    The Neutralization of the Eosinophil Peroxidase Antibody Accelerates Eosinophilic Mucin Decomposition

    Eosinophilic airway inflammation, complicated by bronchial asthma and eosinophilic chronic rhinosinusitis (ECRS), is difficult to treat. The disease may become refractory when eosinophilic mucin associated with eosinophil peroxidase (EPX) and autoantibodies fills in the paranasal sinus and small airway. This study investigated the functional role of an anti-EPX antibody in eosinophilic mucin of ECRS in eosinophilic airway inflammation. Eosinophilic mucin was obtained from patients with ECRS. The effects of the anti-EPX antibody on dsDNA release from eosinophils and eosinophilic mucin decomposition were evaluated. Immunofluorescence or enzyme-linked immunosorbent assays were performed to detect the anti-EPX antibody and its supernatant and serum levels in eosinophilic mucin, respectively. The serum levels of the anti-EPX antibody were positively correlated with sinus computed tomography score and fractionated exhaled nitrogen oxide. Patients with refractory ECRS had higher serum levels of the anti-EPX antibody than those without. However, dupilumab treatment decreased the serum levels of the anti-EPX antibody. Immunoglobulins (Igs) in the immunoprecipitate of mucin supernatants enhanced dsDNA release from eosinophils, whereas the neutralization of Igs against EPX stopped dsDNA release. Furthermore, EPX antibody neutralization accelerated mucin decomposition and restored corticosteroid sensitivity. Taken together, the anti-EPX antibody may be involved in the formulation of eosinophilic mucin and be used as a clinical marker and therapeutic target for intractable eosinophilic airway inflammation

    This paper has focused on analyzing morphological change features of Nha Trang Beach based on the comparison of different beach profiles which were measured from July 2008 to September 2016 corresponding to the Northeast and Southwest monsoons. Additionally, the sediment transport balance was estimated by the method of equity value curve. Studied results show that the beach profiles were accreted and eroded during Southwest and Northeast monsoons respectively, but due to the shortage of materials supply, the beach has the narrowing trend. In this paper, some effects of the existing works in Nha Trang Beach such as Vinpearl, Navy Port, and hydro-dams in upstream area of Cai river have been also analyzed for estimating sedimentary supply to beach segments.Bài báo phân tích đặc điểm biến đổi hình thái địa hình bãi biển Nha Trang trên cơ sở so sánh các trắc diện địa hình bãi biển, với số liệu đo đạc từ tháng 7/2008 đến tháng 9/2016, cụ thể vào các thời kỳ gió mùa Đông Bắc và gió mùa Tây Nam tại các vị trí cố định.     CD69 Signaling in Eosinophils Induces IL-10 Production and Apoptosis via the Erk1/2 and JNK Pathways, Respectively

    INTRODUCTION: Eosinophils contribute to the pathogenesis of allergic diseases, including asthma, allergic rhinitis, and atopic dermatitis. We previously reported that human tissue eosinophils have high CD69 expression compared to blood eosinophils, and its expression is correlated with disease severity and the number of infiltrated eosinophils. However, biological CD69 signaling activity in eosinophils remains unclear. METHODS: CD69 expression on lung tissue eosinophils obtained from mice with ovalbumin-induced asthma was measured using flow cytometry. CD69 crosslinking was performed on eosinophils purified from the spleen of IL-5 transgenic mice to investigate CD69 signaling and its function in eosinophils. Then, qPCR, Western blot, enzyme-linked immunosorbent assay, and survival assay results were analyzed. RESULTS: Surface CD69 expression on lung tissue eosinophils in the asthma mice model was 2.91% ± 0.76%, whereas no expression was detected in the healthy group. CD69-expressed eosinophils intrinsically have an upregulation of IL-10 mRNA expression. Moreover, CD69 crosslinking induced further pronounced IL-10 production and apoptosis; these responses were mediated via the Erk1/2 and JNK pathways, respectively.     Using various chromatography methods, three merosesquiterpenes belonging to sesquiterpene quinone type, neodactyloquinone (1), dactyloquinone D (2), and dactyloquinone C (3) together with two indole derivatives indole-3-aldehyde (4) and indole-3-cacboxylic methyl ester (5) were isolated from the methanol extract of the Vietnamese marine sponge Smenospongia cerebriformis. Their structures were determined by 1D-, 2D-NMR spectra, HR-ESI-MS and in comparison with those reported in the literature. Keywords.     A Methodology to Characterize Riverine Macroplastic Emission Into the Ocean

