    Environmental Flow Scenarios for a Regulated River System: Projecting Catchment-Wide Ecosystem Benefits and Consequences for Hydroelectric Production

    To enable prioritization among measures for ecological restoration, knowing the expected benefits and consequences of implementation is imperative but rarely explicitly quantified. We developed a novel method to prioritize among environmental flow measures to rehabilitate ecosystems in the Ume River catchment in northern Sweden, a river system heavily regulated for hydropower production. Our strategy was to identify measures with minimal impact on hydropower production while providing substantial environmental benefits. Based on field surveys of remaining natural values and potential for ecological rehabilitation, we quantified the projected gain in habitat area of implementing environmental flows for target organism groups, for example, lotic fish species and riparian vegetation, along the whole river length. We quantified the consequences for hydropower production by identifying a set of hydropower operational rules reflecting the constraints added by environmental flows. We then used production optimization software to calculate changes in hydropower production and revenues. Implementing restrictions on zero-flow events by mandating minimum discharge at all run-of-river hydropower stations and allocating 1%-12% of mean annual discharge to bypassed reaches in the entire catchment would result in a 2.1% loss of annual electricity production. Adding flow to fishways would increase the loss to 3.1% per year. With implementation of more natural water-level fluctuations in run-of-river impoundments, the loss increases to 3.8%. These actions would increase the habitat for lotic species like the grayling Thymallus more than threefold and increase the area of riparian vegetation by about 66%. Our method forms a basis for ongoing implementation of nationwide environmental rehabilitation schemes

    Joint effect of phosphorus limitation and temperature on alkaline phosphatase activity and somatic growth in Daphnia magna

    Alkaline phosphatase (AP) is a potential biomarker for phosphorus (P) limitation in zooplankton. However, knowledge about regulation of AP in this group is limited. In a laboratory acclimation experiment, we investigated changes in body AP concentration for Daphnia magna kept for 6 days at 10, 15, 20 and 25°C and fed algae with 10 different molar C:P ratios (95–660). In the same experiment, we also assessed somatic growth of the animals since phosphorus acquisition is linked to growth processes. Overall, non-linear but significant relationships of AP activity with C:P ratio were observed, but there was a stronger impact of temperature on AP activity than of P limitation. Animals from the lowest temperature treatment had higher normalized AP activity, which suggests the operation of biochemical temperature compensation mechanisms. Body AP activity increased by a factor of 1.67 for every 10°C decrease in temperature. These results demonstrate that temperature strongly influences AP expression. Therefore, using AP as a P limitation marker in zooplankton needs to consider possible confounding effects of temperature. Both temperature and diet affected somatic growth. The temperature effect on somatic growth, expressed as the Q10 value, responded non-linearly with C:P, with Q10 ranging between 1.9 for lowest food C:P ratio and 1.4 for the most P-deficient food. The significant interaction between those two variables highlights the importance of studying temperature-dependent changes of growth responses to food quality

    Decreased Th1-Type Inflammatory Cytokine Expression in the Skin Is Associated with Persisting Symptoms after Treatment of Erythema Migrans

    Background: Despite the good prognosis of erythema migrans (EM), some patients have persisting symptoms of various character and duration post-treatment. Several factors may affect the clinical outcome of EM, e. g. the early interaction between Borrelia (B.) burgdorferi and the host immune response, the B. burgdorferi genotype, antibiotic treatment as well as other clinical circumstances. Our study was designed to determine whether early cytokine expression in the skin and in peripheral blood in patients with EM is associated with the clinical outcome. Methods: A prospective follow-up study of 109 patients with EM was conducted at the A land Islands, Finland. Symptoms were evaluated at 3, 6, 12 and 24 months post-treatment. Skin biopsies from the EM and healthy skin were immunohistochemically analysed for expression of interleukin (IL)-4, IL-10, IL-12p70 and interferon (IFN)-gamma, as well as for B. burgdorferi DNA. Blood samples were analysed for B. burgdorferi antibodies, allergic predisposition and levels of systemic cytokines. Findings: None of the patients developed late manifestations of Lyme borreliosis. However, at the 6-month follow-up, 7 of 88 patients reported persisting symptoms of diverse character. Compared to asymptomatic patients, these 7 patients showed decreased expression of the Th1-associated cytokine IFN-gamma in the EM biopsies (p = 0.003). B. afzelii DNA was found in 48%, B. garinii in 15% and B. burgdorferi sensu stricto in 1% of the EM biopsies, and species distribution was the same in patients with and without post-treatment symptoms. The two groups did not differ regarding baseline patient characteristics, B. burgdorferi antibodies, allergic predisposition or systemic cytokine levels. Conclusion: Patients with persisting symptoms following an EM show a decreased Th1-type inflammatory response in infected skin early during the infection, which might reflect a dysregulation of the early immune response.     Genome-wide structural variant analysis identifies risk loci for non-Alzheimer’s dementias

    We characterized the role of structural variants, a largely unexplored type of genetic variation, in two non-Alzheimer’s dementias, namely Lewy body dementia (LBD) and frontotemporal dementia (FTD)/amyotrophic lateral sclerosis (ALS). To do this, we applied an advanced structural variant calling pipeline (GATK-SV) to short-read whole-genome sequence data from 5,213 European-ancestry cases and 4,132 controls. We discovered, replicated, and validated a deletion in TPCN1 as a novel risk locus for LBD and detected the known structural variants at the C9orf72 and MAPT loci as associated with FTD/ALS. We also identified rare pathogenic structural variants in both LBD and FTD/ALS. Finally, we assembled a catalog of structural variants that can be mined for new insights into the pathogenesis of these understudied forms of dementia

    This chapter is about musical leadership and specifically the role of the choral conductor. The conductor role is familiar to anyone who has been singing in a school choir or have been the audience of live or televised concerts. The role is prominent in concert programmes and associated with ensembles whenever they are profiled or critiqued. The presence of a conductor is largely taken for granted by the layman as well as the scholar, as self-evident as the presence of musicians and singers. The purpose of this chapter is to look through this veil of self-evidence by reflecting on what conducting is, why we need it it and what makes it work. Contrary to most writing on conducting and conductors, this chapter will take the choral singer’s viewpoint, describing how musical leadership is perceived and experienced by those whom conducting is intended to impact
