During this period, volunteers undertook \u3e2,000 days of activities, the majority of which were educational displays at events such as boat shows and patrols in coastal, marine and estuarine environs.     Blue swimmer crabs (Portunus armatus) (formerly Portunus pelagicus; Lai et al., 2010) in Cockburn Sound, Western Australia, are the basis of an important commercial and recreational fishery close to the Perth metropolitan area with the commercial fishery achieving a peak catch of 362 t in 1997/98 and a recreational catch of about 18 t. Commercial blue swimmer crab catches declined significantly since 2000 due to low stock abundance, resulting in the closure of the fishery in December 2006

    Substance use and Intimate Partner Abuse (IPA):A descriptive model of the pathways between substance use and IPA perpetration for men

    Intimate partner abuse (IPA) is a pervasive public health and human rights issue disproportionately affecting women. There is a complex link between IPA and substance use; substance use can increase both the frequency and severity of IPA. Pathway models have been applied to explore heterogeneous trajectories into other behaviours and to identify areas for intervention. This approach has not previously been applied in the area of substance use and IPA. Inductive thematic analysis of 37 interviews with heterosexual men aged 28-52 who had reported previous IPA perpetration was conducted. Men were recruited from alcohol and drug services across two areas of England. Three groupings of pathways into substance use-related IPA were generated: 1) Rule Breaking Pathway (n = 11); 2) Entrenched Substance Use Pathway (n = 13); and 3) Relationship Insecurity Pathway (n = 13). Across the three groupings of pathways, the men's childhood and early experiences led to different journeys into SU-related IPA (abuse that was associated with intoxication, withdrawal, acquisition and substance use lifestyle). Each pathway presented differently with varying core features, for example core features of generalised violence, mental health or jealousy, and different predisposing background factors, including types and timing of childhood abuse and trauma. Adopting a pathways approach drawing on principles of equifinality and multifinality can improve understanding of heterogeneity in men who perpetrate IPA and use substances and propose treatment/intervention targets

    Iterative focused screening with biological fingerprints identifies selective Asc-1 inhibitors distinct from traditional high throughput screening

    N-methyl-d-aspartate receptors (NMDARs) mediate glutamatergic signaling that is critical to cognitive processes in the central nervous system, and NMDAR hypofunction is thought to contribute to cognitive impairment observed in both schizophrenia and Alzheimer’s disease. One approach to enhance the function of NMDAR is to increase the concentration of an NMDAR coagonist, such as glycine or d-serine, in the synaptic cleft. Inhibition of alanine–serine–cysteine transporter-1 (Asc-1), the primary transporter of d-serine, is attractive because the transporter is localized to neurons in brain regions critical to cognitive function, including the hippocampus and cortical layers III and IV, and is colocalized with d-serine and NMDARs. To identify novel Asc-1 inhibitors, two different screening approaches were performed with whole-cell amino acid uptake in heterologous cells stably expressing human Asc-1: (1) a high-throughput screen (HTS) of 3 M compounds measuring 35S l-cysteine uptake into cells attached to scintillation proximity assay beads in a 1536 well format and (2) an iterative focused screen (IFS) of a 45 000 compound diversity set using a 3H d-serine uptake assay with a liquid scintillation plate reader in a 384 well format. Critically important for both screening approaches was the implementation of counter screens to remove nonspecific inhibitors of radioactive amino acid uptake. Furthermore, a 15 000 compound expansion step incorporating both on- and off-target data into chemical and biological fingerprint-based models for selection of additional hits enabled the identification of novel Asc-1-selective chemical matter from the IFS that was not identified in the full-collection HTS

    Pygmy blue whale movement, distribution and important areas in the Eastern Indian Ocean

