    Delighting and Detesting Engagement : Emotional Politics of Junk News

    How do audiences make sense of and interact with political junk news on Facebook? How does the platform's "emotional architecture" intervene in these sense-making, interactive processes? What kinds of mediated publics emerge on and through Facebook as a result? We study these questions through topic modeling 40,500 junk news articles, quantitatively analyzing their engagement metrics, and a qualitative comment analysis. This exploratory research design allows us to move between levels of public discourse, zooming in from cross-outlet talking points to microsociological processes of meaning-making, interaction, and emotional entrainment taking place within the comment boxes themselves. We propose the concepts of delighting and detesting engagement to illustrate how the interplay between audiences, platform architecture, and political junk news generates a bivalent emotional dynamic that routinely divides posts into highly "loved" and highly "angering." We argue that high-performing (or in everyday parlance, viral) junk news bring otherwise disparate audience members together and orient their dramatic focus toward objects of collective joy, anger, or concern. In this context, the nature of political junk news is performative as they become resources for emotional signaling and the construction of group identity and shared feeling on social media. The emotions that animate junk news audiences typically refer back to a transpiring social relationship between two political sides. This affectively loaded "us" versus "them" dynamic is both enforced by Facebook's emotional architecture and made use of by junk news publishers.Peer reviewe

    Identifying Teacher, School and District Characteristics Associated with Elementary Teachers' Use of Technology:A Multilevel Perspective

    Over the past decade, investment in technology for schools has increased at a dramatic rate. Although policy makers are eager to understand the ways in which technology use in schools is affecting student learning, we believe that a critical preliminary step toward assessing the impacts of technology on teaching and learning requires the examination of the varied uses of technology in schools as well as the contexts that are likely to affect the use of technology in the classroom as a teaching and learning tool. Previous research examining technology use has focused on teacher characteristics and has neglected to explore the potentially alterable, organizational characteristics that may be affecting the adoption and use of technology in the classroom. In light of this argument and using survey data collected from 1490 elementary classroom teachers in 96 schools in 22 Massachusetts districts, this research examines how technology is being used by elementary school teachers, and examines the school and district organizational characteristics that are associated with increased use of technology as a teaching and learning tool. In addition to examining technology-use as a multi-faceted construct, using multilevel regression techniques this study provides evidence that schools’ organizational characteristics are associated with teachers’ use of technology in the classroom. Organizational characteristics such as districts’ and schools’ leadership practices and emphasis on technology, the type and amount of technology-related professional development available to teachers, as well as the amount of technology-related restrictive policies in place were found to be associated with the four measures of teachers’ use of technology examined in this study. Individual teacher characteristics such as constructivist beliefs, higher confidence using technology and positive beliefs about the efficacy of technology were each found to be associated with increased use of technology in the classroom

    Laminin Modulates the Stem Cell Population in LM05-E Murine Breast Cancer Cells Through the Activation of the MAPK/ERK Pathway

    PURPOSE: We investigated the effects of laminin on the fraction of cells with self-renewing capacity in the estrogen-dependent, tamoxifen-sensitive LM05-E breast cancer cell line. We also determined whether laminin affected the response to tamoxifen. MATERIALS AND METHODS: The LM05-E breast cancer cell line was used as a model for all experiments. Aldehyde dehydrogenase (ALDH) activity, clonogenic and mammosphere assays were performed to measure the effects of laminin on modulation of the stem cell subpopulation. Pluripotent gene expression was analyzed by RT-PCR. The involvement of the MAPK/ERK pathway was determined using specific inhibitors. The effects of laminin on the response to tamoxifen were determined and the involvement of alpha-6 integrin was investigated. RESULTS: We found that pre-treatment with laminin leads to a decrease in cells with the ability to form mammospheres that was accompanied by a decrease in ALDH activity. Moreover, exposure of mammospheres to laminin reduced the capacity to form secondary mammospheres and decreased the expression of Sox-2, Nanog and Oct-4. We previously reported that 4-OH-tamoxifen leads to an increase in the expression of these genes in LM05-E cells. Treatment with signaling pathway inhibitors revealed that the MAPK/ERK pathway mediates the effects of laminin. Finally, laminin induced tamoxifen resistance in LM05-E cells through alpha 6 integrin. CONCLUSIONS: Our results suggest that the final number of cells with self-renewing capacity in estrogen-dependent breast tumors may result from the combined effects of endocrine treatment and microenvironmental cues.Fil: Berardi, Damian Emilio.     Ecological phylogeography and coalescent models suggest a linear population expansion of Anastrepha fraterculus (Diptera: Tephritidae) in southern South America

    This work is a first approach to an integrated view of the genetics, ecology and dispersion patterns of Anastrepha fraterculus in southern South America. We studied the association of genetic variation with geographical patterns and environmental variables to provide insight into the crucial factors that drive the structure and dynamics of fly populations. Data from a 417 bp mitochondrial COII gene fragment from seven Argentinian populations and one South Brazilian population (from five ecoregions grouped in three biomes) were used to identify population clusters using a model-based Bayesian phylogeographical and ecological clustering approach. The sequences were also analysed under a coalescent model to evaluate historical demographic changes. We identified 19 different haplotypes and two clusters differing in all the environmental covariables. The assumption of neutral evolution and constant population size was rejected, and the population growth parameters suggested a linear population expansion starting 2500 years before present. The most likely ancestral location is Posadas, from where A. fraterculus would have expanded southwards and westwards in Argentina. This result is consistent with Holocene changes and anthropic factors related to the expansion of the Tupí-Guaraní culture, 3000-1500 years before present.Fil: Vilardi, Juan Cesar.     Hypothesis and theory : a pathophysiological concept of stroke-induced acute phase response and increased intestinal permeability leading to secondary brain damage

