    Principles of protein structure: An established Internet‐based course in structural biology

    The Department of Crystallography at Birkbeck College, London, UK, has been running a one‐year, part‐time accredited graduate course, ‘Principles of Protein Structure’, entirely over the Internet since 1996. Students on this course learn the basic principles of the increasingly important subject of structural biology using software programs such as Rasmol and Chime to visualize and manipulate molecular structures in three dimensions. They interact with their tutors, based at Birkbeck, using email and text‐based teleconferencing, and can test their knowledge with multiple choice quizzes on the Web. Over 200 students from thirty countries registered for this course in the last four years. Forty, from central and eastern Europe, were supported by bursaries from the Open Society Institute. The course has been well received by students and its success led us to introduce a similar course in protein crystallography

    Bargaining and Trust: The Effects of 36hr Total Sleep Deprivation on Socially Interactive Decisions

    Though it is well known that sleep loss results in poor judgment and decisions, little is known about the influence of social context in these processes. Sixteen healthy young adults underwent three games involving bargaining (‘Ultimatum’ and ‘Dictator’) and trust, following total sleep deprivation (TSD) and during rested wakefulness (RW), in a repeated measures, counterbalanced design. To control for repeatability, a second group (n=16) was tested twice under RW conditions. Paired anonymously with another individual, participants made their simple social interaction decisions facing real monetary incentives. For bargaining, following TSD participants were more likely to reject unequal-split offers made by their partner, despite the rejection resulting in a zero monetary payoff for both participants. For the trust game, participants were less likely to place full trust in their anonymous partner, again affecting final payoff. Overall, we provide novel evidence that following TSD, the conflict between personal financial gain and payoff equality is focused on unfavourable inequality. This results in the rejection of unfair offers, at personal monetary cost, and the lack of full trust, which would expose one to being exploited in the interaction. As such, we suggest that within a social domain a rational decision may not prevail over more emotional options following TSD, which has fundamental consequence for real-world decision making involving social exchange.     Overview This report analyses serious, transnational and organised crime and the harms it causes to Australia’s interest, with the aim of reinvigorating a discussion of this critical matter amongst Australians. This web impacts on our national interests to the sum of an estimated $15 billion per year. That very conservative estimate includes costs to government through denied revenue and increased law enforcement costs. But there are also social, health and economic harms to individuals, community and business. The report poses a series of questions to be considered by the community, business and government

    Science vs. science: the complexities of interdisciplinary research

    Human-Computer Interaction and Web Science are radically interdisciplinary fields, but what does this mean in practical terms? Undertaking research (and writing papers) that encompass multiple disciplinary perspectives and methods is a serious challenge and it is difficult to maintain conferences that fairly review and host contributions from multiple disciplines. The colocation of the ACM WebSci conference with CHI in Paris, offers an unusual opportunity to bring these two communities together. Previous discussions have considered how to conduct interdisciplinary work that bridges HCI/WebSci with specific areas. Our objective is to provide a space for interested researchers from both communities to share their views and approaches to tackling the tensions and complexities associated with interdisciplinary work, whatever fields are being bridged

    Phosphorylation of Subunit Proteins of Intermediate Filaments from Chicken Muscle and Nonmuscle Cells

    The phosphorylation of the subunit proteins of intermediate (10-nm) filaments has been investigated in chicken muscle and nonmuscle cells by using a two-dimensional gel electrophoresis system. Desmin, the 50,000-dalton subunit protein of the intermediate filaments of muscle, had previously been shown to exist as two major isoelectric variants--alpha and ß --in smooth, skeletal, and cardiac chicken muscle. Incubation of skeletal and smooth muscle tissue with 32PO4{}3- reveals that the acidic variant, alpha -desmin, and three other desmin variants are phosphorylated in vivo and in vitro. Under the same conditions, minor components of alpha - and ß -tropomyosin from skeletal muscle, but not smooth muscle, are also phosphorylated. Both the phosphorylated desmin variants and the nonphosphorylated ß -desmin variant remain insoluble under conditions that solubilize actin and myosin filaments, but leave Z-discs and intermediate filaments insoluble. Primary cultures of embryonic chicken muscle labeled with 32PO4{}3- possess, in addition to the desmin variants described above, a major nonphosphorylated and multiple phosphorylated variants of the 52,000-dalton, fibroblast-type intermediate filament protein (IFP). Filamentous cytoskeletons, prepared from primary myogenic cultures by Triton X-100 extraction, contain actin and all of the phosphorylated and nonphosphorylated variants of both desmin and the IFP. Similarly, these proteins are the major components of the caps of aggregated 10-nm filaments isolated from the same cell cultures previously exposed to Colcemid. These results demonstrate that a nonphosphorylated and several phosphorylated variants of desmin and IFP are present in assembled structures in muscle and nonmuscle cells

    Convicts and coolies : rethinking indentured labour in the nineteenth century

    This article seeks to shift the frame of analysis within which discussions of Indian indentured migration take place. It argues that colonial discourses and practices of indenture are best understood not with regard to the common historiographical framework of whether it was 'a new system of slavery', but in the context of colonial innovations in incarceration and confinement. The article shows how Indian experiences of and knowledge about transportation overseas to penal settlements informed in important ways both their own understandings and representations of migration and the colonial practices associated with the recruitment of indentured labour. In detailing the connections between two supposedly different labour regimes, it thus brings a further layer of complexity to debates around their supposed distinctions
