    skeletal muscle expression analysis of fat metabolism genes in pig

    Fat content and backfat thickness are important polygenic parameters influencing meat quality and carcass traits in pigs. Up to now, there is a lack of knowledge on the expression level of the genes encoding for enzymes involved in fatty acid metabolism in porcine skeletal muscle. In the present study we analysed, by quantitative real time PCR, the expression of three genes, acetyl-CoA carboxylase (ACACA), ATP citrate lyase (ACL) and fatty acid synthase (FASN) in skeletal muscle tissue samples of Italian Large White and Italian Duroc pigs with divergent breeding values for backfat thickness or visible intramuscular fat. Significant differences of the expression level for ACACA gene (P=0.04) and for ACL gene (P=0.02) were observed between the two breeds, comparing the samples selected for backfat thickness trait. The expression analysis of FASN gene in the samples with different genotype at the SNP c.265C>T showed that the TT genotype presented the lowest values in both breeds. The differences observed between breeds should be further considered to investigate the putative involvement of these genes on fat deposition traits. - Ricerca e analisi di espressione di geni candidati per la deposizione di grasso nella carne suina in razze con diversa attitudine alla deposizione di grasso intramuscolare

    Satellite networking integration in the 5G ecosystem: Research trends and open challenges

    The envisioned 5G ecosystem will be composed of heterogeneous networks based on different technologies and communication means, including satellite communication networks. The latter can help increase the capabilities of terrestrial networks, especially in terms of higher coverage, reliability, and availability, contributing to the achievement of some of the 5G KPIs. However, technological changes are not immediate. Many current satellite communication networks are based on proprietary hardware, which hinders the integration with future 5G terrestrial networks as well as the adoption of new protocols and algorithms. On the other hand, the two main paradigms that are emerging in the networking scenario \u2014 software defined networking (SDN) and network functions virtualization \u2014 can change this perspective. In this respect, this article presents first an overview of the main research works in the field of SDN satellite networks in order to understand the already proposed solutions. Then some open challenges are described in light of the network slicing concept by 5G virtualization, along with a possible roadmap including different network virtualization levels. The remaining unsolved problems are related to the development and deployment of a complete integration of satellite components in the 5G ecosystem

    Design and construction of a new metering hot box for the in situ hygrothermal measurement in dynamic conditions of historic masonries

    The main purpose of the HeLLo project is to contribute to data available on the literature on the real hygrothermal behavior of historic walls and the suitability of insulation technologies. Furthermore, it also aims at minimizing the energy simulation errors at the design phase and at improving their conservation features. In this framework, one of the preliminary activities of the study is the creation of a real in situ hot box to measure and analyze different insulation technologies applied to a real historic wall, to quantify the hygrothermal performance of a masonry building. Inside this box, 'traditional' experiments can be carried out: recording heat flux, surface temperature, and air temperatures, as well as relative humidity values through the use of a new sensing system (composed of thermocouples and temperature/relative humidity combined sensors). Within this paper, the process of development, construction, and validation of this new metering box is exhibited. The new hot box, specifically studied for historic case studies, when compared to other boxes, presents other advantages compared to previous examples, widely exemplified

    Analysis of melanocortin 1 receptor (MC1R) gene polymorphisms in some cattle breeds: their usefulness and application for breed traceability and authentication of Parmigiano Reggiano cheese

