    Heat-shock pretreatment inhibits sorbitol-induced apoptosis in K562, U937 and HeLa cells.

    The aim of this study was to determine whether heat-shock pretreatment exerted a protective effect against sorbitol-induced apoptotic cell death in K562, U937 and HeLa cell lines and whether such protection was associated with a decreased cytochrome c release from mithocondria and a decreased activation of caspase-9 and -3. Following heat-shock pretreatment (42 6 0.3C for 1 hr), these cell lines were exposed to sorbitol for 1 hr. Apoptosis was evaluated by DNA fragmentation, whereas caspase-9,-3 activation, cytochrome c release and heat-shock protein70 (HSP70) were assayed by Western Blot. Sorbitol exposure-induced apoptosis in these different cell lines with a marked activation of caspase-9 and caspase- 3, whereas heat-shock pretreatment before sorbitol exposure, induced expression of HSP70 and inhibited sorbitol-mediated cytochrome c release and subsequent activation of caspase-9 and caspase- 3. Similarly, overexpression of HSP70 in the three cell lines studied prevented caspase-9 cleavage and activation as well as cell death. Furthermore, we showed that the mRNA expression of iNOS decreased during both the heat-shock treatment and heat-shock pretreatment before sorbitol exposure. By contrast, the expression of Cu-Zn superoxide dismutase (SOD) and Mn-SOD proteins increased during heat-shock pretreatment before sorbitol exposure. We conclude that, heat-shock pretreatment protects different cell lines against sorbitol-induced apoptosis through a mechanism that is likely to involve SOD family members

    Exposition aux facteurs de stress et impact psychosocial du tremblement de terre de Tohoku et de ses conséquences sur les Français présents au Japon en mars 2011 - Étude qualitative

    The Great East Japan Earthquake and Tsunami of the 11 th of March 2011 was followed by a nuclear accident. This study aim to identify the exposure to stress, the psychosocial impact and information need of the French Nationals who were in Japan at this time as well as the care they were looking for. A qualitative study based on 10 semi-structured interviews of French nationals was carried out between June and October 2013. They were chosen among the French who answered to the registry launched by the InVS considering the importance of the event. The interviews were face to face (7 in France, 2 in Japan) except for one in Japan that was performed by videoconference.This study confirms the impact of this disaster on real life experience. Some interviewed relate anxious and thymic manifestations or impacts on personality that might lead to seek care, even two years after the events. Beyond stress factors usually linked to natural disaster, this study showed that the issue of information was considerable, in particular about nuclear contamination that makes the feeling of worrying long lasting. Living abroad is a specificity that modifies the experience of such extreme events. This experience influenced the life-course of the exposed. These results will be useful for future post-disaster epidemiological studies. They plaid also for registering involved people as well as preparing appropriate and sustained information and mental health screening which allows people to stay active in their choices and decisionsLe séisme survenu au Japon le 11 mars 2011 a entraîné un tsunami et un accident nucléaire. L’objectif de cette étude est de mieux connaître, pour les ressortissants français présents, les expositions aux facteurs de stress liés à ces événements, ainsi que l’impact psychosocial et les besoins d’information et de prise en charge qui en ont résulté.Une étude qualitative par entretiens semi-directifs a été réalisée auprès d’une dizaine de personnes sélectionnées parmi celles qui avaient répondu à l’enregistrement proposé par l’InVS. Les entretiens se sont déroulés entre juin et octobre 2013 en face à face (7 en France, 2 au Japon) ou par visioconférence pour une personne domiciliée au Japon. Les résultats confirment le vécu bouleversant de cette catastrophe multiple pouvant contribuer, parfois à distance de l’événement, à la survenue de manifestations anxieuses, thymiques ou des modifications de la personnalité qui peuvent nécessiter un recours aux soins. Les facteurs de stress classiquement identifiés en rapport avec les catastrophes naturelles sont retrouvés. La question de l’information a été centrale, notamment au sujet de la contamination radiologique suscitant une inquiétude prolongée. L’expatriation est une situation particulière qui modifie les modalités de vécu de tels événements. Cette expérience a sensiblement influencé ou précipité des décisions relatives à l’existence. Ces résultats vont être utilisés pour planifier les études épidémiologiques post-catastrophe. Ils confirment l'importance d'un enregistrement initial des personnes impliquées et d'une offre d’information et de dépistage adaptée et maintenue dans le long terme qui donne aux personnes exposées la possibilité de rester actives dans leurs choix et leurs décisions

