    Single Higgs boson production at future linear colliders including radiative corrections

    The next generation of high energy e+ e- linear colliders is expected to operate at \sqrt{s} \gtsim 500 GeV. In this energy range the WW fusion channel dominates the Higgs boson production cross section e+ e- -> \bar\nu \nu h^0/H^0. We calculate the one-loop corrections to this process due to fermion and sfermion loops within the MSSM. We perform a detailed numerical analysis of the total cross section and the distributions of the rapidity, the transverse momentum and the production angle of the Higgs boson. The fermion-sfermion correction is substantial being of the order of -10% and is dominated by the fermion loops. In addition, we explore the possibility of polarized e+ / e- beams. In the so-called "intense coupling" scenario the production of the heavy Higgs boson H^0 is also discussed.Comment: 23 pages, 9 figures, to be published in Nucl. Phys.

    Estudio de la biopelícula formada por dos especies de pseudomonas asociadas a plantas

    Las bacterias pertenecientes al género Pseudomonas constituyen un modelo de estudio de la interacción microorganismo-planta. Pseudomonas syringae es una especie que produce enfermedades en la parte aérea de algunas plantas. Por otro lado, Pseudomonas chlororaphis interacciona con la raíz y ha demostrado ser un agente de control biológico. Ambas especies forman biopelículas y, a pesar de su diferente estilo de vida, análisis in silico han puesto de manifiesto la existencia de regiones genómicas tanto comunes como diferenciales que parecen estar implicadas en la biosíntesis de matriz extracelular. En este trabajo se han construido mutantes de estas regiones y analizado su implicación en la formación de biopelícula y aspectos relacionados.Universidad de Málaga.     A Psl-like polysaccharide has a key role in the biofilm architecture of two plant-associated Pseudomonas

    Comunicación congresoThe Psl polysaccharide has only been studied in Pseudomonas aeruginosa, and its role in bacteria that interact with plants is still unknown. On the one hand, P. syringae is a model of study of plant-pathogen interactions. On the other hand, P. chlororaphis is a biocontrol agent of plant-fungal diseases, as the white root rot caused by Rosellinia necatrix. In silico analysis have let us identify in both species a genomic region which is ortholog to the Psl-encoding region of P. aeruginosa PAO1. We constructed mutants of this region in both strains and analysed its phenotype in biofilm formation, both in static microwell plates and dynamic flow-cell chamber experiments. The results revealed a different and an important role of this polysaccharide in the biofilm architecture of both strains. Furthermore, in P. syringae this Psl-like polysaccharide has a very important role in swarming motility. All these striking phenotypes led us further study the implication of this polysaccharide in the lifestyles of both models of plant-bacteria interaction.Universidad de Málaga.     Human neutrophil clearance of bacterial pathogens triggers anti-microbial gamma delta T cell responses in early infection

    Human blood Vc9/Vd2 T cells, monocytes and neutrophils share a responsiveness toward inflammatory chemokines and are rapidly recruited to sites of infection. Studying their interaction in vitro and relating these findings to in vivo observations in patients may therefore provide crucial insight into inflammatory events. Our present data demonstrate that Vc9/Vd2 T cells provide potent survival signals resulting in neutrophil activation and the release of the neutrophil chemoattractant CXCL8 (IL-8). In turn, Vc9/Vd2 T cells readily respond to neutrophils harboring phagocytosed bacteria, as evidenced by expression of CD69, interferon (IFN)-c and tumor necrosis factor (TNF)-a. This response is dependent on the ability of these bacteria to produce the microbial metabolite (E)-4-hydroxy-3-methyl-but-2-enyl pyrophosphate (HMB-PP), requires cell-cell contact of Vc9/Vd2 T cells with accessory monocytes through lymphocyte function-associated antigen-1 (LFA-1), and results in a TNF-a dependent proliferation of Vc9/Vd2 T cells. The antibiotic fosmidomycin, which targets the HMB-PP biosynthesis pathway, not only has a direct antibacterial effect on most HMB-PP producing bacteria but also possesses rapid anti-inflammatory properties by inhibiting cd T cell responses in vitro. Patients with acute peritoneal-dialysis (PD)-associated bacterial peritonitis – characterized by an excessive influx of neutrophils and monocytes into the peritoneal cavity – show a selective activation of local Vc9/Vd2 T cells by HMB-PP producing but not by HMB-PP deficient bacterial pathogens. The cd T celldriven perpetuation of inflammatory responses during acute peritonitis is associated with elevated peritoneal levels of cd T cells and TNF-a and detrimental clinical outcomes in infections caused by HMB-PP positive microorganisms. Taken together, our findings indicate a direct link between invading pathogens, neutrophils, monocytes and microbe-responsive cd T cells in early infection and suggest novel diagnostic and therapeutic approaches.Martin S.     Sfermion Pair Production in Polarized and Unpolarized γγ\gamma\gamma Collisions

    We calculate total and differential cross sections for the production of sfermion pairs in photon-photon collisions, including contributions from resolved photons and arbitrary photon polarization. Sfermion production in photon collisions depends only on the sfermion mass and charge. It is thus independent of the details of the SUSY breaking mechanism, but highly sensitive to the sfermion charge. We compare the total cross sections for bremsstrahlung, beamstrahlung, and laser backscattering photons to those in e+ee^+e^- annihilation. We find that the total cross section at a polarized photon collider is larger than the e+ee^+e^- annihilation cross section up to the kinematic limit of the photon collider.Comment: 19 pages, Latex, 18 (e)ps-figure

