SejtfelszĂni receptor minták mint lehetsĂ©ges diagnosztikai eszközök = Cell surface receptor patterns as potential diagnostic tools
Modern biofizikai mĂłdszerek kombinált alkalmazásával (FRET, konfokális mikroszkĂłpia, fluoreszcencia korreláciĂłs spektroszkĂłpia, stb.) a T sejtes immunválaszban rĂ©sztvevĹ‘ kulcsfontosságĂş membránfehĂ©rjĂ©k sejtfelszĂni elrendezĹ‘dĂ©sĂ©t, asszociáciĂłs mintázatait, kölcsönhatásait tanulmányoztuk. A kialakulĂł mintázatok a jelátvitel egyes lĂ©pĂ©seinek szervezĂ©se rĂ©vĂ©n alapvetĹ‘en befolyásolják a sejtek fiziolĂłgiás működĂ©sĂ©t, másrĂ©szrĹ‘l potenciális terápiás cĂ©lpontok lehetnek. Megalkottuk az IL-2/15R rendszer dinamikus heterotetramer modelljĂ©t: a releváns citokin kötĹ‘dĂ©se a receptor alegysĂ©gek átrendezĂ©sĂ©vel alakĂtja ki a megfelelĹ‘ nagy affinitásĂş heterotrimert. Kimutattuk, hogy az IL-2/15R Ă©s az MHC molekulák közös szuperklasztereket alkotnak a T sejtek lipid tutajaiban. Jurkat T sejteken Ă©s aktivált CTL-eken kimutattuk a Kv1.3 káliumcsatornák Ă©s a TCR kolokalizáciĂłját. A CTL-target sejt kölcsönhatás során a CTL-ek Kv1.3 csatornái feldĂşsulnak a kĂ©t sejt közötti immunolĂłgiai szinapszisban, felvetve a TCR Ă©s a Kv1.3 kölcsönös szabályozásának lehetĹ‘sĂ©gĂ©t. A korábban limfoid Ă©s kolorektális karcinĂłma sejteken feltárt, az MHC I, II Ă©s az ICAM-1 rĂ©szvĂ©telĂ©vel kialakulĂł asszociáciĂłs mintázatok jelenlĂ©tĂ©t uveális melanĂłma sejteken is kimutattuk. RNS interferencia felhasználásával igazoltuk, hogy a kolorektális karcinĂłma sejteken az MHC I Ă©s ICAM-1 klasztereit Ă©s domĂ©n szintű elrendezĹ‘dĂ©sĂ©t e kĂ©t fehĂ©rje relatĂv expressziĂłs szintje dinamikus mĂłdon szabályozza. | We investigated the cell surface distribution, association motifs and interactions of key membrane proteins involved in T cell mediated immune responses by applying modern biophysical techniques (FRET, confocal microscopy, fluorescence correlation spectroscopy, etc.). The studied protein patterns have a profound influence on the physiological function of the cells via organizing certain signaling steps, and may serve as potential therapeutic targets. We proposed a dynamic heterotetrameric model for the IL-2/15R system: binding of the cytokine rearranges the receptor subunits to form the relevant heterotrimeric high affinity receptor. We demonstrated that IL-2/15 receptors and MHC glycoproteins form superclusters in lipid rafts of T cells. We showed that Kv1.3 K+ channels and TCR are colocalized on Jurkat T cells and CTLs. Kv1.3 channels are enriched in the immune synapse formed between the CTL and the target cell, raising the possibility of reciprocal regulation of TCR and Kv1.3. We identified association patterns of MHC I, II and ICAM-1 on uveal melanoma cells similar to those previously found on lymphoid and colorectal carcinoma cells. By using RNA interference we demonstrated that the clusterization and interaction of MHC I and ICAM-1 are dynamically regulated by the relative expression levels of these proteins
Multiple Binding Sites for Melatonin on Kv1.3
AbstractMelatonin is a small amino acid derivative hormone of the pineal gland. Melatonin quickly and reversibly blocked Kv1.3 channels, the predominant voltage-gated potassium channel in human T-lymphocytes, acting from the extracellular side. The block did not show state or voltage dependence and was associated with an increased inactivation rate of the current. A half-blocking concentration of 1.5mM was obtained from the reduction of the peak current. We explored several models to describe the stoichiometry of melatonin-Kv1.3 interaction