Post-alpine tectonics of the Upper Nysa Kłodzka Graben : a reply
The paper aims to answer the critical remarks presented by Don and Wojewoda (in this issue) related to our papers published in Acta Montana (Badura et al., 2002, Badura et al., 2003). Answering those we have noticed that the terms brachysyncline and brachyanticline are improper with regard to the young Alpine rebuilding of the Upper Nysa Kłodzka Graben. In this paper we present the basic reasons for our hypothesis of young tectonic movements in the studied area more widely. Our
morphotectonic analyses are based on the digital elevation models (DEM) and the electroresistance survey of the researched area located near Kamienna and north of Długopole Zdrój. The results of our research show that despite of the long history of the geological studies - longer than 100 years, the region is still a very challenging research area. In this paper we point out that application of the DEM and electroresistivity methods gives the new important results. Those should definitely be taken into consideration together with palaeontological research and geological mapping. We really wonder that the authors of the critical paper unequivocally negate the results of our studies and suggest that the continuation of such research and geodetic survey in the area is aimless
The influence of cryogenic temperatures on the characteristics of a brushless motor
This article presents the results of a study on the magnetic properties of soft magnetic powder composites at room temperature and at liquid nitrogen temperature. The properties of a magnetoelectric motor with the power of 0.75 kW at room temperature and cryogenic temperatures are also presented in this article. The study of magnetic properties showed that magnetization curves at 20°C and -195.8°C temperatures are almost the same. Iron loss in the composite at liquid nitrogen temperature increased significantly in comparison to loss at room temperature. The study of the magnetoelectric motor showed that the back electromotive force at liquid nitrogen temperature decreases insignificantly in comparison to the back electromotive force at room temperature. Electromagnetic torque at liquid nitrogen temperature increases in comparison to room temperature
ALLPATHS 2: Small Genomes Assembled Accurately and with High Continuity from Short Paired Reads
We demonstrate that genome sequences approaching finished quality can be generated from short paired reads. Using 36 base (fragment) and 26 base (jumping) reads from five microbial genomes of varied GC composition and sizes up to 40 Mb, ALLPATHS2 generated assemblies with long, accurate contigs and scaffolds. Velvet and EULER-SR were less accurate. For example, for Escherichia coli, the fraction of 10-kb stretches that were perfect was 99.8% (ALLPATHS2), 68.7% (Velvet), and 42.1% (EULER-SR).Organismic and Evolutionary Biolog
Cancer immunogenomic approach to neoantigen discovery in a checkpoint blockade responsive murine model of oral cavity squamous cell carcinoma
Head and neck squamous cell carcinomas (HNSCC) are an ideal immunotherapy target due to their high mutation burden and frequent infiltration with lymphocytes. Preclinical models to investigate targeted and combination therapies as well as defining biomarkers to guide treatment represent an important need in the field. Immunogenomics approaches have illuminated the role of mutation-derived tumor neoantigens as potential biomarkers of response to checkpoint blockade as well as representing therapeutic vaccines. Here, we aimed to define a platform for checkpoint and other immunotherapy studies using syngeneic HNSCC cell line models (MOC2 and MOC22), and evaluated the association between mutation burden, predicted neoantigen landscape, infiltrating T cell populations and responsiveness of tumors to anti-PD1 therapy. We defined dramatic hematopoietic cell transcriptomic alterations in the MOC22 anti-PD1 responsive model in both tumor and draining lymph nodes. Using a cancer immunogenomics pipeline and validation with ELISPOT and tetramer analysis, we identified the H-2Kb-restricted ICAM1P315L (mICAM1) as a neoantigen in MOC22. Finally, we demonstrated that mICAM1 vaccination was able to protect against MOC22 tumor development defining mICAM1 as a bona fide neoantigen. Together these data define a pre-clinical HNSCC model system that provides a foundation for future investigations into combination and novel therapeutics
Subcutaneous implantable cardioverter-defibrillator therapy in Poland: Results of the Polish S-ICD Registry
Background: The use of a subcutaneous implantable cardioverter-defibrillator (S-ICD) has been expanding in Poland since 2014. The Polish Registry of S-ICD Implantations was held by the Heart Rhythm Section of the Polish Cardiac Society between May 2020 and September 2022 to monitor the implementation of that therapy in Poland.
Aims: To investigate and present the state-of-the-art of S-ICD implantation in Poland.
