Commentary: Evidence to Guide Gun Violence Prevention in America
Gun violence is a major threat to the public's health and safety in the United States. The articles in this volume's symposium on gun violence reveal the scope of the problem and new trends in mortality rates from gunfire. Leading scholars synthesize research evidence that demonstrates the ability of numerous policies and programs -- each consistent with lessons learned from successful efforts to combat public health problems -- to prevent gun violence. Each approach presents challenges to successful implementation. Future research should inform efforts to assess which approaches are most effective and how to implement evidence-based interventions most effectively
How Cities Can Combat Illegal Guns and Gun Violence
Though gun violence peaked in American cities in the early 1990s, firearms are still used in more than 30 homicides per day in the U.S.One major challenge is how to stanch the flow of guns to criminals. Researchers from the School's Center for Gun Policy and Research have recently zeroed in on illegal sales by licensed dealers (the dominant means of diverting guns into illegal markets). These sales include unrecorded, "off the books" transactions and "straw purchases" in which a person unable to pass a federal background check relies on another individual for the purchase.Daniel Webster, MPH, ScD '91, Center co-director, wants to increase the accountability of gun dealers. High Resolution Nature Runs and the Big Data Challenge
NASA's Global Modeling and Assimilation Office at Goddard Space Flight Center is undertaking a series of very computationally intensive Nature Runs and a downscaled reanalysis. The nature runs use the GEOS-5 as an Atmospheric General Circulation Model (AGCM) while the reanalysis uses the GEOS-5 in Data Assimilation mode. This paper will present computational challenges from three runs, two of which are AGCM and one is downscaled reanalysis using the full DAS. The nature runs will be completed at two surface grid resolutions, 7 and 3 kilometers and 72 vertical levels. The 7 km run spanned 2 years (2005-2006) and produced 4 PB of data while the 3 km run will span one year and generate 4 BP of data. The downscaled reanalysis (MERRA-II Modern-Era Reanalysis for Research and Applications) will cover 15 years and generate 1 PB of data. Our efforts to address the big data challenges of climate science, we are moving toward a notion of Climate Analytics-as-a-Service (CAaaS), a specialization of the concept of business process-as-a-service that is an evolving extension of IaaS, PaaS, and SaaS enabled by cloud computing. In this presentation, we will describe two projects that demonstrate this shift. MERRA Analytic Services (MERRA/AS) is an example of cloud-enabled CAaaS. MERRA/AS enables MapReduce analytics over MERRA reanalysis data collection by bringing together the high-performance computing, scalable data management, and a domain-specific climate data services API. NASA's High-Performance Science Cloud (HPSC) is an example of the type of compute-storage fabric required to support CAaaS. The HPSC comprises a high speed Infinib and network, high performance file systems and object storage, and a virtual system environments specific for data intensive, science applications. These technologies are providing a new tier in the data and analytic services stack that helps connect earthbound, enterprise-level data and computational resources to new customers and new mobility-driven applications and modes of work. In our experience, CAaaS lowers the barriers and risk to organizational change, fosters innovation and experimentation, and provides the agility required to meet our customers' increasing and changing need
Effects of Policies Designed to Keep Firearms from High-Risk Individuals
This article summarizes and critiques available evidence from studies published between 1999 and August 2014 on the effects of policies designed to keep firearms from high-risk individuals in the United States. Some prohibitions for high-risk individuals (e.g., those under domestic violence restraining orders, violent misdemeanants) and procedures for checking for more types of prohibiting conditions are associated with lower rates of violence. Certain laws intended to prevent prohibited persons from accessing firearms -- rigorous permit-to-purchase, comprehensive background checks, strong regulation and oversight of gun dealers, and requiring gun owners to promptly report lost or stolen firearms -- are negatively associated with the diversion of guns to criminals. Future research is needed to examine whether these laws curtail nonlethal gun violence and whether the effects of expanding prohibiting conditions for firearm possession are modified by the presence of policies to prevent diversion
Early and efficient detection of Mycobacterium tuberculosis in sputum by microscopic observation of broth cultures.
