    Abstract IL-2, the first cytokine discovered with T cell growth factor activity, is now known to have pleiotropic effects on T cells. For example, it can promote growth, survival, and differentiation of Ag-selected cells, or facilitate Ag-induced cell death of T cells when Ag persists, and in vivo, it is thought to contribute to the regulation of the size of adaptive T cell response. IL-2 is deficient in HIV-1 infection and has been used in the management of HIV-1-infected individuals undergoing antiretroviral therapy. In this study, we investigated how continuous low-dose IL-2 affected the CD4+ and CD8+ T cell response induced by two inoculations of a canarypox recombinant SIV-based vaccine candidate in healthy macaques chronically infected with SIVmac251. These macaques had normal levels of CD4+ T cells at the beginning of antiretroviral therapy treatment. Vaccination in the presence of IL-2 significantly augmented Gag-specific CD8+ T cell responses, but actually reduced Gag-specific CD4+ T cell responses. Although IL-2 at low doses did not change the overall concentration of circulating CD4+ or CD8+ T cells, it expanded the frequency of CD4+CD25+ T cells. Depletion of the CD4+CD25+ T cells in vitro, however, did not result in a reconstitution of Gag-specific CD4+ responses or augmentation of SIV-specific CD8+ T cell responses. Thus, we conclude that the decrease in virus-specific CD4+ T cell response may be due to IL-2-promoted redistribution of cells from the circulation, or due to Ag-induced cell death, rather than suppression by a T regulatory population

    Riluzole use in presence of contraindications in adults affected by amyotrophic lateral sclerosis and its off-label use in other motor neuron diseases: Findings from an Italian multicentre study (the {CAESAR} project)

    Background: This analysis describes the use of riluzole in amyotrophic lateral sclerosis (ALS) individuals with contraindications and off-label use for subjects with other motor neuron diseases (o-MND) in the Italian regions of Latium, Tuscany and Umbria.Methods: A cohort of adults with ALS prescribed with riluzole during the years 2016–2019 was enrolled from administrative healthcare databases, excluding subjects with o-MND in the preceding 5 years. Being affected by ALS for more than 5 years, presence of tracheostomy, renal or hepatic failure were considered as contraindications to the use of riluzole. A cohort of adults with o-MND was enrolled in 2016–2019 for whom off-label use of riluzole was retrieved up to 4 years, analysing over the time differences related to sex.Results: Among 206 ALS individuals prescribed with riluzole in Latium, 336 in Tuscany and 60 in Umbria, less than 1% were diagnosed with ALS for more than 5 years. Less than 2% were tracheotomised or affected by hepatic failure. Renal failure was documented for 1.9%, 2.7%, and 5.0% of ALS individuals in Latium, Tuscany and Umbria. The o-MND cohort comprised 264 subjects in Latium, 222 in Tuscany, and 66 in Umbria. Non-negligible off-label riluzole use was observed: 8.5%, 33.0%, and 4.2% in females, and 19.9%, 26.5% and 2.4% in males in Latium, Tuscany and Umbria.Discussion: Riluzole use in ALS individuals in the presence of contraindications is rare, with slightly higher numbers in presence of renal failure. Off-label use in o-MND was found to be non-negligible, with variations between sexes

    ChronoMoMoWo is a timeline conceived for the International Travelling Exhibition of the European project "MoMoWo – Women’s creativity since the Modern Movement". This timeline has been devised as a framework that helps to understand the activity carried out between 1918 and 2018 by European female architects, designers and civil engineers. ChronoMoMoWo mirrors the growing of visibility of women's professional creativity and provides a continuum of personal achievements that have contributed to breaking the glass ceiling that is often found in architecture and design. ChronoMoMoWo traces two lines that are related from a historical point of view. The lower area shows a summary of the main achievements of European women in the fields of politics, society, culture, science, education and sports. The upper area presents the most relevant milestones by female architects and designers, including the access of women to higher education, the awards received, and the creation of female professional associations or the most important works by several cohorts of female designers. This timeline represents a historical context that evolves from black and white pictures to colour photographs, portraying a group of capable and talented women that have been –and still are– able to compete with their male counterparts in their professional careers

    Is Oxygen Uptake Measurement Enough to Estimate Energy Expenditure During High-Intensity Intermittent Exercise? Quantification of Anaerobic Contribution by Different Methods

    Purpose: The aim of the present study was to compare the contributions of the anaerobic pathway as determined by two different methods and energy expenditure during a typical high-intensity intermittent exercise (HIIE) protocol.Methods: A descriptive research design was utilized in which thirteen physically active men performed six experimental sessions consisting of an incremental test (session 1), submaximal tests at 40, 50, 60, 70, 75, 80, 85, 90% of velocity associated with maximum oxygen uptake (vV˙O2max) with two intensities per session (sessions 2–5), and the HIIE protocol (session 6; 10 efforts of 1 min at vV˙O2max interspersed by 1 min of passive recovery). The estimation of anaerobic energy system contribution was calculated by: (a) the excess post-exercise oxygen consumption plus delta lactate method and (b) the accumulated oxygen deficit method using the difference between predicted oxygen demand from the submaximal tests of varying intensities and accumulated oxygen uptake during HIIE. Estimation of aerobic energy system contribution was calculated through the measurement of oxygen consumption during activity. Total EE during the entire HIIE protocol (efforts + recovery) and for the efforts only were calculated from each method.Results: For efforts + recovery and efforts only, anaerobic contribution was similar for both methods, and consequently total EE was also equivalent (p = 0.230 for both comparisons). During efforts + recovery, aerobic:anaerobic energy system contribution was (68 ± 4%: 32 ± 4%), while efforts only was (54 ± 5%: 46 ± 5%) with both situations demonstrating greater aerobic than anaerobic contribution (p < 0.001 for both).Conclusion: Anaerobic contribution seems to be relevant during HIIE and must to be taken into account during total EE estimation; however, the type of method employed did not change the anaerobic contribution or total EE estimates

    First evidence of cases of haemophilia dates from ancient Egypt, but it was when Queen Victoria from England in the 19th century transmitted this illness to her descendants, when it became known as the "royal disease". Last decades of the 20th century account for major discoveries that improved the life expectancy and quality of life of these patients. The history and evolution of haemophilia healthcare counts ups and downs. The introduction of prophylactic schemes during the 1970s have proved to be more effective that the classic on-demand replacement of clotting factors, nevertheless many patients managed with frequent plasma transfusions or derived products became infected with the Human Immunodeficiency Virus (HIV) and Hepatitis C virus during the 1980s and 1990s. Recombinant factor VIII inception has decreased the risk of blood borne infections and restored back longer life expectancies. Main concerns for haemophilia healthcare are shifting from the pure clinical aspects to the economic considerations of long-term replacement therapy. Nowadays researchers' attention has been placed on the future costs and cost-effectiveness of costly long-term treatment. Equity considerations are relevant as well, and alternative options for less affluent countries are under the scope of further research. The aim of this review was to assess the evidence of different treatment options for haemophilia type A over the past four decades, focusing on the most important technological advances that have influenced the natural course of this "royal disease"
