    Pomolic acid inhibits proliferation of human lung carcinoma cells via induction of apoptosis and suppression of cell migration and invasion

    Purpose: To investigate the anti-proliferative effect of pomolic acid on lung cancer cells (A549), and theunderlying mechanism.Methods: The viability of pomolic acid-treated A549 cells was determined by MTT and colony formation assays. Cell colony formation was monitored with acridine orange/ethidium bromide (AO/EB) staining. Protein expressions of Bax and Bcl-2 were assayed by western blotting.Results: Pomolic acid suppressed the growth of A549 cells, with an half-maximal inhibitory concentration of (IC50) of 10 μM (p < 0.05). However, the IC50 of pomolic acid for normal BEAS-2B cells was 80 μM. Pomolic acid also decreased colony formation of A549 cells. At 20 μM, the percentage of A549 colonies decreased to 14 % of control. The dose-dependent cytotoxicity of pomolic acid against A549 cells was mediated via induction of apoptosis and oxidative stress. Pomolic acid treatment enhanced the expression of Bax and decreased the expression of Bcl-2 in A549 cells. Moreover, pomolic acid inhibited the migration and invasion in A549 cells in a dose-dependent manner (p < 0.05).Conclusion: These results indicate the potent anticancer effect of pomolic acid against human lung cancer cells. Thus, pomolic acid has promising potential as a lead molecule for the development of chemotherapy

    Urania: Visualizing Data Analysis Pipelines for Natural Language-Based Data Exploration

    Exploratory Data Analysis (EDA) is an essential yet tedious process for examining a new dataset. To facilitate it, natural language interfaces (NLIs) can help people intuitively explore the dataset via data-oriented questions. However, existing NLIs primarily focus on providing accurate answers to questions, with few offering explanations or presentations of the data analysis pipeline used to uncover the answer. Such presentations are crucial for EDA as they enhance the interpretability and reliability of the answer, while also helping users understand the analysis process and derive insights. To fill this gap, we introduce Urania, a natural language interactive system that is able to visualize the data analysis pipelines used to resolve input questions. It integrates a natural language interface that allows users to explore data via questions, and a novel data-aware question decomposition algorithm that resolves each input question into a data analysis pipeline. This pipeline is visualized in the form of a datamation, with animated presentations of analysis operations and their corresponding data changes. Through two quantitative experiments and expert interviews, we demonstrated that our data-aware question decomposition algorithm outperforms the state-of-the-art technique in terms of execution accuracy, and that Urania can help people explore datasets better. In the end, we discuss the observations from the studies and the potential future works

    Methylation levels at IGF2 and GNAS DMRs in infants born to preeclamptic pregnancies

    BACKGROUND: Offspring of pregnancy complicated with preeclampsia are at high risk for hypertension, stroke and possibly obesity. The mechanisms behind the association of intrauterine exposure to preeclampsia and high risk of health problems in the later life remain largely unknown. The aims of the current investigation were to determine the changes in DNA methylation at IGF2 and GNAS DMR in offspring of preeclamptic pregnancy and to explore the possible mechanisms underlying the association between maternal preeclampsia and high risk for health problems in the later life of their offspring. RESULTS: Umbilical cord blood was taken from infants born to women of preeclampsia (n=56), gestational hypertension (n=23) and normal pregnancy (n=81). DNA methylation levels of IGF2 and GNAS DMR were determined by Massarray quantitative methylation analysis. Methylation levels at IGF2 DMR were significantly lower in preeclampsia than normal pregnancy. The average methylation level at IGF2 DMR was significantly correlated with preeclampsia even after birth weight, maternal age, gestational age at delivery and fetal gender were adjusted. The difference in methylation level was not significantly different between mild and severe preeclampsia. The methylation level at GNAS DMR was not significantly correlated with birth weight, maternal age, gestational age at delivery, fetal gender, preeclampsia or gestational hypertension. CONCLUSIONS: We concluded preeclampsia induced a decrease in methylation level at IGF 2 DMR, and this might be among the mechanisms behind the association between intrauterine exposure to preeclampsia and high risk for metabolic diseases in the later life of the infants

    PARP inhibitor maintenance treatment for newly diagnosed ovarian cancer patients: a real-world study from China

