    Mission description and in-flight operations of ERBE instruments on ERBS, NOAA 9, and NOAA 10 spacecraft

    Instruments of the Earth Radiation Budget Experiment (ERBE) are operating on three different Earth-orbiting spacecraft. The Earth Radiation Budget Satellite (ERBS) is operated by NASA, and NOAA 9 and NOAA 10 weather satellites are operated by the National Oceanic and Atmospheric Administration (NOAA). This paper is the second in a series that describes the ERBE mission, and data processing and validation procedures. This paper describes the spacecraft and instrument operations for the second full year of in-orbit operations, which extend from February 1986 through January 1987. Validation and archival of radiation measurements made by ERBE instruments during this second year of operation were completed in July 1991. This period includes the only time, November 1986 through January 1987, during which all ERBE instruments aboard the ERBE, NOAA 9, and NOAA 10 spacecraft were simultaneously operational. This paper covers normal and special operations of the spacecraft and instruments, operational anomalies, and the responses of the instruments to in-orbit and seasonal variations in the solar environment

    Profiles of Disruptive Behavior Across Early Childhood: Contributions of Frustration Reactivity, Physiological Regulation, and Maternal Behavior

    Disruptive behavior, including aggression, defiance, and temper tantrums, typically peaks in early toddlerhood and decreases by school entry; however, some children do not show this normative decline. The current study examined disruptive behavior in 318 boys and girls at 2, 4, and 5 years of age and frustration reactivity, physiological regulation, and maternal behavior in the laboratory at 2 years of age. A latent profile analysis (LPA) resulted in 4 longitudinal profiles of disruptive behavior, which were differentiated by interactions between reactivity, regulation, and maternal behavior. A high profile was associated with high reactivity combined with high maternal control or low regulation combined with low maternal control. Results are discussed from a developmental psychopathology perspective

    Longitudinal stability of temperamental exuberance and social-emotional outcomes in early childhood

    The goals of the current study were to investigate the stability of temperamental exuberance across infancy and toddlerhood and to examine the associations between exuberance and social-emotional outcomes in early childhood. The sample consisted of 291 4-month-olds followed at 9, 24, and 36 months and again at 5 years of age. Behavioral measures of exuberance were collected at 9, 24, and 36 months. At 36 months, frontal electroencephalogram (EEG) asymmetry was assessed. At 5 years, maternal reports of temperament and behavior problems were collected, as were observational measures of social behavior during an interaction with an unfamiliar peer in the laboratory. Latent profile analysis revealed a high, stable exuberance profile that was associated with greater ratings of 5-year externalizing behavior and surgency, as well as observed disruptive behavior and social competence with unfamiliar peers. These associations were particularly true for children who displayed left frontal EEG asymmetry. Multiple factors supported an approach bias for exuberant temperament but did not differentiate between adaptive and maladaptive social-emotional outcomes at 5 years of age

    NKp46<sup>+</sup>CD3<sup>+</sup> Cells: A Novel Nonconventional T Cell Subsetin Cattle Exhibiting Both NK Cell and T Cell Features

    The NKp46 receptor demonstrates a high degree of lineage-specificity, being expressed almost exclusively in natural killer cells. Previous studies have demonstrated NKp46 expression by T-cells, but NKp46(+)CD3(+) cells are rare and almost universally associated with NKp46 acquisition by T-cells following stimulation. In this study we demonstrate the existence of a population of NKp46(+)CD3(+) cells resident in normal bovine PBMC which include cells of both the αβ TCR(+) and γδ TCR(+) lineages and is present at a frequency of 0.1-1.7%. NKp46(+)CD3(+) cells express transcripts for a broad repertoire of both natural killer (NKR) and T-cell receptors (TCR) and also the CD3ζ, DAP10 and FcεR1γ but not DAP12 adaptor proteins. In vitro functional analysis of NKp46(+)CD3(+) cells confirm that NKp46, CD16 and CD3 signalling pathways are all functionally competent and capable of mediating-re-direct cytolysis. However, only CD3 cross-ligation elicits IFN-γ release. NKp46(+)CD3(+) cells exhibit cytotoxic activity against autologous Theileria parva infected cells in vitro and during in vivo challenge with this parasite an expansion of NKp46(+)CD3(+) cells was observed in some animals, indicating the cells have the potential to act as an anti-pathogen effector population. The results presented herein identifies and describes a novel non-conventional NKp46(+)CD3(+) T-cell subset that is phenotypically and functionally distinct from conventional NK and T-cells. The ability to exploit both NKR and TCR suggests these cells may fill a functional niche at the interface of innate and adaptive immune responses

    A Post-Synaptic Scaffold at the Origin of the Animal Kingdom

    The evolution of complex sub-cellular structures such as the synapse requires the assembly of multiple proteins, each conferring added functionality to the integrated structure. Tracking the early evolution of synapses has not been possible without genomic information from the earliest branching animals. As the closest extant relatives to the Eumetazoa, Porifera (sponges) represent a pivotal group for understanding the evolution of nervous systems, because sponges lack neurons with clearly recognizable synapses, in contrast to eumetazoan animals.We show that the genome of the demosponge Amphimedon queenslandica possesses a nearly complete set of post-synaptic protein homologs whose conserved interaction motifs suggest assembly into a complex structure. In the critical synaptic scaffold gene, dlg, residues that make hydrogen bonds and van der Waals interactions with the PDZ ligand are 100% conserved between sponge and human, as is the motif organization of the scaffolds. Expression in Amphimedon of multiple post-synaptic gene homologs in larval flask cells further supports the existence of an assembled structure. Among the few post-synaptic genes absent from Amphimedon, but present in Eumetazoa, are receptor genes including the entire ionotropic glutamate receptor family.Highly conserved protein interaction motifs and co-expression in sponges of multiple proteins whose homologs interact in eumetazoan synapses indicate that a complex protein scaffold was present at the origin of animals, perhaps predating nervous systems. A relatively small number of crucial innovations to this pre-existing structure may represent the founding changes that led to a post-synaptic element
