    Comparação entre duas técnicas de biópsia renal guiadas por laparoscopia em eqüinos

    A biópsia renal é técnica de diagnóstico útil para identificação do tipo e severidade e acompanhamento da evolução da doença renal. A alta incidência de lesões renais em eqüinos, geralmente em conseqüência de distúrbios digestivos ou respiratórios, freqüentemente associados a endotoxemia, nos motivou a estudar a técnica de biópsia renal guiada por laparoscopia, comparando o uso de agulha com pinça de biópsia. Foram utilizados 10 eqüinos hígidos, distribuídos aleatoriamente em dois grupos. Em cinco animais foi realizada biópsia renal com agulha descartável do tipo tru-cut e nos outros cinco com pinça de biópsia de 5mm, sendo que o procedimento foi guiado em ambos os grupos com auxílio da laparoscopia. Os animais permaneceram em estação durante a biópsia, que foi realizada mediante sedação e analgesia e bloqueio anestésico local em 2 pontos no flanco esquerdo, sendo um para introdução do trocarte e laparoscópio e outro para introdução da agulha ou pinça. As técnicas foram comparadas quanto à segurança, intensidade do sangramentro, tempo de execução e significância da amostra colhida. Em todos os procedimentos, foi possível a inspeção da cavidade peritoneal e escolha do local de colheita do material, com ausência de acidentes ou complicações o que nos permitiu validar ambas as técnicas. A análise histológica dos fragmentos demonstrou maior porcentagem de biópsias consideradas adequadas no grupo em que foi utilizada pinça laparoscópica o que nos leva a indicar preferencialmente seu uso.Renal biopsy is a useful diagnostic technique for the identification to the type of lesion, severity of the disease and its evolution. The high incidence of renal lesions in horses, which usually are a consequence of digestive and respiratory diseases and are frequently associated with endotoxemia, prompted us to study the laparoscopic technique for renal biopsy compair the use of needle and forceps. For the purpose of this study ten helthy horses were randomly chosen and divided into two grups of five animals each: in one group laparoscopic guided renal biops was performed using tru-cut needles and in the other group using 5mm laparoscopic forceps. Procedures were carried out with the animals in standing position. Animals were given a sedative analgesic combination and local anesthetic was infiltrated in two specific points on the left flank. One point was used to introduce the trocar and laparoscopic, the other was used to introduce the forceps or the needle. The techniques were compared as regards safety, intensity of bleeding, time for execution and significance of the sample obtained. In all of the procedures, were possible to inspect the abdominal cavity, and chose the place to collect samples, without acidents or complications. The histological analysis of the sample showed a higher percentage of adequate biopsies in the group where laparoscopic forceps were used that permit us to viable both of them with preference for the forcps use

    Uso da pele de tilápia (Oreochromis niloticus) como curativo biológico oclusivo em feridas de equinos:: nota prévia

    Tilapia skin is being already use in humans and wild animals present burning wounds and showed a great result. The objective is to evaluate if tilapia skin used as an occlusive curative improves equine wound healing in two horses present chronic wound. Both animals are males, adults, both of breed Mangalarga Marchador South America. Every seven days wound we measured, photographed, biopsied for histopathological analysis, cleaned and tilapia curative was changed. Image J software was used to measure wound area. Tilapia skin as an occlusive biological factor seemed to improve healing process, wounds present an area reduction and clinical improvement during 35 days treatment, even though is still waiting for complete wound healing. In equine tilapia skin curative seemed to speed up healing process and allowed reduced curative change from every two days to once a week. This implies in decrease animal´s stress, less pain and treatment cost reduction since we used less bandage amount. Beside that tilapia skin industrial waste. Furthermore, it allowed avoid using antibiotics, which reduces environment pollution and there´s no antibiotic resistance issues.  A pele de tilápia está sendo utilizada em humanos e animais silvestres com feridas por queimadura demonstrando um excelente resultado. O objetivo do estudo é avaliar se a pele de tilápia utilizada como curativo oclusivo melhora o processo de cicatrização em dois equinos machos adultos da raça Mangalarga Machador, os quais apresentam feridas crônicas. A cada 7 dias as feridas eram medidas, fotografadas, biopsiadas para a análise histopatológica, limpas e o curativo de pele de tilápia trocado. O programa Image J foi utilizado para calcular a área da ferida. A pele de tilápia utilizada como curativo oclusivo parece ter um efeito positivo na cicatrização das feridas, a área diminuiu e a o aspecto clínico melhorou nos 35 dias avaliados, no entanto é necessário esperar pela cicatrização completa das feridas. Em equinos, a utilização de curativo de pele de tilápia parece diminuir o tempo de cicatrização e permite a redução do número de trocas de curativos de a cada dois dias para uma vez por semana. Isso implica em menos estresse e dor para o animal devido à menor manipulação e menor custo de tratamento, pois há menor quantidade de material de curativo sendo utilizado. Além disso, permite evitar a utilização de antibióticos, o que diminui o impacto ambiental e não gera resistência.&nbsp