    Cellular and humoral immune responses and protection against schistosomes induced by a radiation-attenuated vaccine in chimpanzees

    The radiation-attenuated Schistosoma mansoni vaccine is highly effective in rodents and primates but has never been tested in humans, primarily for safety reasons. To strengthen its status as a paradigm for a human recombinant antigen vaccine, we have undertaken a small-scale vaccination and challenge experiment in chimpanzees (Pan troglodytes). Immunological, clinical, and parasitological parameters were measured in three animals after multiple vaccinations, together with three controls, during the acute and chronic stages of challenge infection up to chemotherapeutic cure. Vaccination induced a strong in vitro proliferative response and early gamma interferon production, but type 2 cytokines were dominant by the time of challenge. The controls showed little response to challenge infection before the acute stage of the disease, initiated by egg deposition. In contrast, the responses of vaccinated animals were muted throughout the challenge period. Vaccination also induced parasite-specific immunoglobulin M (IgM) and IgG, which reached high levels at the time of challenge, while in control animals levels did not rise markedly before egg deposition. The protective effects of vaccination were manifested as an amelioration of acute disease and overall morbidity, revealed by differences in gamma-glutamyl transferase level, leukocytosis, eosinophilia, and hematocrit. Moreover, vaccinated chimpanzees had a 46% lower level of circulating cathodic antigen and a 38% reduction in fecal egg output, compared to controls, during the chronic phase of infection

    WMAPing out Supersymmetric Dark Matter and Phenomenology

    The recent WMAP data provide a rather restricted range of the Cold Dark Matter (CDM) density ΩCDMh2 \Omega_{CDM} h^2 of unprecedented accuracy. We combine these new data along with data from BNL E821 experiment measuring (gμ2) {(g_{\mu}-2)}, the {b\goes s \gamma} branching ratio and the light Higgs boson mass bound from LEP, to update our analysis of the allowed boundaries in the parameter space of the Constrained Minimal Supersymmetric Standard Model (CMSSM). The prospects of measuring Supersymmetry at LHC look like a very safe bet, and the potential of discovering SUSY particles at a s=1.1TeV \sqrt{s} = 1.1 \mathrm{TeV} linear collider is enhanced considerably.     NirA Is an Alternative Nitrite Reductase from Pseudomonas aeruginosa with Potential as an Antivirulence Target.

    The opportunistic pathogen Pseudomonas aeruginosa produces an arsenal of virulence factors causing a wide range of diseases in multiple hosts and is difficult to eradicate due to its intrinsic resistance to antibiotics. With the antibacterial pipeline drying up, antivirulence therapy has become an attractive alternative strategy to the traditional use of antibiotics to treat P. aeruginosa infections. To identify P. aeruginosa genes required for virulence in multiple hosts, a random library of Tn5 mutants in strain PAO1-L was previously screened in vitro for those showing pleiotropic effects in the production of virulence phenotypes. Using this strategy, we identified a Tn5 mutant with an insertion in PA4130 showing reduced levels of a number of virulence traits in vitro Construction of an isogenic mutant in this gene presented results similar to those for the Tn5 mutant. Furthermore, the PA4130 isogenic mutant showed substantial attenuation in disease models of Drosophila melanogaster and Caenorhabditis elegans as well as reduced toxicity in human cell lines. Mice infected with this mutant demonstrated an 80% increased survival rate in acute and agar bead lung infection models. PA4130 codes for a protein with homology to nitrite and sulfite reductases. Overexpression of PA4130 in the presence of the siroheme synthase CysG enabled its purification as a soluble protein. Methyl viologen oxidation assays with purified PA4130 showed that this enzyme is a nitrite reductase operating in a ferredoxin-dependent manner. The preference for nitrite and production of ammonium revealed that PA4130 is an ammonia:ferredoxin nitrite reductase and hence was named NirA.IMPORTANCE The emergence of widespread antimicrobial resistance has led to the need for development of novel therapeutic interventions. Antivirulence strategies are an attractive alternative to classic antimicrobial therapy; however, they require identification of new specific targets which can be exploited in drug discovery programs. The host-specific nature of P. aeruginosa virulence adds complexity to the discovery of these types of targets. Using a sequence of in vitro assays and phylogenetically diverse in vivo disease models, we have identified a PA4130 mutant with reduced production in a number of virulence traits and severe attenuation across all infection models tested. Characterization of PA4130 revealed that it is a ferredoxin-nitrite reductase and hence was named NirA. These results, together with attenuation of nirA mutants in different clinical isolates, high level conservation of its gene product in P. aeruginosa genomes, and the lack of orthologues in human genomes, make NirA an attractive antivirulence target

    Extinction of cue-evoked drug-seeking relies on degrading hierarchical instrumental expectancies

    There has long been need for a behavioural intervention that attenuates cue-evoked drug-seeking, but the optimal method remains obscure. To address this, we report three approaches to extinguish cue-evoked drug-seeking measured in a Pavlovian to instrumental transfer design, in non-treatment seeking adult smokers and alcohol drinkers. The results showed that the ability of a drug stimulus to transfer control over a separately trained drug-seeking response was not affected by the stimulus undergoing Pavlovian extinction training in experiment 1, but was abolished by the stimulus undergoing discriminative extinction training in experiment 2, and was abolished by explicit verbal instructions stating that the stimulus did not signal a more effective response-drug contingency in experiment 3. These data suggest that cue-evoked drug-seeking is mediated by a propositional hierarchical instrumental expectancy that the drug-seeking response is more likely to be rewarded in that stimulus. Methods which degraded this hierarchical expectancy were effective in the laboratory, and so may have therapeutic potential
