    Tissue culture and subsequent field evaluation of strawberry

    Strawberry plants of cv. Redlands Crimson, free of strawberry mild yellow edge virus (MYEV), have been produced by apical tip culture on a modified Adams (1972) medium. Explants from six other cultivars were grown on this medium and considerable variation in growth response was observed. No shoot growth was obtained on this medium with explants of cv. Phenomenal, however, growth was initiated on a modified Lee and De Fossard (1975) medium. Low media pH (4.2) inhibited shoot growth. Yield of six clones of cv. Redlands Crimson, free of MYEV and one virus infected clone were monitored for a fruiting season. Removal of MYEV had no effect on fruit quality, but uninfected clones had a 13% yield advantage of marketable fruit over the whole season. This increase was shown to occur during the two fruiting flushes and was a result of more vigorous plants having the capacity to produce more fruit. Commercial plantings of clones free of MYEV over five seasons have been characterised by high early yields

    An improved embryo-rescue protocol for a Carica interspecific hybrid

    An improved embryo-rescue protocol was developed for embryos (90 days old) of Carica papaya L. (Clone 2001), and subsequently was utilised for efficient production of interspecific hybrids of C. papaya × C. cauliflora Jacq. from 90- to 120-day-old embryos. Pre-incubation of C. papaya embryos for 7 days on a germination medium containing half-strength De Fossard nutrients supplemented with gibberellic acid (10 μM), 6-benzylamino-purine (0.25 μM), alpha-naphthalene-acetic acid (0.25 μM). sucrose (58 mM) and agar (8 g L-1) supported 100% germination. Subsequent transfer of germinated embryos to a nutrient medium that was identical, except that it was free of plant growth regulator, allowed good growth but induced shoot etiolation and callus production. Reducing the pre-incubation of C. papaya embryos on this medium to 5 days before transfer to the medium free of plant growth regulator produced similarly high germination (96%), but allowed for the production of good quality seedlings that were unetiolated and free of unwanted callus. For interspecific hybrids, a 5-day pre-incubation of the embryos on a liquid formulation was better than the solid formulation as it promoted better growth and vigour of the normally abortive interspecific hybrid embryos. Using the improved protocol, 1981 of 2100 (94%) interspecific hybrid embryos consisting of single and multiple forms were germinated. In all cases, the germinating multiple embryos underwent further embryogenesis that allowed for the production of 485 (25%) morphologically normal hybrid plants grown in soil in the glasshouse

    Sub-arcsecond radio and optical observations of the likely counterpart to the gamma-ray source 2FGL J2056.7+4939

    We have searched and reviewed all multi- wavelength data available for the region towards the gamma-ray source 2FGL J2056.7+4939 in order to con- strain its possible counterpart at lower energies. As a result, only a point-like optical/infrared source with flat-spectrum radio emission is found to be consistent with all X-ray and gamma-ray error circles. Its struc- ture is marginally resolved at radio wavelengths at the sub-arcsecond level.     Chasing the identification of ASCA Galactic Objects (ChIcAGO): An X-ray survey of unidentified sources in the galactic plane. I : Source sample and initial results

    We present the Chasing the Identification of ASCA Galactic Objects (ChIcAGO) survey, which is designed to identify the unknown X-ray sources discovered during the ASCA Galactic Plane Survey (AGPS). Little is known about most of the AGPS sources, especially those that emit primarily in hard X-rays (2-10 keV) within the Fx 10-13 to 10-11 erg cm -2 s-1 X-ray flux range. In ChIcAGO, the subarcsecond localization capabilities of Chandra have been combined with a detailed multiwavelength follow-up program, with the ultimate goal of classifying the >100 unidentified sources in the AGPS. Overall to date, 93 unidentified AGPS sources have been observed with Chandra as part of the ChIcAGO survey. A total of 253 X-ray point sources have been detected in these Chandra observations within 3′ of the original ASCA positions. We have identified infrared and optical counterparts to the majority of these sources, using both new observations and catalogs from existing Galactic plane surveys. X-ray and infrared population statistics for the X-ray point sources detected in the Chandra observations reveal that the primary populations of Galactic plane X-ray sources that emit in the Fx 10-13 to 10-11 erg cm -2 s-1 flux range are active stellar coronae, massive stars with strong stellar winds that are possibly in colliding wind binaries, X-ray binaries, and magnetars. There is also another primary population that is still unidentified but, on the basis of its X-ray and infrared properties, likely comprises partly Galactic sources and partly active galactic nuclei.Peer reviewedSubmitted Versio