    Promyelocytic leukemia zinc finger protein (PLZF) is a protein involved in various signaling, growth regulatory, and differentiation pathways, including development/function of some T cells. Here, we aimed at the detection of PLZF during colorectal carcinogenesis, using immunofluorescence, and at the evaluation of the colocalization of PLZF with CD2 and CD56 positive cells (T, \u3b3\u3b4, NK, and NKT cells), using confocal-microscopy, along colorectal carcinogenesis, since its earliest stages, that is, dysplastic aberrant crypt foci (ACF). Furthermore, we analyzed PLZF in the normal colonic mucosa (NM) according to anthropometric parameters of the subject. NM exhibited strong CD56 fluorescent staining. This infiltration was lost in both ACF and colorectal carcinoma (CRC), while PLZF presence increased from NM to ACF and CRC. Strong association was found between CD56+ colonic mucosa cell infiltration and body mass index. Interestingly, an increased stromal PLZF-reactivity was present in NM of obese subjects. This study shows that overexpression of PLZF and exclusion of NK cells in dysplastic microenvironment are very early events in the stepwise sequence leading to CRC and that lower levels of CD56+ cells in NM, together with increased levels of PLZF+ cells, can be a reflection of colon cancer risk due to obesity

    Transcribed-ultra conserved region expression profiling from low-input total RNA

    <p>Abstract</p> <p>Background</p> <p>Ultra Conserved Regions (UCRs) are a class of 481 noncoding sequences located in both intra- and inter-genic regions of the genome. The recent findings that they are significantly altered in adult chronic lymphocytic leukemias, carcinomas, and pediatric neuroblastomas lead to the hypothesis that UCRs may play a role in tumorigenesis.</p> <p>Results</p> <p>We present a novel application of Ribo-SPIA™ isothermal linear amplification of minute RNA quantities for quantifying Transcribed-UCR (T-UCR) expression by quantitative PCR. Direct comparison of non-amplified with amplified cDNA in two neuroblastoma cell lines showed that the amplification approach increases sensitivity and repeatability in T-UCR quantification. It is noteworthy that the Ribo-SPIA™ step allowed us to analyze all 481 T-UCRs by using 150 ng of RNA, while introducing a minimal bias and preserving the magnitude of relative expression. Only the less abundant T-UCRs have high intra-assay variability, consistently with the Poisson distribution statistics and stochastic effects on PCR repeatability.</p> <p>Conclusions</p> <p>We demonstrated that the quantification procedure shown here is an accurate and reliable technique for genome-wide non coding gene (i.e., UCRs) profiling using small amounts of RNA. This issue is particularly important because studies of transcription regulation are increasingly conducted in small homogeneous samples, such as laser capture microdissected or sorted cell populations.</p

    Radiofrequency echographic multispectrometry (REMS): an innovative technique for the assessment of bone status in young women with anorexia nervosa

    Purpose Reduced bone mineral density (BMD) and increase risk of fragility fracture are common complication of anorexia nervosa (AN). BMD by dual-energy X-ray absorptiometry (DXA) present several limits in subjects with AN. This study aimed to evaluate the usefulness of the new Radiofrequency echographic multispectrometry (REMS) technique in the assessment of bone status in young women with AN. Methods In a cohort of 50 subjects with restrictive AN and in 30 healthy controls, we measured BMD at the lumbar spine (LS-BMD), at femoral neck (FN-BMD) and total hip (TH-BMD) using both DXA and REMS technique. Results BMD evaluated by DXA and REMS technique at all measurement sites were all significantly (p &lt; 0.01) lower in subjects suffering from AN subjects than in controls. Good correlations were detected between BMD by DXA and BMD by REMS measurements at LS (r = 0.64, p &lt; 0.01) at FN (r = 0.86, p &lt; 0.01) and at TH (r = 0.84, p &lt; 0.01) in subjects suffering from AN. Moreover, Bland-Altman analysis confirmed the good agreement between the two techniques. The subjects suffering from AN with previous vertebral fragility fractures presented lower values of both BMD-LS and BMD-TH by DXA and by REMS with respect to those without fractures; however, the difference was significant only for BMD-TH by REMS (p &lt; 0.05). Conclusions Our data suggest that REMS technique due to its characteristic of precision and reproducibility may represent an important tool for the evaluation of the changes in bone status in AN young women, especially during the fertile age and in case of pregnancy and breastfeeding. © 2022, The Author(s)