Methods: Implanting centers reported clinical data of patients undergoing S-ICD implantations and replacements, including: age, gender, height, weight, underlying disease, history of pacemaker and defibrillator implantations, indications for S-ICD, electrocardiographical parameters, procedural techniques, and complications.
Results: 440 patients undergoing S-ICD implantation (411) or replacement (29) were reported by 16 centers. Most patients were in New York Heart Association class II (218 patients, 53%) or I (150 patients, 36.5%). Left ventricular ejection fraction was 10-80%, median (IQR) 33% (25%‒55%). Primary prevention indications were present in 273 patients (66.4%). Non-ischemic cardiomyopathy was reported in 194 patients (47.2%). The main reason for the choice of S-ICD were: young age (309, 75.2%), risk of infective complications (46, 11.2%), prior infective endocarditis (36, 8.8%), hemodialysis (23, 5.6%), and immunosuppressive therapy (7, 1.7%). Electrocardiographic screening was performed in 90% of patients. The rate of adverse events was low (1.7%). No surgical complications were observed.
Conclusions: Qualification for S-ICD in Poland was slightly different when compared to the rest of Europe. The implantation technique was mostly consistent with the current guidelines. S-ICD implantation was safe, and the complication rate was low
A skeleton of peat-trapped forest rhinoceros Stephanorhinus kirchbergensis (Jäger, 1839) from Gorzów Wielkopolski, Northwestern Poland: a record of life and death of the Eemian large mammals
We present a description of an almost complete skeleton of the forest rhino (Stephanorhinus kirchbergensis) found near Gorzów Wielkopolski (Northwest Poland) in its geological and palaeoenvironmental context. While finds of bones and teeth are common, the importance of this find resides in, that it is the most complete specimen of this species, almost perfectly preserved in a well-studied and dated stratigraphic and sedimentological context, along with other fossil fauna and flora. The OSL dates of glaciofluvial sediments sandwiching skeleton-bearing horizons indicate a Middle Eemian age of the Pleistocene paleolake land. This warm climate is also indicated by the results of sedimentological, geochemical, paleobotanical, and isotopic analyses. The rhino skeleton was deposited in a shallow area near the lake’s littoral zone. Bite marks of a large predator (cave hyena) were recognized on the rhino’s pelvis, which, due to their location, were probably created post-mortem. With an estimated height of 1.82 m at the withers, this was a large adult Stephanorhinus kirchbergensis. The results of osteometric analyses indicate that we are dealing with one of the largest known individuals of this species. Although it suffered from a pathologically deformed spine, the individual died at a relatively old age.Peer reviewe
A new approach to ticagrelor-based de-escalation of antiplatelet therapy after acute coronary syndrome. A rationale for a randomized, double-blind, placebo-controlled, investigator-initiated, multicenter clinical study
© 2021 Via Medica. This article is available in open access under Creative Common Attribution-Non-Commercial-No Derivatives 4.0 International (CC BY-NC-ND 4.0) license. https://creativecommons.org/licenses/by/4.0/The risk of ischemic events gradually decreases after acute coronary syndrome (ACS), reaching a stable level after 1 month, while the risk of bleeding remains steady during the whole period of dual antiplatelet treatment (DAPT). Several de-escalation strategies of antiplatelet treatment aiming to enhance safety of DAPT without depriving it of its efficacy have been evaluated so far. We hypothesized that reduction of the ticagrelor maintenance dose 1 month after ACS and its continuation until 12 months after ACS may improve adherence to antiplatelet treatment due to better tolerability compared with the standard dose of ticagrelor. Moreover, improved safety of treatment and preserved anti-ischemic benefit may also be expected with additional acetylsalicylic acid (ASA) withdrawal. To evaluate these hypotheses, we designed the Evaluating Safety and Efficacy of Two Ticagrelor-based De-escalation Antiplatelet Strategies in Acute Coronary Syndrome — a randomized clinical trial (ELECTRA-SIRIO 2), to assess the influence of ticagrelor dose reduction with or without continuation of ASA versus DAPT with standard dose ticagrelor in reducing clinically relevant bleeding and main-taining anti-ischemic efficacy in ACS patients. The study was designed as a phase III, randomized, multicenter, double-blind, investigator-initiated clinical study with a 12-month follow-up.Peer reviewedFinal Published versio
