    Regulation of AQP0 water permeability is enhanced by cooperativity.

    Aquaporin 0 (AQP0), essential for lens clarity, is a tetrameric protein composed of four identical monomers, each of which has its own water pore. The water permeability of AQP0 expressed in Xenopus laevis oocytes can be approximately doubled by changes in calcium concentration or pH. Although each monomer pore functions as a water channel, under certain conditions the pores act cooperatively. In other words, the tetramer is the functional unit. In this paper, we show that changes in external pH and calcium can induce an increase in water permeability that exhibits either a positive cooperativity switch-like increase in water permeability or an increase in water permeability in which each monomer acts independently and additively. Because the concentrations of calcium and hydrogen ions increase toward the center of the lens, a concentration signal could trigger a regulatory change in AQP0 water permeability. It thus seems plausible that the cooperative modes of water permeability regulation by AQP0 tetramers mediated by decreased pH and elevated calcium are the physiologically important ones in the living lens

    Cartilage on the Move: Cartilage Lineage Tracing During Tadpole Metamorphosis

    The reorganization of cranial cartilages during tadpole metamorphosis is a set of complex processes. The fates of larval cartilage-forming cells (chondrocytes) and sources of adult chondrocytes are largely unknown. Individual larval cranial cartilages may either degenerate or remodel, while many adult cartilages appear to form de novo during metamorphosis. Determining the extent to which adult chondrocytes/cartilages are derived from larval chondrocytes during metamorphosis requires new techniques in chondrocyte lineage tracing. We have developed two transgenic systems to label cartilage cells throughout the body with fluorescent proteins. One system strongly labels early tadpole cartilages only. The other system inducibly labels forming cartilages at any developmental stage. We examined cartilages of the skull (viscero- and neurocranium), and identified larval cartilages that either resorb or remodel into adult cartilages. Our data show that the adult otic capsules, tecti anterius and posterius, hyale, and portions of Meckel\u27s cartilage are derived from larval chondrocytes. Our data also suggest that most adult cartilages form de novo, though we cannot rule out the potential for extreme larval chondrocyte proliferation or de- and re-differentiation, which could dilute our fluorescent protein signal. The transgenic lineage tracing strategies developed here are the first examples of inducible, skeleton-specific, lineage tracing in Xenopus

    Effects of urbanization on the occurrence of Batrachochytrium dendrobatidis: do urban environments provide refuge from the amphibian chytrid fungus?

    Batrachochytrium dendrobatidis (Bd) is a widespread pathogenic fungus that is known to cause the disease, chytridiomycosis, which can be lethal to many amphibians. We compared occurrence rates on spring peepers (Pseudacris crucifer) in urban and forested breeding sites in eastern Texas, USA. All study sites were at approximately the same latitude and altitude, and samples were collected at the same time of year to isolate differences in Bd infection rates between habitat types. We found significant differences (p\u3c0.001) in the occurrence of Bd between habitats; with dramatically lower rates of occurrence at urban sites (19.5 %), compared to forested sites (62.9 %). The exact reason for the observed differences in the occurrence of Bd is not known, however, we suspect that warmer temperatures or lower population densities and lower species richness at urban sites all could play a role in our results. Our findings suggest that urban environments may provide a refuge for some amphibians from the pathogen

    Differential jumping performance in newly metamorphosed Blanchard\u27s cricket frogs, Acris blanchardi (Anura: Hylidae), from fish- and invertebrate-dominated ponds

    Organisms that adopt phenotypically plastic anti-predator strategies during larval stages may incur fitness costs later in development. These costs are typically difficult to define in many species. The difficulty of identifying ecological trade-offs may result from experimental comparisons that do not adequately mirror naturally occurring predator-prey relatioships. To examine this, we captured 61 newly metamorphosed Acris blanchardi from ponds dominated either by fish or invertebrate predators. These predators are known to induce alternate phenotypic anti-predator responses at the larval stage. We use jumping performance as a measure of post-metamorphic fitness, and compared the morphological traits and jumping ability of frogs from ponds with and without fish. Frogs from fishless ponds can jump significantly farther than frogs from fish-dominated ponds. Morphological measurements indicated that overall frog size determined differences in jumping ability, with hind-leg length primarily explaining jumping performance. Differences in hind-leg length positively correlated with overall body size, which differed between the two types of ponds and did not result from allometric growth between them.     Maximal uniform convergence rates in parametric estimation problems

    This paper considers parametric estimation problems with independent, identically nonregularly distributed data. It focuses on rate efficiency, in the sense of maximal possible convergence rates of stochastically bounded estimators, as an optimality criterion, largely unexplored in parametric estimation. Under mild conditions, the Hellinger metric, defined on the space of parametric probability measures, is shown to be an essentially universally applicable tool to determine maximal possible convergence rates. These rates are shown to be attainable in general classes of parametric estimation problems

    The Intentional Andragogy Model: A Teaching Framework for Counselor Educators

    Counselor education programs can help support doctoral students in developing teaching philosophies. Yet, limited guidance exists about how counselor educators can help doctoral students integrate andragogies into their teaching philosophy statements. Overlooking andragogy may impede educators from deepening their philosophical beliefs and teaching with authenticity and intentionality. Therefore, we offer the Intentional Andragogy Model (IAM), a process-oriented reimagining of Halbur and Halbur’s Intentional Theory Selection model (ITSM). Counselor educators-in-training can follow this scaffolded step-by-step process to write teaching philosophy statements rooted in their life philosophies and their unique contexts. This model can help counselor educators-in-training root their teaching philosophy statements in andragogy and link andragogy and teaching practice. To illustrate this model, we present a step-by-step case example of how counselor educators can implement the IAM in a doctoral teaching course. We also present considerations for implementation of this model

    Gemini-South + FLAMINGOS Demonstration Science: Near-Infrared Spectroscopy of the z=5.77 Quasar SDSS J083643.85+005453.3

    We report an infrared 1-1.8 micron (J+H-bands), low-resolution (R=450) spectrogram of the highest-redshift radio-loud quasar currently known, SDSS J083643.85+005453.3, obtained during the spectroscopic commissioning run of the FLAMINGOS multi-object, near-infrared spectrograph at the 8m Gemini-South Observatory. These data show broad emission from both CIV 1549 and CIII] 1909, with strengths comparable to lower-redshift quasar composite spectra. The implication is that there is substantial enrichment of the quasar environment, even at times less than a billion years after the Big Bang. The redshift derived from these features is z = 5.774 +/- 0.003, more accurate and slightly lower than the z = 5.82 reported in the discovery paper based on the partially-absorbed Lyman-alpha emission line. The infrared continuum is significantly redder than lower-redshift quasar composites. Fitting the spectrum from 1.0 to 1.7 microns with a power law f(nu) ~ nu^(-alpha), the derived power law index is alpha = 1.55 compared to the average continuum spectral index = 0.44 derived from the first SDSS composite quasar. Assuming an SMC-like extinction curve, we infer a color excess of E(B-V) = 0.09 +/- 0.01 at the quasar redshift. Only approximately 6% of quasars in the optically-selected Sloan Digital Sky Survey show comparable levels of dust reddening.Comment: 10 pages, 1 figure; to appear in the Astrophysical Journal Letter
