    BLOC-2 subunit HPS6 deficiency affects the tubulation and secretion of von Willebrand factor from mouse endothelial cells

    Hermansky-Pudlak syndrome (HPS) is a recessive disorder with bleeding diathesis, which has been linked to platelet granule defects. Both platelet granules and endothelial Weibel-Palade bodies (WPBs) are members of lysosome-related organelles (LROs) whose formation is regulated by HPS protein associated complexes such as BLOC (biogenesis of lysosome organelles complex) -1, -2, -3, AP-3 (adaptor protein complex-3) and HOPS (homotypic fusion and protein sorting complex). Von Willebrand factor (VWF) is critical to hemostasis, which is stored in a highly-multimerized form as tubules in the WPBs. In this study, we found the defective, but varying, release of VWF into plasma after desmopressin (DDAVP) stimulation in HPS1 (BLOC-3 subunit), HPS6 (BLOC-2 subunit), and HPS9 (BLOC-1 subunit) deficient mice. In particular, VWF tubulation, a critical step in VWF maturation, was impaired in HPS6 deficient WPBs. This likely reflects a defective endothelium, contributing to the bleeding tendency in HPS mice or patients. The differentially defective regulated release of VWF in these HPS mouse models suggests the need for precise HPS genotyping before DDAVP administration to HPS patients

    miR-10a is aberrantly overexpressed in Nucleophosmin1 mutated acute myeloid leukaemia and its suppression induces cell death

    <p>Abstract</p> <p>Background</p> <p>Acute myeloid leukaemia (AML) with nucleophosmin-1 (<it>NPM1</it>) mutation is a major subtype of AML. The <it>NPM1 </it>mutation induces a myeloproliferative disorder, but evidence indicates that other insults are necessary for the development of AML. We utilised microRNA microarrays and functional assays to determine if microRNA dysregulation could be involved in the pathogenesis of in <it>NPM1 </it>mutated (<it>NPM1<sup>mut</sup></it>)-AML.</p> <p>Results</p> <p>We used a stringent locked nucleic acid (LNA) based microRNA microarray platform to profile bone marrow samples of patients with normal karyotype AML. A panel of five microRNAs dichotomised AML patients according to their <it>NPM1 </it>mutational status. miR-10a, let-7b and let-7c were significantly over-expressed, while miR-130a and miR-335 were under-expressed in <it>NPM1<sup>mut</sup></it>-AML when compared to <it>NPM1<sup>wildtype</sup></it>-AML. Of these, miR-10a is the most differentially expressed in <it>NPM1<sup>mut</sup></it>-AML versus <it>NPM1<sup>wildtype</sup></it>-AML (> 10 fold higher as confirmed by qRT-PCR). To investigate the functions of miR-10a, the OCI-AML3 cell line was utilised, which is the only commercially available cell line bearing <it>NPM1<sup>mut</sup></it>. OCI-AML3 cells were firstly demonstrated to have a similarly high miR-10a expression to primary <it>NPM1<sup>mut</sup></it>-AML patient samples. Inhibition of miR-10a expression by miRCURY LNA Inhibitors (Exiqon) in these cells resulted in increased cell death as assessed by MTS, cell cycle and Annexin-V assays and reduced clonogenic capacity, indicative of an involvement in leukaemic cell survival. <it>In silico </it>filtering of bioinformatically predicted targets of miR-10a identified a number of potential mRNA targets with annotated functions in haematopoiesis, cell growth and apoptosis. Lucferase reporter assays confirmed a number of these putative tumorogenic genes that are miR-10a suppressible including <it>KLF4 </it>and <it>RB1CC1</it>. This provides a potential mechanism for the pathogenic role of miR-10a in <it>NPM1<sup>mut</sup></it>-AML.</p> <p>Conclusions</p> <p>This study provides, for the first time, <it>in vitro </it>evidence of a pro-survival role of miR-10a in <it>NPM1<sup>mut</sup></it>-AML, that it may contribute to the pathogenesis of <it>NPM1<sup>mut</sup></it>-AML and identifies putative tumorogenic targets.</p

    Decoder Hardware Architecture for HEVC

    This chapter provides an overview of the design challenges faced in the implementation of hardware HEVC decoders. These challenges can be attributed to the larger and diverse coding block sizes and transform sizes, the larger interpolation filter for motion compensation, the increased number of steps in intra prediction and the introduction of a new in-loop filter. Several solutions to address these implementation challenges are discussed. As a reference, results for an HEVC decoder test chip are also presented.Texas Instruments Incorporate

