    Whole genome sequence analysis reveals the broad distribution of the RtxA type 1 secretion system and four novel putative type 1 secretion systems throughout the Legionella genus.

    Type 1 secretion systems (T1SSs) are broadly distributed among bacteria and translocate effectors with diverse function across the bacterial cell membrane. Legionella pneumophila, the species most commonly associated with Legionellosis, encodes a T1SS at the lssXYZABD locus which is responsible for the secretion of the virulence factor RtxA. Many investigations have failed to detect lssD, the gene encoding the membrane fusion protein of the RtxA T1SS, in non-pneumophila Legionella, which has led to the assumption that this system is a virulence factor exclusively possessed by L. pneumophila. Here we discovered RtxA and its associated T1SS in a novel Legionella taurinensis strain, leading us to question whether this system may be more widespread than previously thought. Through a bioinformatic analysis of publicly available data, we classified and determined the distribution of four T1SSs including the RtxA T1SS and four novel T1SSs among diverse Legionella spp. The ABC transporter of the novel Legionella T1SS Legionella repeat protein secretion system shares structural similarity to those of diverse T1SS families, including the alkaline protease T1SS in Pseudomonas aeruginosa. The Legionella bacteriocin (1-3) secretion systems T1SSs are novel putative bacteriocin transporting T1SSs as their ABC transporters include C-39 peptidase domains in their N-terminal regions, with LB2SS and LB3SS likely constituting a nitrile hydratase leader peptide transport T1SSs. The LB1SS is more closely related to the colicin V T1SS in Escherichia coli. Of 45 Legionella spp. whole genomes examined, 19 (42%) were determined to possess lssB and lssD homologs. Of these 19, only 7 (37%) are known pathogens. There was no difference in the proportions of disease associated and non-disease associated species that possessed the RtxA T1SS (p = 0.4), contrary to the current consensus regarding the RtxA T1SS. These results draw into question the nature of RtxA and its T1SS as a singular virulence factor. Future studies should investigate mechanistic explanations for the association of RtxA with virulence

    Reconfiguring an Irrigation Landscape to Improve Provision of Ecosystem Services

    Over-allocation of fresh water resources to consumptive uses, coupled with recurring drought and the prospect of climate change, is compromising the stocks of natural capital in the world’s basins and reducing their ability to provide ecosystem services. To combat this, governments world wide are making significant investment in efforts to improve sharing of water between consumptive uses and the environment, with many investments centred on modernisation of inefficient irrigation delivery systems, and the purchase of water by government for environmental flows. In this study, spatial targeting was applied within a cost-benefit framework to reconfigure agricultural land use in an irrigation district to achieve a 20% reduction in agricultural water use to increase environmental flows and improve the provision of other ecosystem services. We demonstrate using spatial planning and optimisation models that a targeted land use reconfiguration policy approach could potentially increase the net present value of ecosystem services by up to AUS463.7m.Thisprovidesathresholdlevelofinvestmentthatwouldbejustifiedonthebasisofbenefitsthattheinvestmentproduces.Theincreaseinecosystemservicesincluderecovering61GLofwaterforenvironmentalflows,thesequestrationof10.6mtonnesofCO2e/yr,a13EC(?S/cm)reductioninriversalinity,andanoverall24463.7m. This provides a threshold level of investment that would be justified on the basis of benefits that the investment produces. The increase in ecosystem services include recovering 61 GL of water for environmental flows, the sequestration of 10.6m tonnes of CO2-e/yr, a 13 EC (?S/cm) reduction in river salinity, and an overall 24% increase in the value of agriculture. Without a targeted approach to planning, a 20% reduction in water for irrigation could result in the loss of AUS68.7m in economic returns to agriculture which may be only marginally offset by the increased value of ecosystem services resulting from the return of 61 GL of water to the environment.landscape planning, geographic information systems, cost-benefit analysis, irrigation, climate change, water management, spatial targeting, environmental valuation

    Amish Economic Transformations: New Forms of Income and Wealth Distribution in a Traditionally “Flat” Community

    The basic contours of the Amish economic transformation over the past few decades have been well documented, including the demographic squeeze that pushed many Amish out of farming, their embrace of cottage industries and, to a lesser extent, factory labor, and the social and cultural dilemmas created by successful entrepreneurship. Yet the effects of increasing market entanglement on the distribution of income and assets in Amish communities are still poorly understood. In this exploratory study, we draw on publicly available data from the U.S. Census, the Ohio Amish Directory, and records from real estate transactions to map out the distribution of income and land wealth in one predominantly Amish-populated Census Tract in Holmes County, OH. Our findings illustrate economic differentiation within the Amish community, as well as the ways in which affiliation and church leadership are associated with land holdings. Our case study raises important questions about growing economic inequalities that merit further exploration

