    Digestible lysine effects on gene expression by Japanese quails in the pre-laying phase

    This study aimed to determine the effects of digestible lysine levels in the diets of Japanese quail (Coturnix coturnix japonica) on performance, blood parameters and the expression of insulin-like growth factor I, and growth hormone receptor (GHR), apolipoprotein A-I (APOA-I), acetyl-CoA-carboxylase (ACC), and fatty acid synthase (FAS) genes. A total of 288 seven-day-old female Japanese quails were randomly assigned to one of three diets that contained 0.8%, 1.10%, or 1.40% digestible lysine. The birds were slaughtered at 42 days old, and relative gene expression was evaluated in the liver by qRT-PCR using the 2-ΔCT method. Lysine supplementation had no effect on weight gain and feed conversion. Abdominal fat was lower in birds supplemented with 0.8% digestible lysine than those supplemented with 1.10% and 1.40%. Increased total cholesterol and triglycerides were elevated in quails that received supplementation of 1.10% digestible lysine compared with the other diets. High density lipoproteins were decreased in birds that received 0.8% digestible lysine. Quails fed with 1.40% digestible lysine had greater expression of GHR and APOA-I than quails fed diets with 0.8 and 1.10% (P <0.05). The greatest expressions of ACC and FAS were observed in the liver of quails fed with 0.8% digestible lysine. The current results suggest that lysine supplementation in the pre-laying phase allows birds to deposit muscle mass to reach the optimal conformation and body fatness that provides an energetic reserve for the productive phase by modulating the expression of genes related to growth and lipid metabolism.     Inclusion of shrimp waste meal in diet of free-range chickens

    Shrimp waste meal (SWM) is a by-product from the processing of shrimp for human consumption. The value of SMW in feeding poultry is not well documented. The objective of this study was to determine the energy value and optimal inclusion level of SWM in the diet of growing chickens. A total of 180 one-day-old broilers were randomly assigned to five treatments with 0, 50, 100, 150 and 200 g/kg of SWM included in their diet. There were six replicates of six birds for each treatment. Dry matter intake (DMI) was not affected by the level of SWM that was fed. Retained dry matter varied from 72.39% in the diet that did not contain SWM to 66.97% in the diet with 200 g/kg of SWM. Nitrogen retention (NR) ranged from 54.70% to 70.10%; N ingested was between 18.71% and 24.03%. Energy intake ranged from 73.57% to 69.33% for the control and the diet with 200 g/kg of SWM, respectively. NR improved with increasing SWM inclusion levels. The apparent metabolizable energy (AME) and corrected apparent energy metabolizable (AMEn) ranged from 2928 to 2527 kcal/kg and 2774 to 2329 kcal/kg, respectively, relative to the control and 200 g/kg SWM diets. The energy consumption, in kcal/kg, of SWM consumed was AME = 2332-6.971 x SWM and AMEn = 2113-8.128 x SWM. High levels of SWM reduce the dry matter metabolization coefficient and metabolizable energy values in broilers during the growing phase, so it is recommended that up to 100 g/kg should be included, which would provide an AMEn of 1300.2 kcal/kg for free-range chickens in dry matter

    Antioxidant activity and duodenum transporter gene expression in quail fed citric acid

    The study aimed to evaluate the effects of citric acid supplementation on the performance and the gene expression in the duodenum of Japanese quail. Genes related to antioxidant activity: superoxide dismutase (SOD) and glutathione peroxidase 7 (GPX7); and genes related to nutrient uptake in the enterocytes’ apical membrane: sodium-glucose cotransporter 1 (SGLT1, responsible for the glucose absorption), glucose transporter 2 (GLUT2) and B0AT1 (responsible for the absorption of neutral amino acids in brush board membrane) were evaluated. For this, 270 laying quails (Coturnix japonica) were fed a diet supplemented with 0%, 0.6% or 1.2% of citric acid. Gene expression was evaluated by the quantitative reverse transcription-polymerase chain reaction (qRT-PCR) at 35 days old (n = 5). The highest expression of the SGLT1 gene was observed in quails fed the 1.2% citric acid supplemented diet (0.120 AU). Birds fed diets with both levels of citric acid supplementation showed a higher B0AT1 gene expression than quails fed no citric acid. Quails fed the 1.2% of citric acid supplemented diet had the lowest SOD gene expression (0.3455 AU). The birds receiving the diets supplemented with citric acid showed a lower GPX7 gene expression than the quails fed the 0% citric acid diet. These results suggests that the effect of citric acid on gene expression in the digestive tract may have contributed to the greater weight gain and lower feed intake observed in birds fed diets supplemented with citric acid.Keywords: B0AT1, intestinal health, organic acid, pathogenic microorganisms, SGLT

    Mitochondrial gene expression in the liver and muscle of high and low feed efficiency Japanese quail layers subjected to different environmental temperatures

    We evaluated the adenine nucleotide translocator (ANT) and cytochrome oxidase subunit III (COX III) mRNA expressions in the muscle and liver of Japanese quails presenting high and low feed efficiency (FE), and subjected them to three different environmental temperatures: comfort, heat stress (38°C), and cold stress (10°C). ANT mRNA expression was lower in the liver of heat-stressed animals. In the muscle, higher ANT and COX III mRNA expressions were observed in high-FE and cold-stressed animals. In the liver, much higher expression of COX III mRNA was observed in cold-stressed animals. These results suggest a possible correlation between the genes involved in energy production by the mitochondria and FE phenotypes, and that environmental temperature can affect the ANT and COX III mRNA expressions.