    A survey on equine neoplasias over a 15-year period in a veterinary hospital

    Apesar de vários estudos fornecerem informações sobre a incidência de neoplasias em equinos, há grande variação na distribuição, em decorrência de diferenças regionais, como clima e população racial. Foram analisados 133 casos de neoplasias dentre os 6669 equídeos atendidos no Hospital Veterinário da Faculdade de Medicina Veterinária e Zootecnia da Universidade de São Paulo, durante um período de 15 anos, representando 2% do total de atendimentos. A neoplasia mais frequente foi o carcinoma de células escamosas (45%), diferindo da maioria dos estudos realizados mundialmente. No sistema tegumentar, a neoplasia mais comum foi o melanoma (34,2%); nos sistemas ocular e urogenital, foi o carcinoma de células escamosas (76,9% e 40,9% respectivamente). Em 68% dos casos foi realizado tratamento cirúrgico, em 10%, tratamento clínico e em 22%, nenhum tipo de tratamento, por opção do proprietário ou indicação de eutanásia. Óbitos por causas relacionadas às neoplasias ocorreram em 14% dos casos. Os achados sugerem que o carcinoma de células escamosas ocorre mais frequentemente em nosso atendimento hospitalar. Este fato pode estar relacionado à exposição prolongada à luz ultravioleta, cuja quantidade de radiação em regiões tropicais vem aumentando consideravelmente nas últimas décadas.Although several prior studies have provided information regarding incidence of tumors in horses, there is a wide variation due to regional differences, such as climate and breed population. A number of 133 cases involving tumors was found among the 6669 equides submitted to the Veterinary Hospital (HOVET) of the School of Veterinary Medicine of the University of São Paulo, during a 15 years period, representing 2% of all records. The most commonly found neoplasm was the squamous cells carcinoma (45%), different from most of the studies worldwide. The most common neoplasia in the integument system was melanoma (34,2%) and in the ocular (76,9%) and urogenital systems (40,9%) was squamous cells carcinoma. Surgical intervention was recommended in 68% of the cases, while a clinical approach was chosen in 10% of them. Twenty two percent of the horses were not treated due to of the owner option or euthanasia indication. Death associated with tumors occurred on 14% of the animals during the hospitalization period or treatment. The squamous cell carcinoma occurs more often in our veterinary hospital and may be related to prolonged exposure to ultraviolet radiation, whose amount in the tropics has increased considerably in the last decades

    In vitro evaluation of hydroxyapatite, chitosan, and carbon nanotube composite biomaterial to support bone healing

    O compósito à base de hidroxiapatita, quitosana e nanotubo de carbono foi desenvolvido com o intuito de auxiliar na consolidação óssea. Estudos anteriores sugerem que a combinação de substitutos ósseos e células-tronco mesenquimais (CTM) podem auxiliar a potencializar e promover a regeneração óssea. No presente estudo, o biomaterial foi desenvolvido e a viabilidade e a citotoxicidade de células Vero (ATCC CCL-81) e CTM obtidas de medula óssea provenientes de ovinos utilizando ensaios metil-tiazol-tetrazólio, MTT e iodeto de propídeo (PI) foram avaliadas em diferentes concentrações de extrato desse compósito. O compósito demonstrou ausência de citotoxicidade com comportamento semelhante ao grupo controle. Amostras com 50% e 10% de concentração de extrato do compósito mostraram resultados maiores comparados ao grupo controle (ensaio MTT). Esses resultados também sugerem que a presença do biomaterial pode ser utilizada em associação a CTM. Assim, esse estudo conclui que o compósito apresentado de hidroxiapatita, quitosana e nanotubo de cabono não foi considerado citotóxico e pode ser utilizado em teste in vivo.Hydroxyapatite, chitosan, and carbon nanotube composite biomaterial were developed to improve bone healing. Previous studies suggested that a combination of biomaterials and mesenchymal stem cells (MSCs) can potentially help promote bone regeneration. In the present study, we first developed hydroxyapatite, chitosan, and carbon nanotube composite biomaterial. Then, the effect of different concentrations of the extract on the viability of Vero cells (ATCC CCL-81) and MSCs obtained from sheep bone marrow using methylthiazol tetrazolium (MTT) and propidium iodide (PI) assays were evaluated. The biomaterial group demonstrated an absence of cytotoxicity, similar to the control group. Samples with 50% and 10% biomaterial extract concentrations showed higher cell viability compared to samples from the control group (MTT assay). These results suggest that the presence of this composite biomaterial can be used with MSCs. This study also concluded that hydroxyapatite, chitosan, and carbon nanotube composite biomaterial were not cytotoxic. Therefore, these could be used for performing in vivo tests