Il maggior prezzo che questi prodotti spuntano sul mercato fa emergere l\u2019esigenza di poter autenticare o tracciare i prodotti mono-razza per smascherare e scoraggiare possibili frodi. A questo scopo sono stati proposti sistemi di analisi del DNA, alcuni dei quali utilizzano marcatori in geni che determinano il colore del mantello, che \ue8 uno dei principali caratteri che differenziano tra di loro le razze. Diverse mutazioni nel gene melanocortin 1 receptor (MC1R) sono gi\ue0 state associate a particolari effetti sul colore del mantello nella specie bovina. In questa ricerca abbiamo studiato la presenza dei principali alleli al locus MC1R, per valutare la possibilit\ue0 di utilizzare questo gene per l\u2019autenticazione e la tracciabilit\ue0 di razza dei prodotti lattiero-caseari. Le mutazioni che permettono di distinguere questi alleli sono state analizzate utilizzando protocolli di PCR-RFLP e PCR-APLP su un totale di 1360 animali appartenenti a 18 razze bovine. Per ognuna delle seguenti razze, Frisona Italiana, Bruna Italiana, Pezzata Rossa Italiana, Jersey, Rendena, Reggiana e Modenese, \ue8 stato possibile analizzare pi\uf9 di 70 animali. L\u2019allele Ed \ue8 stato identificato nella razza Frisona Italiana con una frequenza dello 0,886. L\u2019allele E (nomenclatura che include tutti gli alleli tranne che e, Ed e E1) \ue8 stato identificato con alta frequenza nella Bruna Italiana (0,591), Rendena (0,738), Jersey (0,955) e Modenese (0,961) e con bassa frequenza nella Pezzata Rossa Italiana (0,029). Inoltre, questo allele \ue8 stato osservato nella Rossa Svedese, Rossa Danese, Grigio Alpina, Piemontese, Romagnola, Marchigiana e Chianina. In alcune di queste razze (Bruna Italiana, Rendena, Grigio Alpina, Piemontese, Rossa Svedese e Rossa Danese) \ue8 stato identificato anche l\u2019allele E1. L\u2019allele e \ue8 risultato fissato nella razza Reggiana e quasi fissato nella razza Pezzata Rossa Italiana. Inoltre, con bassa frequenza, \ue8 stato identificato in tutte le altre razze analizzate, tranne che nella Marchigiana. Le differenze osservate tra razze esaminate indicano che, almeno in alcuni casi, \ue8 possibile utilizzare i polimorfismi del gene MC1R per escludere o confermare l\u2019impiego di latte di una determinata razza nella produzione di un prodotto lattiero-caseario. Il caso pi\uf9 interessante \ue8 quello del formaggio Parmigiano Reggiano prodotto con l\u2019uso esclusivo di latte di bovine di razza Reggiana. Infatti, essendo presente in questa razza soltanto l\u2019allele e il rilievo analitico di qualsiasi altro allele nel DNA estratto dal formaggio rivela l\u2019uso di latte proveniente da altre razze. La messa a punto di un metodo PCR-RFLP per l\u2019analisi del DNA estratto da prodotti lattiero caseari, incluso il Parmigiano Reggiano di oltre 24 mesi di stagionatura, rappresenta uno strumento importante per la difesa di questo prodotto mono-razza da eventuali frodi. I risultati ottenuti su 10 forme di formaggio prodotto esclusivamente con latte di bovine di razza Reggiana e su 15 forme di Parmigiano Reggiano commerciale ottenuto senza restrizione della razza di origine del latte hanno mostrato la validit\ue0 del metodo del quale \ue8 stata valutata anche la sensibilit\ue0n cattle, the MC1R gene has been the subject of several studies with the aim to elucidate the biology of coat colour. Then, polymorphisms of this gene have been proposed as tools for breed identification and animal products authentication. As a first step to identify breed specific DNA markers that can be used for the traceability of mono-breed dairy cattle products we investigated, using PCR-RFLP and PCR-APLP protocols, the presence and distribution of some alleles at the MC1R locus in 18 cattle breeds for a total of 1360 animals. For each of seven breeds (Italian Holstein, Italian Brown, Italian Simmental, Rendena, Jersey, Reggiana and Modenese) a large number of animals (>70) was genotyped so the obtained results can be considered with more confidence. Allele Ed was identified only in black pied cattle (Italian Holstein and Black Pied Valdostana). Allele E (this nomenclature includes all alleles except Ed, E1 and e) was observed in Italian Brown, Rendena, Jersey, Modenese, Italian Simmental, Grigio Alpina, Piedmontese, Chianina, Romagnola, Marchigiana, Swedish Red and White and Danish Red. Allele E1 was identified in Italian Brown, Rendena, Grigio Alpina, Piedmontese, Swedish Red and White and Danish Red. The recessive allele e, known to cause red coat colour, was fixed in Reggiana and almost fixed in Italian Simmental. This allele was observed also in Italian Holstein, Italian Brown, Rendena, Jersey and Modenese albeit with low frequency. Moreover, this allele was detected in Valdostana, Pezzata Rossa d\u2019Oropa, Piedmontese, Romagnola, Swedish Red and White, Danish Red, Charoleis and Salers. In the case of the Reggiana breed, which is fixed for allele e, the MC1R locus is highly informative with respect to breeds that carry other alleles or in which allele e is at very low frequency. In theory, using the MC1R locus it is possible to identify the presence of milk from some other breeds in Parmigiano Reggiano cheese labelled as exclusively from the Reggiana breed. This possibility was practically tested by setting up protocols to extract and analyse polymorphisms of the MC1R locus in several dairy products, including Parmigiano Reggiano cheese cured for 30 months. The lower detection limit was estimated to be 5% of non expected DNA. This test can represent a first deterrent against fraud and an important tool for the valorisation and authentication of Parmigiano Reggiano cheese obtained from only Reggiana milk