Early, efficient and inexpensive methods for the detection of pulmonary tuberculosis are urgently needed for effective patient management as well as to interrupt transmission. These methods to detect M. tuberculosis in a timely and affordable way are not yet widely available in resource-limited settings. In a developing-country setting, we prospectively evaluated two methods for culturing and detecting M. tuberculosis in sputum. Sputum samples were cultured in liquid assay (micro broth culture) in microplate wells and growth was detected by microscopic observation, or in Löwenstein-Jensen (LJ) solid media where growth was detected by visual inspection for colonies. Sputum samples were collected from 321 tuberculosis (TB) suspects attending Bugando Medical Centre, in Mwanza, Tanzania, and were cultured in parallel. Pulmonary tuberculosis cases were diagnosed using the American Thoracic Society diagnostic standards. There were a total of 200 (62.3%) pulmonary tuberculosis cases. Liquid assay with microscopic detection detected a significantly higher proportion of cases than LJ solid culture: 89.0% (95% confidence interval [CI], 84.7% to 93.3%) versus 77.0% (95% CI, 71.2% to 82.8%) (p = 0.0007). The median turn around time to diagnose tuberculosis was significantly shorter for micro broth culture than for the LJ solid culture, 9 days (interquartile range [IQR] 7-13), versus 21 days (IQR 14-28) (p<0.0001). The cost for micro broth culture (labor inclusive) in our study was US 11.35 per sample for the LJ solid culture. The liquid assay (micro broth culture) is an early, feasible, and inexpensive method for detection of pulmonary tuberculosis in resource limited settings
Regulation of normal B-cell differentiation and malignant B-cell survival by OCT2.
The requirement for the B-cell transcription factor OCT2 (octamer-binding protein 2, encoded by Pou2f2) in germinal center B cells has proved controversial. Here, we report that germinal center B cells are formed normally after depletion of OCT2 in a conditional knockout mouse, but their proliferation is reduced and in vivo differentiation to antibody-secreting plasma cells is blocked. This finding led us to examine the role of OCT2 in germinal center-derived lymphomas. shRNA knockdown showed that almost all diffuse large B-cell lymphoma (DLBCL) cell lines are addicted to the expression of OCT2 and its coactivator OCA-B. Genome-wide chromatin immunoprecipitation (ChIP) analysis and gene-expression profiling revealed the broad transcriptional program regulated by OCT2 that includes the expression of STAT3, IL-10, ELL2, XBP1, MYC, TERT, and ADA. Importantly, genetic alteration of OCT2 is not a requirement for cellular addiction in DLBCL. However, we detected amplifications of the POU2F2 locus in DLBCL tumor biopsies and a recurrent mutation of threonine 223 in the DNA-binding domain of OCT2. This neomorphic mutation subtly alters the DNA-binding preference of OCT2, leading to the transactivation of noncanonical target genes including HIF1a and FCRL3 Finally, by introducing mutations designed to disrupt the OCT2-OCA-B interface, we reveal a requirement for this protein-protein interface that ultimately might be exploited therapeutically. Our findings, combined with the predominantly B-cell-restricted expression of OCT2 and the absence of a systemic phenotype in our knockout mice, suggest that an OCT2-targeted therapeutic strategy would be efficacious in both major subtypes of DLBCL while avoiding systemic toxicity.This research was supported by the Intramural Research Program of the NIH, National Cancer Institute, Center for Cancer Research. allows the Murchison Widefield Array to employ a simple RFI blanking scheme and
still calibrate visibilities and form images in the FM radio band. The
techniques described are running autonomously in our calibration and imaging
software, which is currently being used to process an FM-band survey of the

Estimating the Effects of Law Enforcement and Public Health Interventions Intended to Reduce Gun Violence in Baltimore
Baltimore has long been plagued by high rates of homicides, with guns playing an important role. City and law enforcement officials in Baltimore have attributed much of the gun violence to the illegal drug economy and the availability of guns for criminal use. For many years, the most visible and direct approaches employed by the Baltimore Police Department (BPD) to curb gun violence have focused on enforcement of drug laws to reduce violent crime associated with the drug trade. In the most ambitious and resource-intensive efforts, the objective of law enforcement actions has been to "take down" or severely weaken organized groups selling illegal drugs through targeted arrests and prosecutions. Such efforts are intended to both remove violent criminals from communities and, ideally, deter violent crime. Most of these targeted drug law enforcement efforts have been place-focused, targeting "hot spots" for homicides and shootings. Within these hot spots, there is often some degree of targeting of individuals believed to be important drivers of gun violence, based on intelligence gathered, individuals' histories of criminal offending, and individuals' criminal associates.In the early 2000s, Baltimore City leadership encouraged aggressive enforcement of drug laws, resulting in the arrests of tens of thousands of individuals for drug possession and drug distribution. However, beginning mid-2007, the BPD shifted its focus to initiatives aimed at apprehending violent criminals and targeting illegal gun possession. We used data from January 1, 2003, through December 23, 2017, to estimate the effects of place-focused policing and prevention initiatives that were focused on criminal offending involving guns and/or drugs to estimate the effects of those interventions on homicides and nonfatal shootings.
