanisms involving the intracytoplasmic portion of ICAM-1 is not known. To characterize precisely the molecular scaffolding associated with ICAM-1, we have used the yeast two-hybrid system, and we have identified six different proteins interacting with the ICAM-1 cytoplasmic domain. In this study, we report that the two forms of nonmuscle alpha-actinin (i.e., alpha-actinin 1 and alpha-actinin 4) associate with ICAM-1, and that these interactions are essential for leukocyte extravasation. These interactions were further confirmed by coimmunoprecipitation and immunofluorescence in endothelial cells and in ICAM-1-transfected Chinese hamster ovary cells. The function of these interactions was analyzed by point mutation of charged amino acids located on ICAM-1 cytoplasmic domain. We have identified three charged amino acids (arginine 480, lysine 481, and arginine 486) which are essential in the binding of alpha-actinins to the ICAM-1 cytoplasmic tail. Mutation of these amino acids completely inhibited ICAM-1-mediated diapedesis. Experiments with siRNA inhibiting specifically alpha-actinin 1 or alpha-actinin 4 on endothelial cells indicated that alpha-actinin 4 had a major role in this phenomenon. Thus, our data demonstrate that ICAM-1 directly interacts with cytoplasmic alpha-actinin 1 and 4 and that this interaction is required for leukocyte extravasation