    Fine-scale spatial genetic structure and dispersal among Italian smooth newt populations in a rural landscape

    Amphibians are particularly sensitive to habitat loss and fragmentation caused by the intensification and modernization of farming occurring in the second half of the twentieth century in the Mediterranean basin. However, artificial water bodies, associated with traditional husbandry, proved to be important surrogate for amphibian feeding and reproduction. Here, multilocus genotypes were used to investigate the spatial population structure of Lissotriton vulgaris meridionalis and the role of drinking troughs in supporting viable breeding populations within a rural landscape interested by traditional husbandry and agriculture. Our genetic analysis highlighted the conservation value and the potential stepping-stone function of artificial aquatic sites in the dispersal of the species and for the gene flow maintenance. Indeed, populations of drinking troughs show allelic richness and heterozygosity levels comparable to those from natural ponds and there is no great evidence of genetic bottlenecks. A complex system of artificial aquatic sites and few natural wetlands was identified sustaining a well-structured network of demes highly interconnected with themselves and natural aquatic sites. The conservation of the identified genetic clusters may be useful to prevent further population declines and future loss of genetic diversity within the study area characterized by scarce natural wetlands that frequently dried because of agricultural practices and strong seasonality. Site-specific protection measures are needed to contrast the progressive disappearance of drinking troughs observed in the last years in Italy because of the abandonment of traditional farming practices in favour of modern agriculture and intensive farming

    Atomic force microscopy techniques for nanomechanical characterization : a polymer case study

    Atomic force microscopy (AFM) is a versatile tool to perform mechanical characterization of surface samples at the nanoscale. In this work, we review two of such methods, namely contact resonance AFM (CR-AFM) and torsional harmonics AFM (TH-AFM). First, such techniques are illustrated and their applicability on materials with elastic moduli in different ranges are discussed, together with their main advantages and limitations. Then, a case study is presented in which we report the mechanical characterization using both CR-AFM and TH-AFM of polyaniline and polyaniniline doped with nanodiamond particles tablets prepared by a pressing process. We determined the indentation modulus values of their surfaces, which were found in fairly good agreement, thus demonstrating the accuracy of the techniques. Finally, the determined surface elastic moduli have been compared with the bulk ones measured through standard indentation testing. INTRODUCTION In the field of nanotechnology, the development of innovative and nondestructive characterization techniques plays a crucial role. Indeed, the characterization of nanostructured hybrid materials (e.g., thin films and nanocomposites) and devices requires the capability of acquiring maps of the local mechanical properties at the nanoscale. Nanoindentation is the most common method for determining the mechanical properties of thin films. However, its applicability is strictly limited by the thickness of the sample. Furthermore, its poor spatial resolution does not allow the reconstruction of an accurate distribution of the sample surface mechanical properties. For this reason, alternative methods, based on atomic force microscopy (AFM), have been developed. By exploiting the high resolution of the AFM, maps of the surface mechanical properties (i.e., indentation modulus) can be achieved.     Applied Research of the hygrothermal behaviour of an internally insulated historicwall without vapour barrier: In situ measurements and dynamic simulations