    Youth futures and a masculine development ethos in the regional story of Uttarakhand

    Research on the Uttarakhand region, which became a new state in 2000, has focused largely on agrarian livelihoods, religious rituals, development demands, ecological politics and the role of women in regional social movements. This essay discusses another dimension of the regional imaginary—that of a masculine development ethos. Based on ethnographic research and print media sources, this essay focuses on stories, politics, mobilities and imaginations of young men in the years immediately after the achievement of statehood. Despite increased outmigration of youth in search of employment, many young men expressed the dream of maintaining livelihoods in the familiar towns and rural spaces of Uttarakhand, describing their home region as a source of power and agency. In rallies and in print media, young (mostly upper caste) men expressed their disillusionment with the government and the promises of statehood, arguing that their aspirations for development and employment were left unfulfilled. Gendered stories of the region, told in Hindi in rallies and print media, contained references to local places, people and historical events and were produced through local connections and know-how, fostering a regional youth politics. The article argues that Uttarakhand as a region is shaped by the politics of local actors as well as embodied forms of aspiration, affiliation and mobility.IS

    Adsorption of mono- and multivalent cat- and anions on DNA molecules

    Adsorption of monovalent and multivalent cat- and anions on a deoxyribose nucleic acid (DNA) molecule from a salt solution is investigated by computer simulation. The ions are modelled as charged hard spheres, the DNA molecule as a point charge pattern following the double-helical phosphate strands. The geometrical shape of the DNA molecules is modelled on different levels ranging from a simple cylindrical shape to structured models which include the major and minor grooves between the phosphate strands. The densities of the ions adsorbed on the phosphate strands, in the major and in the minor grooves are calculated. First, we find that the adsorption pattern on the DNA surface depends strongly on its geometrical shape: counterions adsorb preferentially along the phosphate strands for a cylindrical model shape, but in the minor groove for a geometrically structured model. Second, we find that an addition of monovalent salt ions results in an increase of the charge density in the minor groove while the total charge density of ions adsorbed in the major groove stays unchanged. The adsorbed ion densities are highly structured along the minor groove while they are almost smeared along the major groove. Furthermore, for a fixed amount of added salt, the major groove cationic charge is independent on the counterion valency. For increasing salt concentration the major groove is neutralized while the total charge adsorbed in the minor groove is constant. DNA overcharging is detected for multivalent salt. Simulations for a larger ion radii, which mimic the effect of the ion hydration, indicate an increased adsorbtion of cations in the major groove.Comment: 34 pages with 14 figure

    Prospects for Studies of Stellar Evolution and Stellar Death in the JWST Era

    I review the prospects for studies of the advanced evolutionary stages of low-, intermediate- and high-mass stars by the JWST and concurrent facilities, with particular emphasis on how they may help elucidate the dominant contributors to the interstellar dust component of galaxies. Observations extending from the mid-infrared to the submillimeter can help quantify the heavy element and dust species inputs to galaxies from AGB stars. JWST's MIRI mid-infrared instrument will be so sensitive that observations of the dust emission from individual intergalactic AGB stars and planetary nebulae in the Virgo Cluster will be feasible. The Herschel Space Observatory will enable the last largely unexplored spectral region, the far-IR to the submillimeter, to be surveyed for new lines and dust features, while SOFIA will cover the wavelength gap between JWST and Herschel, a spectral region containing important fine structure lines, together with key water-ice and crystalline silicate bands. Spitzer has significantly increased the number of Type II supernovae that have been surveyed for early-epoch dust formation but reliable quantification of the dust contributions from massive star supernovae of Type II, Type Ib and Type Ic to low- and high-redshift galaxies should come from JWST MIRI observations, which will be able to probe a volume over 1000 times larger than Spitzer.Comment: 24 pages, 19 figures.     The velocity potential and the interacting force for two spheres moving perpendicularly to the line joining their centers

    The velocity potential around two spheres moving perpendicularly to the line joining their centers is given by a series of spherical harmonics. The appropriateness of the truncation is evaluated by determining the residual normal surface velocity on the spheres. In evaluating the residual normal velocity, a recursive procedure is constructed to evaluate the spherical harmonics to reduce computational effort and truncation error as compared to direct transformation or numerical integration.