    Diagnosis, treatment, and follow-up of patients with cerebral amyloid angiopathy-related inflammation

    Purpose Cerebral amyloid angiopathy-related inflammation (CAA-ri) is a rare potentially reversible encephalopathy associated with an autoimmune process against proteins deposited in the walls of cortical and leptomeningeal brain vessels. Definite diagnosis requires histopathological features of vascular inflammation and amyloid deposition from brain biopsy. Clinical-neuroradiological criteria have been recently introduced and validated to reduce the need for biopsy. The purpose of this paper is to report a historical retrospective review of clinical-neuroradiological follow-up of two patients with probable CAA-ri and five patients with a reasonably probable suspect of CAA-ri (4 females, 3 males, patient's age at admission: 66-79 years) seen at our institution between 2007 and 2021, focusing on clinical and neuroradiological awareness to this entity and variable response to immunotherapy. Materials and methods Clinical features at presentation included subacute to acute confusion (6/7), seizures (4/7), cognitive impairment (5/7), and focal neurological signs (3/7). Neuroradiology included braincomputed tomography followed by magnetic resonance imaging. Infectious diseases and autoimmune workups were then performed. Results CSF analysis was performed in two patients. Cerebral angiography was performed in two patients, to rule out vascular malformations. Hemorrhagic posterior reversible encephalopathy syndrome has been suspected in two patients. Four patients underwent immunotherapy with corticosteroids followed by reduction of brain dysfunctions. Three patients did not undergo immunotherapy but underwent clinical and/or neuroradiological remission. Conclusions Patients with CAA-ri present a rare steroid-responsive acute to subacute brain dysfunction. Thus, it has to be known and recognized both clinically and neuroradiologically. Spontaneous clinical and/or neuroradiological improvement is possible in patients with mild symptoms

    ESCOLA E COMUNIDADE: PENSANDO EM PARCERIAS PARA A EDUCAÇÃO EM VALORES

    In this paper we will discuss the possible relationship between Values Education projects developed in public schools and the effect on their surrounding communities. We investigated these relationships through the use of specific literature and analyzed 169 experiments of Moral Education collected through questionnaires in public secondary schools (6th to 9th grade) and high school in the state of São Paulo. To analyze these answers, we used software Alceste, and, in addition, the content analysis. The relationship between school and community is highlighted in the literature as complex, nonlinear and marked by tensions and contradictions. The results from of the experiments conducted by the schools, found that more than half of the projects was related to the external community. The projects analyzed were related to the external community because of the problems, needs or values experienced by students and their families or for the account of the specificities and needs of the neighborhood, as well as the need to involve parents and projects. http://dx.doi.org/10.14572/nuances.v26i0.2796En este trabajo se discute la posible relación entre los proyectos de educación en valores desarrollados en las escuelas públicas y sus comunidades aledañas. Para el análisis de los cuestionarios utilizado el Alceste y, además, el análisis de contenido. Las relaciones se observaron en la literatura tan complejo, no lineal y marcadas por tensiones y contradicciones . En el análisis de los experimentos llevados a cabo por las escuelas, se encontró que más de la mitad de los proyectos se relaciona con la comunidad externa. Nas análises das experiências desenvolvidas pelas escolas, verificamos que mais da metade dos projetos relacionou-se com a comunidade externa.
