Digital and algorithmic technology: the impact on employment and the workforce
L’obiettivo di questo articolo è rispondere alle domande inerenti al futuro
del lavoro dal punto di vista della trasformazione tecnologica, con l’uso della
robotica, dell’intelligenza algoritmica, dei social media e dell’evoluzione
dei modelli di business. Lo studio parte da un’analisi della metà degli anni
2000, durante una seconda rivoluzione informatica, in cui l’affermazione
dell’economia digitale è stata osservata con l’intensificazione del processo di
outsourcing e la contingenza dei modelli contemporanei di organizzazione del
lavoro e della produzione. Si parte dall’analisi del lavoro mediante piattaforma,
con la premessa che si tratta di un fenomeno distinto da quello dell’Industria
Digitale 4.0 perché riguarda la realizzazione di intensi processi di produzione
di beni e servizi che vengono completamente esternalizzati da qualsiasi luogo
produttivo tradizionale. Così, l’articolo mira ad analizzare questa nuova
evoluzione della produzione nell’ultima era digitale algoritmica, studiandone il
funzionamento e l’impatto sul lavoro, i salari e la società . La prospettiva teorica

Ancient DNA: genomic amplification of Roman and medieval bovine bones.
Cattle remains (bones and teeth) of both roman and medieval age were collected in the
archaeological site of Ferento (Viterbo, Italy) with the aim of extracting and characterising nucleic acids. Procedures to minimize contamination with modern DNA and to help ancient DNA (aDNA) preservation of the archaeological remains were adopted. Different techniques to extract aDNA
(like Phenol/chloroform extraction) from bovine bones were tested to identify the method that applies to the peculiar characteristics of the study site. Currently, aDNA investigation is mainly based on mtDNA, due to the ease of amplification of the small and high-copied genome and to its usefulness in evolutionary studies. Preliminary amplification of both mitochondrial and nuclear aDNA fragments from samples of Roman and medieval animals were performed and partial specific sequences of mitochondrial D-loop as well as of nuclear genes were obtained. Obiettivo primario di questo scritto è colmare il gap informativo relativo alle caratteristiche socio-anagrafiche di quanti lavorano per le piattaforme digi- tali di lavoro in Italia. A tal fine sono utilizzati i dati provenienti dall’edizione 2018 dell’indagine Inapp PLUS (Participation Labour Unemployment Survey). Oltre ad essere ricostruita ai fini comparativi la risposta regolativa (o giurisprudenziale) adottata in altri Paesi europei per disciplinare il fenomeno, è commentato il cosiddetto Decreto Rider – che ha introdotto una specifica disciplina di tutela per i ciclo-fattorini in Italia. Dalle evidenze empiriche emergono dei bisogni ricollegabili allo svolgimento del lavoro digitale, cui solo in parte la legislazione approvata ha saputo/potuto rispondere

GIV/Girdin is a central hub for profibrogenic signalling networks during liver fibrosis.
Progressive liver fibrosis is characterized by the deposition of collagen by activated hepatic stellate cells (HSCs). Activation of HSCs is a multiple receptor-driven process in which profibrotic signals are enhanced and antifibrotic pathways are suppressed. Here we report the discovery of a signalling platform comprising G protein subunit, Gαi and GIV, its guanine exchange factor (GEF), which serves as a central hub within the fibrogenic signalling network initiated by diverse classes of receptors. GIV is expressed in the liver after fibrogenic injury and is required for HSC activation. Once expressed, GIV enhances the profibrotic (PI3K-Akt-FoxO1 and TGFβ-SMAD) and inhibits the antifibrotic (cAMP-PKA-pCREB) pathways to skew the signalling network in favour of fibrosis, all via activation of Gαi. We also provide evidence that GIV may serve as a biomarker for progression of fibrosis after liver injury and a therapeutic target for arresting and/or reversing HSC activation during liver fibrosis
Endoplasmic Reticulum stress induces hepatic stellate cell apoptosis and contributes to fibrosis resolution.
BACKGROUND:
Survival of hepatic stellate cells (HSCs) is a hallmark of liver fibrosis, while the induction of HSC apoptosis may induce recovery. Activated HSC are resistant to many pro-apoptotic stimuli. To this issue, the role of Endoplasmic Reticulum (ER) stress in promoting apoptosis of HSCs and consequently fibrosis resolution is still debated.
AIM:
To evaluate the potential ER stress-mediated apoptosis of HSCs and fibrosis resolution
METHODS:
HSCs were incubated with the ER stress agonists, tunicamycin or thapsigargin. In vivo, HSC were isolated from normal, bile duct-ligated (BDL) and bile duct-diverted (BDD) rats.
RESULTS:
In activated HSC, the specific inhibitor of ER stress-induced apoptosis, calpastatin, is significantly increased vs. quiescent HSCs. Calpain is conversely reduced in activated HSCs. This pattern of protein expression provides HSCs resistance to the ER stress signals of apoptosis (apoptosis-resistant phenotype). However, both tunicamycin and thapsigargin are able to induce apoptosis in HSCs in vitro, completely reversing the calpain/calpastatin pattern expression. Furthermore, in vivo, the fibrosis resolution observed in rat livers subjected to bile duct ligation (BDL) and subsequent bile duct diversion (BDD), leads to fibrosis resolution through a mechanism of HSCs apoptosis, potentially associated with ER stress: in fact, BDD rat liver shows an increased number of apoptotic HSCs associated with reduced calapstatin and increased calpain protein expression, leading to an apoptosis-sensible phenotype.
CONCLUSIONS:
ER stress sensitizes HSC to apoptosis both in vitro and in vivo. Thus, ER stress represents a key target to trigger cell death in activated HSC and promotes fibrosis resolution