    PurposeThis study evaluated the efficacy and safety in a real-world population of epithelial ovarian cancer (EOC) treated with poly (ADP-ribose) polymerase inhibitor (PARPi) as first-line maintenance therapy in the largest gynecologic oncology center in Western China.MethodsThis study included patients newly diagnosed EOC who received PARPi as first-line maintenance therapy in West China Second University Hospital from August 1, 2018 to September 31, 2022. The primary endpoints were progression-free survival (PFS) and safety evaluated by Common Terminology Criteria for Adverse Events Version 5.0(CTCAE 5.0). The secondary endpoints were overall survival (OS) and prognostic factors influencing the PFS of patients in real world.ResultsAmong the eligible 164 patients, 104 patients received olaparib and 60 patients received niraparib. 100 patients (61.0%) had mutations in breast cancer susceptibility gene (BRCA). 87 patients (53.0%) received primary debulking surgery (PDS) while 77 patients (47.0%) received interval debulking surgery (IDS). 94 patients (94/164, 57.3%) achieved R0 and 39 patients (23.8%) achieved R1 after PDS/IDS. 112 (68.3%) achieved complete response (CR) after first-line chemotherapy, while 49 (29.9%) achieved partial response (PR). The median follow-up time was 17.0 months (95% CI 15.6-18.4), and the median PFS has not been reached yet. Multivariate analysis demonstrated that BRCA mutations and CR/PR after platinum-based chemotherapy were independent factors associated with prolonged PFS. Hematologic toxicity was the most common grade≥3 AE. There were no incidence of myelodysplastic syndromes/acute myelogenous leukemia (MDS/AML).ConclusionFocusing on PARPi as first-line maintenance therapy for patients with EOC, this study represented the largest single-center real-world study in China to date. Two independent factors were identified to prolong the PFS of patients: BRCA mutated type and CR/PR after primary treatment, which should be further confirmed with long-term follow-up and large sample sizes

    FENGYUN-4A Advanced Geosynchronous Radiation Imager Layered Precipitable Water Vapor Products’ Comprehensive Evaluation Based on Quality Control System

    A physical retrieval algorithm has been developed for deriving the layered precipitable water vapor (LPWs) product from infrared radiances of the Advanced Geosynchronous Radiation Imager (AGRI) onboard FengYun-4A (FY-4A), the first of the new generation of Chinese geostationary weather satellites (FengYun-4, or FY-4 Series). The FY-4A AGRI LPWs are evaluated with different types of reference datasets based on Quality Control System (QCS), including those from Himawari-8 AHI (Advanced Himawari Imager), MODIS (Moderate Resolution Imaging Spectroradiometer), Radiosonde, ERA5 (European Centre for Medium-Range Weather Forecasts Reanalysis v5), NCEP (National Centers for Environmental Prediction) reanalysis and CMA (China Meteorological Administration) forecast product from global medium range numerical weather prediction (NWP) system. QCS is one of the important components of FY-4A ground segment, which mainly focuses on the satellite products’ evaluation and validation. It is found that the AGRI LPW product has a good agreement with different evaluating sources and the quality is favorable and stable. With the capability of frequent (5-min interval) observations over the East Asia and Western Pacific regions, the AGRI LPW products can be used to investigate the atmospheric temporal and spatial variations in the pre-landfall environment for typhoons. The QCS is a useful tool to monitor, evaluate, and validate the AGRI LPW products

    Coclustering Based Parcellation of Human Brain Cortex Using Diffusion Tensor MRI

    Abstract. The fundamental goal of computational neuroscience is to discover anatomical features that reflect the functional organization of the brain. Investigations of the physical connections between neuronal structures and measurements of brain activity in vivo have given rise to the concepts of anatomical and functional connectivity, which have been useful for our understanding of brain mechanisms and their plasticity. However, at present there is no generally accepted computational framework for the quantitative assessment of cortical connectivity. In this paper, we present accurate analytical and modeling tools that can reveal anatomical connectivity pattern and facilitate the interpretation of high-level knowledge regarding brain functions are strongly demanded. We also present a coclustering algorithm, called Business model based Coclustering Algorithm (BCA), which allows an automated and reproducible assessment of the connectivity pattern between different cortical areas based on Diffusion Tensor Imaging (DTI) data. The proposed BCA algorithm not only partitions the cortical mantel into well-defined clusters, but at the same time maximizes the connection strength between these clusters. Moreover, the BCA algorithm is computationally robust and allows both outlier detection as well as operator-independent determination of the number of clusters. We applied the BCA algorithm to human DTI datasets and show good performance in detecting anatomical connectivity patterns in the human brain.