    This study was conducted as part of AIMS’ North West Shoals to Shore Research Program (NWSSRP) and was supported by Santos as part of the company’s commitment to better understand Western Australia’s marine environment. Hydrophone pressure data from Ocean Bottom Seismometers (OBS) were provided by the CANPASS project, jointly funded by the National Natural Science Foundation of China (NSFC grants 91955210, 41625016), and the China Academy of Science (CAS program GJHZ1776). Instruments were provided by the Australian National instrument pool ANSIR (http://ansir.org.au/). ANSIR, OBS data was also made data available from the Geoscience Australia and Shell. Data was sourced from Australia’s Integrated Marine Observing System (IMOS).Pygmy blue whales in the South-east Indian Ocean migrate from the southern coast of Australia to Indonesia, with a significant part of their migration route passing through areas subject to oil and gas production. This study aimed at improving our understanding of the spatial extent of the distribution, migration and foraging areas, to better inform impact assessment of anthropogenic activities in these regions. Using a combination of passive acoustic monitoring of the NW Australian coast (46 instruments from 2006 to 2019) and satellite telemetry data (22 tag deployments from 2009 to 2021) we quantified the pygmy blue whale distribution and important areas during their northern and southern migration. We show extensive use of slope habitat off Western Australia and only minimal use of shelf habitat, compared to southern Australia where use of the continental shelf and shelf break predominates. In addition, movement behaviour estimated by a state-space model on satellite tag data showed that in general pygmy blue whales off Western Australia were mostly engaged in migration, interspersed with mostly relatively short periods (median = 28hours, range = 2 – 1080hours) of low move persistence (slow movement with high turning angles), which is indicative of foraging. Using the spatial overlap of time and number of whales in area analysis of the satellite tracking data (top 50% of grid cells) with foraging movement behaviour, we quantified the spatial extent of pygmy blue whale high use areas for foraging and migration. We compared these areas to the previously described areas of importance to foraging and migrating whales (Biologically Important Areas; BIAs). In some cases these had good agreement with the most important areas we calculated from our data, but others had only low (5%) to moderate (13%) overlap. Month was the most important variable predicting the number of pygmy blue whale units and number of singers (acting as indices of pygmy blue whale density). Whale density was highest in the southern part of the NW Australian coast and whales were present there between April-June, and November-December, a pattern also confirmed by the satellite tracking data. Available data indicated pygmy blue whales spent up to 124 days in Indonesian waters (34% of annual cycle). Since this area may also be the calving ground for this population, inter-jurisdictional management is necessary to ensure their full protection.Publisher PDFPeer reviewe

    The type III secretion system effector SeoC of salmonella enterica subsp. salamae and S. enterica subsp. arizonae ADP-Ribosylates Src and inhibits opsonophagocytosis

    Salmonella species utilize type III secretion systems (T3SSs) to translocate effectors into the cytosol of mammalian host cells, subverting cell signaling and facilitating the onset of gastroenteritis. In this study, we compared a draft genome assembly of Salmonella enterica subsp. salamae strain 3588/07 against the genomes of S. enterica subsp. enterica serovar Typhimurium strain LT2 and Salmonella bongori strain 12419. S. enterica subsp. salamae encodes the Salmonella pathogenicity island 1 (SPI-1), SPI-2, and the locus of enterocyte effacement (LEE) T3SSs. Though several key S Typhimurium effector genes are missing (e.g., avrA, sopB, and sseL), S. enterica subsp. salamae invades HeLa cells and contains homologues of S. bongori sboK and sboC, which we named seoC SboC and SeoC are homologues of EspJ from enteropathogenic and enterohemorrhagic Escherichia coli (EPEC and EHEC, respectively), which inhibit Src kinase-dependent phagocytosis by ADP-ribosylation. By screening 73 clinical and environmental Salmonella isolates, we identified EspJ homologues in S. bongori, S. enterica subsp. salamae, and Salmonella enterica subsp. arizonae The β-lactamase TEM-1 reporter system showed that SeoC is translocated by the SPI-1 T3SS. All the Salmonella SeoC/SboC homologues ADP-ribosylate Src E310 in vitro Ectopic expression of SeoC/SboC inhibited phagocytosis of IgG-opsonized beads into Cos-7 cells stably expressing green fluorescent protein (GFP)-FcγRIIa. Concurrently, S. enterica subsp. salamae infection of J774.A1 macrophages inhibited phagocytosis of beads, in a seoC-dependent manner. These results show that S. bongori, S. enterica subsp. salamae, and S. enterica subsp. arizonae share features of the infection strategy of extracellular pathogens EPEC and EHEC and shed light on the complexities of the T3SS effector repertoires of Enterobacteriaceae