considering one or four independent binding sites per channel. The model in which the occupancy of one of four binding sites by melatonin is sufficient to block the channels gives the best fit to the dose-response relationship, although all four binding sites can be occupied by the drug. The dissociation constant for the individual binding sites is 8.11mM. Parallel application of charybdotoxin and melatonin showed that both compounds can simultaneously bind to the channels, thereby localizing the melatonin binding site out of the pore region. However, binding of tetraethylammonium to its receptor decreases the melatonin affinity, and vice versa. Thus, the occupancy of the two separate receptor sites allosterically modulates each other

ProjektĂĽnk cĂ©lkitűzĂ©se a transzmembrán jelátvitelben rĂ©sztvevĹ‘ egyes receptorok Ă©s ioncsatornák működĂ©sĂ©nek multidiszciplináris megközelĂtĂ©se volt. Kimutattuk a TASK-3 K+-csatorna expressziĂłját Ă©s döntĹ‘en mitokondriális lokalizáciĂłját melanoma malignum, ill. malignus transzformáciĂłt nem mutatĂł sejteken egyaránt. JellemeztĂĽk a dendritikus sejtek (DS) Ă©rĂ©se Ă©s ioncsatorna expressziĂłja közötti kapcsolatot. Feltártuk, hogy az elsĹ‘dleges Ca2+ jelátviteli utat a CRAC csatornák biztosĂtják, fĂĽggetlenĂĽl a DS-ek differenciáltsági fokátĂłl. Tanulmányoztuk az ic. Ca2+ homeosztázis Ă©s a purinerg jelátvitel kapcsolatát ingerlĂ©keny Ă©s nem-ingerlĂ©keny sejteken Ă©s karakterizáltuk a folyamatban szerepet játszĂł purinoreceptorok tĂpusát. Kimutattuk, hogy az SR-ban kifejezĹ‘dĹ‘ Trisk 95 fehĂ©rje negatĂvan szabályozza a RyR funkciĂłját; Ăgy mind a lokális, mind pedig a globális Ca2+-jelek kialakulását gátolja. Rámutattunk, hogy a közös gamma receptorcsaládba tartozĂł IL-2/15 Ă©s -9 receptorok sejtfelszĂni szervezĹ‘dĂ©sĂ©t feltehetĹ‘en azonos rendezĹ‘ elvek alakĂtják ki. Tanulmányoztuk a T sejtek membránpotenciálját meghatározĂł Kv1.3 K+-csatorna immunszinapszis-beli lokalizáciĂłjának funkcionális következmĂ©nyeit. Kidolgoztunk egy Ăşjfajta FRET mĂ©rĂ©si mĂłdszert, amivel az IL-2 Ăştvonal által aktivált transzkripciĂłs faktorok konformáciĂłját határoztuk meg. EredmĂ©nyeink hozzásegĂtenek a T-sejtes immunválasz mechanizmusának megĂ©rtĂ©sĂ©hez. | The main aim of our project was to characterize selected receptors and ion channels in transmembrane signaling processes using a multidisciplinary approach. We detected expression and mainly mitochondrial localization of TASK-3 K+-channels both in malignant melanomas and benign melanocytes. We described the relationship between maturation and ion channel expression in dendritic cells (DCs). We showed that the primary Ca2+ signaling pathway is mediated by CRAC channels, independent of the degree of maturation. We studied the connection between Ca2+ homeostasis and purinerg signaling in excitable and non-excitable cells, and characterized the type of purinerg receptors involved in this process. We demonstrated that the Trisk 95 protein expressed in the SR negatively regulates RYR function; thus it inhibits the generation of both local and global Ca2+ signals. We revealed that the cell surface organization of the members of the common gamma chain family, IL-2/15 and 9 receptors, is probably organized by the same general principles. We studied the functional consequences of the localization of Kv1.3 K+ channels in the immune synapse. We elaborated a new FRET method, by which we described the conformation of transcription factors induced by the IL-2 pathway. Our results contribute to understanding the mechanism of T cell mediated immune responses