    Land-based macroplastic is considered one of the major sources of marine plastic debris. However, estimations of plastic emission from rivers into the oceans remain scarce and uncertain, mainly due to a severe lack of standardized observations. To properly assess global plastic fluxes, detailed information on spatiotemporal variation in river plastic quantities and composition are urgently needed. In this paper, we present a new methodology to characterize riverine macroplastic dynamics. The proposed methodology was applied to estimate the plastic emission from the Saigon River, Vietnam. During a 2-week period, hourly cross-sectional profiles of plastic transport were made across the river width. Simultaneously, sub-hourly samples were taken to determine the weight, size and composition of riverine macroplastics (>5 cm). Finally, extrapolation of the observations based on available hydrological data yielded new estimates of daily, monthly and annual macroplastic emission into the ocean. Our results suggest that plastic emissions from the Saigon River are up to four times higher than previously estimated. Importantly, our flexible methodology can be adapted to local hydrological circumstances and data availability, thus enabling a consistent characterization of macroplastic dynamics in rivers worldwide. Such data will provide crucial knowledge for the optimization of future mediation and recycling efforts

    The lumbosacral angle does not reflect progressive tethered cord syndrome in children with spinal dysraphism

    Purpose: Our goal was to validate the hypothesis that the lumbosacral angle (LSA) increases in children with spinal dysraphism who present with progressive symptoms and signs of tethered cord syndrome (TCS), and if so, to determine for which different types and/or levels the LSA would be a valid indicator of progressive TCS. Moreover, we studied the influence of surgical untethering and eventual retethering on the LSA. Methods: We retrospectively analyzed the data of 33 children with spinal dysraphism and 33 controls with medulloblastoma. We measured the LSA at different moments during follow-up and correlated this with progression in symptomatology. Results: LSA measurements had an acceptable intra- and interobserver variability, however, some children with severe deformity of the caudal part of the spinal column, and for obvious reasons those with caudal regression syndrome were excluded. LSA measurements in children with spinal dysraphism were significantly different from the control group (mean LSA change, 21.0° and 3.1° respectively). However, both groups were not age-matched, and when dividing both groups into comparable age categories, we no longer observed a significant difference. Moreover, we did not observe a significant difference between 26 children with progressive TCS as opposed to seven children with stable TCS (mean LSA change, 20.6° and 22.4° respectively). Conclusions: We did not observe significant differences in LSA measurements for children with clinically progressive TCS as opposed to clinically stable TCS.     Aperture Photometry Tool Versus SExtractor for Noncrowded Fields

    Outputs from new software program Aperture Photometry Tool (APT) are compared with similar outputs from SExtractor for sources extracted from R-band optical images acquired by the Palomar Transient Factory (PTF), infrared mosaics constructed from Spitzer Space Telescope images, and a processed visible/near-infrared image from the Hubble Legacy Archive (HLA). Two large samples from the PTF images are studied, each containing around 3 × 10^3 sources from noncrowded fields. The median values of source-intensity relative percentage differences between the two software programs, computed separately for two PTF samples, are +0.13% and +0.17%, with corresponding statistical dispersions of 1.43% and 1.84%, respectively. For the Spitzer mosaics, a similar large sample of extracted sources for each of channels 1–4 of Spitzer’s Infrared Array Camera (IRAC) are analyzed with two different sky annulus sizes, and we find that the median and modal values of source-intensity relative percentage differences between the two software programs are between -0.5% and +2.0%, and the corresponding statistical dispersions range from 1.4 to 6.7%, depending on the Spitzer IRAC channel and sky annulus. The results for the HLA image are mixed, as might be expected for a moderately crowded field. The comparisons for the three different kinds of images show that there is generally excellent agreement between APT and SExtractor. Differences in source-intensity uncertainty estimates for the PTF images amount to less than 3% for the PTF sources, and these are potentially caused by SExtractor’s omission of the sky background uncertainty term in the formula for source-intensity uncertainty, as well as differing methods of sky background estimation