    Subclinical infection of macaques and baboons with a baboon simarterivirus

    Simarteriviruses (Arteriviridae: Simarterivirinae) are commonly found at high titers in the blood of African monkeys but do not cause overt disease in these hosts. In contrast, simarteriviruses cause severe disease in Asian macaques upon accidental or experimental transmission. Here, we sought to better understand the host-dependent drivers of simarterivirus pathogenesis by infecting olive baboons (n = 4) and rhesus monkeys (n = 4) with the simarterivirus Southwest baboon virus 1 (SWBV-1). Surprisingly, none of the animals in our study showed signs of disease following SWBV-1 inoculation. Three animals (two rhesus monkeys and one olive baboon) became infected and sustained high levels of SWBV-1 viremia for the duration of the study. The course of SWBV-1 infection was highly predictable: plasma viremia peaked between 1 × 107 and 1 × 108 vRNA copies/mL at 3–10 days post-inoculation, which was followed by a relative nadir and then establishment of a stable set-point between 1 × 106 and 1 × 107 vRNA copies/mL for the remainder of the study (56 days). We characterized cellular and antibody responses to SWBV-1 infection in these animals, demonstrating that macaques and baboons mount similar responses to SWBV-1 infection, yet these responses are ineffective at clearing SWBV-1 infection. SWBV-1 sequencing revealed the accumulation of non-synonymous mutations in a region of the genome that corresponds to an immunodominant epitope in the simarterivirus major envelope glycoprotein GP5, which likely contribute to viral persistence by enabling escape from host antibodies

    Relations Between Stream Chemistry, Fish Diversity, and Land Use in the Upper Little Miami Watershed

    Streams are susceptible to numerous threats to their water quality and biodiversity. In southwest Ohio a major driver of these impacts is associated with current agricultural practices and associated legacy effects. These structural and chemical impacts are known to affect biodiversity in these streams. The objective of this study was to document and examine relationships among land-use/land cover, stream chemistry, and fish diversity in the headwaters streams of the Little Miami Watershed. Three streams (Little Miami River, Massies Creek - North Fork, and Massies Creek - South Fork) were sampled in the upper headwaters as well as downstream. Air and water temperature, pH, dissolved oxygen, alkalinity, hardness and turbidity were measured in early September and late November. With the exception of temperature, the measurement of these parameters was generally consistent between sampling periods (CV \u3c 22). A total of 517 fish were collected using an electroshock backpack along 75m stretches at the sites (N = 6). In total 25 species were identified while individual sites yielded 7 - 12 species each. Fish diversity (Simpson’s Diversity, 1 - D) at our sites ranged between 0.77 and 0.86. A significant correlation (r = 0.918, p = 0.01) was found between fish diversity and the % of developed land in these watersheds. Our landscape analysis revealed that all six watersheds had 86 - 97% agricultural land use which had a negative (r = - 0.796), but not statistically significant correlation (p = 0.0581) impact on fish diversity. This work will serve as the baseline for future study of land use impacts on fish diversity, phylogeography, and water quality

    L’affectation disproportionnée des étudiants de couleur dans l’enseignement spécialisé a été un problème récurrent aux États-Unis et ailleurs. Les auteurs retracent les contextes historiques ainsi que des tendances contemporaines associées à cette disproportion. Ils analysent ensuite les façons dont la surreprésentation s’est appuyée sur les héritages problématiques de la théorie du déficit et des tentatives d’inclusion avortées. Sont ensuite présentés des développements prometteurs dans la théorie, la recherche et la pratique, visant à annuler les effets nocifs du clivage racial dans les écoles et la société. Il faut un engagement continu pour étudier et mieux comprendre la question de la disproportion à travers des champs d’étude interdisciplinaires.The disproportionate placement of students of color in special education has been a persistent problem in the USA and beyond. This article traces the historical contexts, as well as contemporary trends associated with disproportionality. The authors then discuss ways that overrepresentation has relied upon problematic legacies of deficit thinking and frustrated attempts toward inclusion. Next, they document promising developments in theory, research, and practice that seek to undo the harmful effects of racially divided schools and society. Finally, they urge for a continued commitment to researching and better understanding the issue of disproportionality through interdisciplinary fields of study.La afectación desproporcionada de estudiantes de color en la enseñanza especializada ha sido un problema recurrente en Estados Unidos y en otras partes del mundo. Los autores esbozan los contextos históricos así como unas tendencias contemporáneas asociadas a esta desproporción.