    The hygrothermal behaviour of an internally insulated historic wall is still hard to predict, mainly because the physical characteristics of the materials composing the historic wall are unknown. In this study, the hygrothermal assessment of an internally thermal insulated masonry wall of an historic palace located in Ferrara, in Italy, is shown. In situ non-destructive monitoring method is combined with a hygrothermal simulation tool, aiming to better analyse and discuss future refurbishment scenarios. In this context, the original U-value of the wall (not refurbished) is decreased from 1.44W/m2K to 0.26W/m2K (10 cm stone wool). Under the site specific conditions of this wall, not reached by the sun or rain, it was verified that even in the absence of vapour barrier, no frost damage is likely to occur and the condensation risk is very limited. Authors proposed further discussion based on simulation. The results showed that the introduction of a second gypsum board to the studied technology compensated such absence, while the reduction of the insulation material thickness provides a reduction of RH peaks in the interstitial area by 1%; this second solution proved to be more efficient, providing a 3% RH reduction and the avoidance of further thermal losses

    Reproductive payoffs of territoriality are snow-dependent in a mountain ungulate, the Alpine chamois

    Female density and distribution are dependent on resource phenology and female availability strongly influences male mating behaviour and success. When a male adopts a ‘resource defence’ tactic, his reproductive success depends on the location and attractiveness of his territory. Environmental factors associated with territory quality are expected to influence mating success, for example, through territory features or male–male competition. In a protected population of a mountain-dwelling polygynous herbivore, the Alpine chamois Rupicapra r. rupicapra, we investigated the relationships among mating opportunities, some environmental variables (snow depth, topographic features and size of territories) and male intra-sexual competition for mating. We recorded the mating behaviour and territory size of 15 GPS-GSM radio-tagged territorial males, during five rutting seasons (early November to early December: N = 8 individuals in 2011, N = 9 in 2012, N = 8 in 2015, N = 11 in 2016, N = 7 in 2017; 80% of them were observed for more than one mating season) and related them to snow depth and topography of territories. In ruts with deep snow cover, territorial males had smaller territories and higher number of mating opportunities than in ruts with lower snow cover. Smaller territories showed the highest values of terrain roughness, in turn with little or no snow cover in the mating season, and were visited by a greater number of females, than larger territories. Number of wins was positively influenced by snow depth and negatively related to the frequency of aggressions. The frequency of male–male aggressive interactions was greater during ruts with deep snow cover and for males with territories at higher elevations; additionally, it was negatively related to interactions won. Thus, snow depth, which influences resource distribution and female movements, is confirmed as a strong determinant of male mating opportunities and mating behaviour

    The knowledge of genes and molecular processes controlling stress reactions and involved in the genetic system determining resistance to stress in pigs could be important for the improvement of meat quality. This research aimed to compare the expression profiles of skeletal muscle between physically stressed and not stressed pigs of different breeds immediately before slaughter. DNA microarray analysis showed that different functional categories of genes are up-regulated in stressed compared to not stressed pigs and relevant differences among breeds were found. - Utilizzando la tecnica microarray vengono confrontate liste di geni differenzialmente espressi in suini di diverse razze sottoposti oppure no a trattamenti stressanti pre macellazione per identificare le basi genetiche della resistenza e suscettibilità allo stress nel suino nella fase premacellazione e vengono analizzati gli effetti dello stress sulla qualità della carne nei soggetti stressati e non stressati.     Bovine chromosome 20: milk production QTL and candidate gene analysis in the Italian Holstein-Friesian breed

    Bovine chromosome 20 (BTA20) was studied to identify QTL for milk yield and protein percentage in the Italian Holstein-Friesian breed using a selective milk DNA pooling strategy in a daughter design with sire haplotype analysis. Several QTL were identified. The effect of the known GHR F279Y and PRLR S18N mutations were in for the most part confirmed. However, it was also shown that these markers cannot explain all significant effects observed on BTA20 for the investigated traits