    Gut integrity impairment leading to increased intestinal permeability (IP) is hypothesized to be a trigger of critically illness. Approximately 15–20% of human ischemic stroke (IS) victims require intensive care, including patients with impaired level of consciousness or a high risk for developing life-threatening cerebral edema. Local and systemic inflammatory reactions are a major component of the IS pathophysiology and can significantly aggravate brain tissue damage. Intracerebral inflammatory processes following IS have been well studied. Until now, less is known about systemic inflammatory responses and IS consequences apart from a frequently observed post-IS immunosuppression. Here, we provide a hypothesis of a crosstalk between systemic acute phase response (APR), IP and potential secondary brain damage during acute and subacute IS stages supported by preliminary experimental data. Alterations of the acute phase proteins (APPs) C-reactive protein and lipopolysaccharide-binding protein and serum level changes of antibodies directed against Escherichia coli-cell extract antigen (IgA-, IgM-, and IgG-anti-E. coli) were investigated at 1, 2, and 7 days following IS in ten male sheep. We found an increase of both APPs as well as a decrease of all anti-E. coli antibodies within 48 h following IS. This may indicate an early systemic APR and increased IP, and underlines the importance of the increasingly recognized gut-brain axis and of intestinal antigen release for systemic immune responses in acute and subacute stroke stages

    Protection efficacy of Argentinian isolates of Mycobacterium avium subsp. paratuberculosis with different genotypes and virulence in a murine model

    Paratuberculosis is a chronic disease caused by Mycobacterium avium subsp. paratuberculosis (Map). The disease causes economic losses and, therefore, it is imperative to follow proper control strategies, which should include an effective vaccine. Several strategies have assessed the virulence and immune response of Map strains that could be used as a vaccine. This study evaluates the degree of virulence, immune response, and protection of Argentinian strains of Map with different genotype in a murine model. Four local isolates (Cattle type) with different genotypes (analyzed by MIRU-VNTR and SSRs) were selected and evaluated in a virulence assay in BALB/c mice. This assay allowed us to differentiate virulent and low-virulence Map strains. The less virulent strains (1543/481 and A162) failed to induce a significant production of the proinflammatory cytokine IFNg, whereas the virulent strain 6611 established infection along with a proinflammatory immune response. On the other hand, the virulent strain 1347/498 was efficient in establishing a persistent infection, but failed to promote an important Th1 response compared with 6611 at the evaluated time. We selected the low-virulence strain 1543/498 as a live vaccine and the virulent strain 6611 as a live and inactivated vaccine in a protection assay in mice. Strain 1543/481 failed to protect the animals from challenge, whereas strain 6611, in its live and inactivated form, significantly reduced the CFUs count in the infected mice, although they had different immunological response profiles. The inactivated virulent strain 6611 is a potential vaccine candidate against paratuberculosis to be tested in cattle.Fil: Colombatti Olivieri, María Alejandra.     Experimental Certification of Quantum Transmission via Bell's Theorem

    Quantum transmission links are central elements in essentially all implementations of quantum information protocols. Emerging progress in quantum technologies involving such links needs to be accompanied by appropriate certification tools. In adversarial scenarios, a certification method can be vulnerable to attacks if too much trust is placed on the underlying system. Here, we propose a protocol in a device independent framework, which allows for the certification of practical quantum transmission links in scenarios where minimal assumptions are made about the functioning of the certification setup. In particular, we take unavoidable transmission losses into account by modeling the link as a completely-positive trace-decreasing map. We also, crucially, remove the assumption of independent and identically distributed samples, which is known to be incompatible with adversarial settings. Finally, in view of the use of the certified transmitted states for follow-up applications, our protocol moves beyond certification of the channel to allow us to estimate the quality of the transmitted state itself. To illustrate the practical relevance and the feasibility of our protocol with currently available technology we provide an experimental implementation based on a state-of-the-art polarization entangled photon pair source in a Sagnac configuration and analyze its robustness for realistic losses and errors.Comment: 34 pages, 14 figure

    Autophagy: Friend or Foe in Breast Cancer Development, Progression, and Treatment

    Autophagy is a catabolic process responsible for the degradation and recycling of long-lived proteins and organelles by lysosomes. This degradative pathway sustains cell survival during nutrient deprivation, but in some circumstances, autophagy leads to cell death. Thereby, autophagy can serve as tumor suppressor, as the reduction in autophagic capacity causes malignant transformation and spontaneous tumors. On the other hand, this process also functions as a protective cell-survival mechanism against environmental stress causing resistance to antineoplastic therapies. Although autophagy inhibition, combined with anticancer agents, could be therapeutically beneficial in some cases, autophagy induction by itself could lead to cell death in some apoptosis-resistant cancers, indicating that autophagy induction may also be used as a therapy. This paper summarizes the most important findings described in the literature about autophagy and also discusses the importance of this process in clinical settings