    An integrated atom-photon junction

    Photonic chips that integrate guides, switches, gratings and other components, process vast amounts of information rapidly on a single device. A new branch of this technology becomes possible if the light is coupled to cold atoms in a junction of small enough cross section, so that small numbers of photons interact appreciably with the atoms. Cold atoms are among the most sensitive of metrological tools and their quantum nature also provides a basis for new information processing methods. Here we demonstrate a photonic chip which provides multiple microscopic junctions between atoms and photons. We use the absorption of light at a junction to reveal the presence of one atom on average. Conversely, we use the atoms to probe the intensity and polarisation of the light. Our device paves the way for a new type of chip with interconnected circuits of atoms and photons.Comment: 5 pages, 4 figure. Submitted to Nature Photonic

    Comparison of Sentiment Analysis and User Ratings in Venue Recommendation

    Venue recommendation aims to provide users with venues to visit, taking into account historical visits to venues. Many venue recommendation approaches make use of the provided users’ ratings to elicit the users’ preferences on the venues when making recommendations. In fact, many also consider the users’ ratings as the ground truth for assessing their recommendation performance. However, users are often reported to exhibit inconsistent rating behaviour, leading to less accurate preferences information being collected for the recommendation task. To alleviate this problem, we consider instead the use of the sentiment information collected from comments posted by the users on the venues as a surrogate to the users’ ratings. We experiment with various sentiment analysis classifiers, including the recent neural networks-based sentiment analysers, to examine the effectiveness of replacing users’ ratings with sentiment information. We integrate the sentiment information into the widely used matrix factorization and GeoSoCa multi feature-based venue recommendation models, thereby replacing the users’ ratings with the obtained sentiment scores. Our results, using three Yelp Challenge-based datasets, show that it is indeed possible to effectively replace users’ ratings with sentiment scores when state-of-the-art sentiment classifiers are used. Our findings show that the sentiment scores can provide accurate user preferences information, thereby increasing the prediction accuracy. In addition, our results suggest that a simple binary rating with ‘like’ and ‘dislike’ is a sufficient substitute of the current used multi-rating scales for venue recommendation in location-based social networks

    Structure and mechanism of human DNA polymerase η

    The variant form of the human syndrome xeroderma pigmentosum (XPV) is caused by a deficiency in DNA polymerase eta (Pol eta), a DNA polymerase that enables replication through ultraviolet-induced pyrimidine dimers. Here we report high-resolution crystal structures of human Pol eta at four consecutive steps during DNA synthesis through cis-syn cyclobutane thymine dimers. Pol eta acts like a 'molecular splint' to stabilize damaged DNA in a normal B-form conformation. An enlarged active site accommodates the thymine dimer with excellent stereochemistry for two-metal ion catalysis. Two residues conserved among Pol eta orthologues form specific hydrogen bonds with the lesion and the incoming nucleotide to assist translesion synthesis. On the basis of the structures, eight Pol eta missense mutations causing XPV can be rationalized as undermining the molecular splint or perturbing the active-site alignment. The structures also provide an insight into the role of Pol eta in replicating through D loop and DNA fragile sites

    EQ-5D in skin conditions: an assessment of validity and responsiveness

    Aims and objectives This systematic literature review aims to assess the reliability, validity and responsiveness of three widely used generic preference-based measures of health-related quality of life (HRQL), i.e., EQ-5D, Health Utility Index 3 (HUI3) and SF-6D in patients with skin conditions. Methods A systematic search was conducted to identify studies reporting health state utility values obtained using EQ-5D, SF-6D, or HUI3 alongside other HRQL measures or clinical indices for patients with skin conditions. Data on test-retest analysis for reliability, known group differences or correlation and regression analyses for validity, and change over time or responsiveness indices analysis were extracted and reviewed. Results A total of 16 papers reporting EQ-5D utilities in people with skin conditions were included in the final review. No papers for SF-6D and HUI3 were found. Evidence of reliability was not found for any of these measures. The majority of studies included in the review (12 out of 16) examined patients with plaque psoriasis or psoriatic arthritis and the remaining four studies examined patients with either acne, hidradenitis suppurativa, hand eczema, or venous leg ulcers. The findings were generally positive in terms of performance of EQ-5D. Six studies showed that EQ-5D was able to reflect differences between severity groups and only one reported differences that were not statistically significant. Four studies found that EQ-5D detected differences between patients and the general population, and differences were statistically different for three of them. Further, moderate-to-strong correlation coefficients were found between EQ-5D and other skin-specific HRQL measures in four studies. Eight studies showed that EQ-5D was able to detect change in HRQL appropriately over time and the changes were statistically significant in seven studies. Conclusions Overall, the validity and responsiveness of the EQ-5D was found to be good in people with skin diseases, especially plaque psoriasis or psoriatic arthritis. No evidence on SF-6D and HUI3 was available to enable any judgments to be made on their performance