    Characterization of equine adipose tissue-derived progenitor cells before and after cryopreservation

    In horses, stem cell therapies are a promising tool to the treatment of many injuries, which are common consequences of athletic endeavor, resulting in high morbidity and often compromising the performance. In spite of many advantages, the isolation of stem cells similar to human, from equine adipose tissue, occurred only recently. The aim of this study was to isolate equine adipose tissue derived progenitor cells (eAT-PC), to characterize their proliferative potential, and to study their differentiation capacity before and after cryopreservation. The cells, isolated from horse adipose tissue, presented similar fibroblast-like cell morphology in vitro. Their proliferation rate was evaluated during 63 days (23 passages) before and after cryopreservation. After the induction of osteogenic differentiation, von Kossa staining and positive immunostaining studies revealed the formation of calcified extracellular matrix confirming the osteogenic potential of these cells. Adipogenic differentiation was induced using two protocols: routine and other one developed by us, while our protocol requires a shorter time (Oil Red O staining revealed significant accumulation of lipid droplets after 7 days). Chondrogenic differentiation was observed after 21 days of induced pellet culture, as evidenced by histological (toluidine blue) and immunohistochemistry studies. Our data demonstrate that eAT-PC can be easily isolated and successfully expanded in vitro while presenting significant proliferating rate. These cells can be maintained undifferentiated in vitro and can efficiently undergo differentiation at least into mesodermal derivates. These eAT-PC properties were preserved even after cryopreservation. Our findings classify eAT-PC as a promising type of progenitor cells that can be applied in different cell therapies in equine

    Primary Paranasal Maxillary Hematoma: Clinical Characteristics, Diagnosis and Surgical Treatment

    Background: The equine paranasal sinus have a complex anatomy and large compartiments. For this reason, deseases that affect these structures may develop for long periods before the animal show any clinical signs, making it difficult to stablish a definitive diagnosis and institute an adequate treatment.  Usually, maxillary hematomas reports come from progression of ethmoidal hematomas, and the descriptions of primary maxillary hematomas are rare. This study aims to report the clinical features, diagnosis and treatment of a case of a maxillary hematoma not associated with ethmoidal turbinates.Case: An 8-year-old male horse, Mangalarga Paulista, was referred to the Centro de Apoio ao Ensino e Pesquisa FMVZ-USP with history of nasal bleeding for over a year. The animal presented deformity on the right side of the face, with significant volume increase on the maxillary bone region and dull sound at percussion, in addition to great painfull sensibility when palpated. The right nare had no airflow, suggesting complete obstruction of the right nasal cavity. In order to better evaluate, endoscopic and radiographic exams were performed. At the radiographic exam, in ventrodorsal projection, it was observed an increase of volume and radiopacity, occupying the right antimer of the nasal cavity, with left nasal septum deviation. In the right dorsoventral oblique projection, it was observed the filling of the rostral and caudal maxillary sinus with the content radiopacity as previously described. At endoscopy exam of the right nasal cavity, it was observed a rounded greenish structure on the middle meatus The diagnosis of maxillary paranasal sinus cist was suggested and surgical removal, through maxillary sinusotomy was recommended. During the surgical procedure, it was noted that the structure previously observed, was not a cavitary organization filled with liquid, but a deorganized, dark and friable mass, like a hematoma.   A fragment was sent to histopathological evaluation, which revealed the presence of intact eythrocytes interspersed by fibrilar eosinophilic material (fibrin). There were no indications of an infectious or neoplastic process. The diagnosis of maxillary hematoma was concluded. Forty days after admission, the patient had no surgical complications and showed adequate respiratory flow, at which point the animal was discharged.Discussion: Primary maxillary hematomas are rare on equines. Although benign, it has destructives and expansives characteristics, and for this reason, it can be confused with malignant tumor formation. It has unknown ethiology, and the clinical signs observed are diverse and non specific, which makes it hard to stablish a definitive diagnosis based only on the clinical manifestations of the animal. In this particular case presented, the diagnosis was defined from the hitophatological examination. Initially the image exams (radiography and endoscopy) were usefull to stablish differential diagnosis, as well as the extention of the sinus mass. The surgery was performed with the animal on quadrupedal position, under sedation and local anethesia, in order to dimish haemorrhage occurance during the procedure. This paper describes a primary maxillary hematoma on a horse, characterizing it as to the clinical and pathological manifestations, as well as its surgical treatment. Although rare descriptions and with non specific clinical signs, it is suggested that its occurrence is superior to that described in the literature.
