    An LC inventory based on representative and coherent farm types

    There is a need for valid and representative data regarding the production, resource use and emissions from typical farming systems in Denmark for analysis of the environmental impact of different systems and as input to product oriented analyses such as Life Cycle Assess-ments of basic food items. An inventory of 31 farm types was constructed on the basis of 2138 farm accounts from 1999 selected and weighted to be representative for the Danish farming sector. The farm accounts were grouped according to the major soil types, the num-ber of standard working hours, the most important enterprise (dairy, pig, different cash crops) and the stocking rate (livestock units per hectare). For each group the account data on the average inputs and outputs, land use and herd structure was used to establish a farm type model with coherency between livestock production, total feed use, land use, yields, im-ported feed, home-grown feed, manure production, fertiliser use and crop production. The set of farm types were scaled up to national level thus representing the whole Danish agricul-tural sector for the included products. The sum of area and yield by crop, number and pro-duction by livestock type and the use of fertiliser, energy and concentrated feed was checked against national level statistics and corrected accordingly across all farm types. Resource use and emissions in each farm type was established using standard nutrient concentrations and models for nutrient cycling, energy use and emissions of e.g. ammonia, nitrous oxides and methane. For LCA the product oriented inventory was established using system expan-sion rather than allocations to account for the secondary enterprises in the livestock farm types. Data are made available on a web-based database and may be used for analyses of the primary production systems or as input for LCA across the whole production chain

    Identification of a novel type of spacer element required for imprinting in fission yeast

    Asymmetrical segregation of differentiated sister chromatids is thought to be important for cellular differentiation in higher eukaryotes. Similarly, in fission yeast, cellular differentiation involves the asymmetrical segregation of a chromosomal imprint. This imprint has been shown to consist of two ribonucleotides that are incorporated into the DNA during laggingstrand synthesis in response to a replication pause, but the underlying mechanism remains unknown. Here we present key novel discoveries important for unravelling this process. Our data show that cis-acting sequences within the mat1 cassette mediate pausing of replication forks at the proximity of the imprinting site, and the results suggest that this pause dictates specific priming at the position of imprinting in a sequence-independent manner. Also, we identify a novel type of cis-acting spacer region important for the imprinting process that affects where subsequent primers are put down after the replication fork is released from the pause. Thus, our data suggest that the imprint is formed by ligation of a not-fullyprocessed Okazaki fragment to the subsequent fragment.     Mapping leaf nitrogen and leaf area index in European landscapes using high spatial resolution satellite data and the REGularized canopy reFLECtance (REGFLEC) model

    Leaf biochemistry and biophysical parameters are important for simulating soil‐vegetation‐atmosphere exchanges of energy, water, CO2 and ammonia. The accumulation of leaf nitrogen (N) in vegetation canopies is a major component of the ecosystem N balance, and leaf N concentration and leaf area index (LAI) are important determinants of the maximum CO2 uptake by plants and trees. In NEU, high spatial resolution remote sensing data from the SPOT satellite were acquired to prepare maps of leaf N and LAI for 5 European landscapes. Mapping was conducted using the REGFLEC model which is an automatic and image‐based methodology developed for regional chlorophyll (Cab) and LAI estimation (ie. Houborg and Andersen, JARS 3, 2009). REGFLEC combines models for atmospheric correction (6S), canopy reflectance (ACRM) and leaf optics (PROSPECT). Model performance previously proved promising in Denmark and in Maryland, USA. In this study, REGFLEC performance is evaluated and discussed using field measurements of leaf N, SPADmeter data and LAI in Denmark, Poland, Scotland, the Netherlands and Italy. The inverse model estimations of soil reflectance parameters and canopy parameters are discussed in relation to the prevailing soil types and vegetation characteristics of land cover classes across the 5 European landscapes

    Contamination of the Arctic reflected in microbial metagenomes from the Greenland ice sheet

    Globally emitted contaminants accumulate in the Arctic and are stored in the frozen environments of the cryosphere. Climate change influences the release of these contaminants through elevated melt rates, resulting in increased contamination locally. Our understanding of how biological processes interact with contamination in the Arctic is limited. Through shotgun metagenomic data and binned genomes from metagenomes we show that microbial communities, sampled from multiple surface ice locations on the Greenland ice sheet, have the potential for resistance to and degradation of contaminants. The microbial potential to degrade anthropogenic contaminants, such as toxic and persistent polychlorinated biphenyls, was found to be spatially variable and not limited to regions close to human activities. Binned genomes showed close resemblance to microorganisms isolated from contaminated habitats.     Standardized high-throughput evaluation of cell-based compound screens

    <p>Abstract</p> <p>Background</p> <p>High-throughput screening of pharmaceutical compound activity in tissue culture experiments requires time-consuming repeated analysis of the large amounts of data generated. Automation of the evaluation procedure and assessment of measurement accuracy can save time and improve the comparability of results.</p> <p>Results</p> <p>We present a tool for simultaneous evaluation of an arbitrary number of compound screens including a standardized statistical validation. It is provided as a novel R package with a Tcl/Tk-based GUI for convenient use in the lab and runs on usual platforms like Linux, Windows and Mac OS. In a compound screen of lung cancer cells, the tool was successfully and efficiently applied for data analysis.</p> <p>Conclusion</p> <p>The package provides an efficient and intuitive platform for automatic evaluation of compound screens, improving the performance and standardization of data analysis.</p

    Combined Exposure to Anti-Androgens Exacerbates Disruption of Sexual Differentiation in the Rat

    OBJECTIVE: The aim of this study was to assess whether the joint effects of three androgen receptor antagonists (vinclozolin, flutamide, procymidone) on male sexual differentiation after in utero and postnatal exposures can be predicted based on dose-response data of the individual chemicals. METHODS: Test chemicals and mixtures were administered by gavage to time-mated nulliparous, young adult Wistar rats from gestational day 7 to the day before expected birth, and from postnatal days 1-16. Changes in anogenital distance (AGD) and nipple retention (NR) in male offspring rats were chosen as end points for extensive dose-response studies. Vinclozolin, flutamide, and procymidone were combined at a mixture ratio proportional to their individual potencies for causing retention of six nipples in male offspring. RESULTS: With AGD as the end point, the joint effects of the three anti-androgens were essentially dose additive. The observed responses for NR were slightly higher than those expected on the basis of dose addition. A combination of doses of each chemical, which on its own did not produce statistically significant AGD alterations, induced half-maximal mixture effects. At individual doses associated with only modest effects on NR, the mixture induced NR approaching female values in the males. CONCLUSIONS: Effects of a mixture of similarly acting anti-androgens can be predicted fairly accurately on the basis of the potency of the individual mixture components by using the dose addition concept. Exposure to anti-androgens, which individually appears to exert only small effects, may induce marked responses in concert with, possibly unrecognized, similarly acting chemicals
