Avaliação da eficácia do aluminosilicato de sódio hidratado na prevenção de câncer hepático induzido por aflatoxina em truta arco-íris
The use of aluminum silicates for decontaminating animal feed containing aflatoxins has yielded encouraging results in chicken and turkey poults. In contrast, very few studies have tested these substances in aquaculture. In this work, we investigated the efficacy of a trout diet containing 0.5% hydrated sodium aluminosilicate (HSAS) in protecting against contamination with aflatoxin B1. Trout were reared on these diets for one year and the experimental groups were examined monthly for hepatic presumptive preneoplastic and neoplastic lesions. Regardless of the presence of HSAS, all of the fish that received aflatoxin in their diet have shown hepatic lesions indicative of a carcinogenic process, presenting also the development of cancer in some fish. The concentration of HSAS used in this study was ineffective in preventing the onset of hepatic lesions induced by aflatoxin B1 in rainbow trout.Resultados encorajadores têm sido observados com o emprego de aluminosilicatos para descontaminação de ração contendo aflatoxina destinada à avicultura. No entanto, raros estudos têm sido destinados a testar essa substância em aqüicultura. Assim, no presente trabalho, foi investigada a eficácia do aluminosilicato de sódio hidratado (HSAS, 0,5%) em proteger a truta arco-íris dos danos hepáticos causados por ração contaminada com aflatoxina B1. Os grupos experimentais foram alimentados com suas respectivas dietas durante 12 meses, mensalmente cada grupo foi amostrado para análises anatomopatológica e histopatológica para verificação da presença de lesões hepáticas. Apesar da presença de HSAS, todos os grupos que receberam dieta contaminada com aflatoxina B1 apresentaram lesões hepáticas indicativas do processo carcinogênico, sendo que exemplares alimentados com essas dietas por mais longo prazo apresentaram hepatocarcinoma ou colangiocarcinoma. Histological changes caused by experimental Riedeliella graciliflora (Leg. Papilionoideae) poisoning in cattle and laboratory animals Alterações histológicas na intoxicação experimental por Riedeliella graciliflora (Leg. Papilionoideae) em bovinos e animais de laboratório
Tissues from cattle, mice, rats and guinea pigs experimentally intoxicated by Riedeliella graciliflora were studied histologically. Cattle lymph nodes, spleen, Peyer patches and peribronchial lymphoid tissues had diffuse necrosis of lymphocytes, mainly in the germinal centers of the follicles. This lesion was less severe in laboratory animals. All species had severe enteritis with infiltration of the lamina propria by mononuclear cells. Some cells in this infiltrate were necrotic. Degeneration and necrosis of the epithelial cells, mainly in the tip of the villi, and detachment of the epithelial lining from the lamina propria were also observed. In the liver the trabecular structure was disrupted and the hepatocytes had some degree of individual necrosis and degeneration. A tubular nephrosis was observed in the kidneys. Liver, lung, kidney, intestine and lymph nodes had different degrees of congestion. Those lesions are similar than those caused by Polygala klotzschii, a plant that contains 5-metoxi-podophyllotoxin.Foi realizado um estudo histológico de bovinos, camundongos, ratos e coelhos intoxicados experimentalmente por Riedeliella graciliflora. Em bovinos os gânglios linfáticos, baço, placas de Peyer e tecido peribronquial apresentavam necrose do tecido linfático, afetando, principalmente, os centros germinativos dos folículos. Esta lesão foi menos severa em animais de laboratório. Em todas as espécies havia severa enterite e infiltração da lâmina propria do intestino por células mononucleares. Algumas destas células estavam necróticas. Degeneração e necrose do epitélio, principalmente na superfície das vilosidades, e descamação das células epiteliais foram, também, observadas. No fígado havia desorganização da estrutura trabecular e alguns hepatócitos apresentavam-se degenerados ou necróticos. Os rins apresentavam nefrose tubular. Diferentes graus de congestão foram observados no fígado, pulmão, rins, intestinos e linfonodos. As lesões observadas são similares às causadas por Polygala klotzschii, uma planta que contém 5-metoxi-podofilotoxina
Extramedullary plasmacytoma of the third eyelid gland in a dog
A case of extramedullary plasmacytoma of the third eyelid gland in a 7-year-old American Cocker Spaniel is reported. An enlargement of the third eyelid gland, abundant mucopurulent discharge, mild hyperemia and corneal pigmentation in the OD was present. Excisional biopsy of the mass revealed the gland was infiltrated and partially destroyed by a uniform population of neoplastic plasma cells. the neoplastic cells were positive for CD138, Ki-67 and lambda light chain. CD20, CD3, kappa light chain and cytokeratin were negative. Twelve months following surgery, no recurrence was observed. Transient disruption of liver gap junctional intercellular communication and induction of apoptosis after administration of 1,4-bis[2-(3,5 dichloropyridyloxy)]benzene in mice
Gap junctional intercellular communication (GJIC) and connexin expression (Cx26 and Cx32) in mouse liver were studied after administration of 4-bis[2-(3,5 dichloropyridyloxy)]benzene (TCPOBOP), a phenobarbital-like enzyme inducer. Female C57BI/6 mice were administered TCPOBOP (5.8 mg/kg BW) and euthanized 0, 24, 48 and 72 hours later. Liver samples were snap frozen, or fixed in formalin, or submitted to GJIC analysis. The proliferating cell nuclear antigen (PCNA) immunohistochemistry and the Western blotting for Cx26 and Cx32 were performed. After 48 and 72 h of drug administration the liver-to-body weight ratio was increased 70% and 117% (p < 0.0001), respectively. There were temporal-dependent alterations in liver histopathology and a significant increase in cell proliferation was noted after 48h and sustained after 72h, though to a lesser extent (p < 0.0001). In addition. TCPOBOP administration induced apoptosis, which appeared to be time-dependent showing statistical significance only after 72h (p < 0.0001). Interestingly, a transient disruption by nearly 50% of GJIC capacity was detected after 48 h of drug ingestion, which recovered after 72 h (p = 0.003). These GJIC changes were due to altered levels of Cx26 and Cx32 in the livers of TCPOBOP-treated mice. We concluded that a single administration of TCPOBOP transiently disrupted the levels of GJIC due to decreased expression of connexins and increased apoptotic cell death in mouse liver.
