    Asymmetric wall ingrowth deposition in Arabidopsis phloem parenchyma transfer cells is tightly associated with sieve elements

    In Arabidopsis, polarized deposition of wall ingrowths in phloem parenchyma (PP) transfer cells (TCs) occurs adjacent to cells of the sieve element/companion cell (SE/CC) complex. However, the spatial relationships between these different cell types in minor veins, where phloem loading occurs, are poorly understood. PP TC development and wall ingrowth localization were compared with those of other phloem cells in leaves of Col-0 and the transgenic lines AtSUC2::AtSTP9-GFP (green fluorescent protein) and AtSWEET11::AtSWEET11-GFP that identify CCs and PP cells, respectively. The development of PP TCs in minor veins, indicated by deposition of wall ingrowths, proceeded basipetally in leaves. However, not all PP cells develop wall ingrowths, and higher levels of deposition occur in abaxial- compared with adaxial-positioned PP TCs. Furthermore, the deposition of wall ingrowths was exclusively initiated on and preferentially covered the PP TC/SE interface, rather than the PP TC/CC interface, and only occurred in PP cells that were adjacent to SEs. Collectively, these results demonstrate a tight association between SEs and wall ingrowth deposition in PP TCs and suggest the existence of two subtypes of PP cells in leaf minor veins. Compared with PP cells, PP TCs showed more abundant accumulation of AtSWEET11–GFP, indicating functional differences in phloem loading between PP and PP TCs

Laser action at 1080nm was obtained in a 22mm long gallium lanthanum sulphide glass fibre with a neodymium doped core, fabricated by the rod-in-tube technique.     Host-directed therapy, an untapped opportunity for antimalarial intervention

    Host-directed therapy (HDT) is gaining traction as a strategy to combat infectious diseases caused by viruses and intracellular bacteria, but its implementation in the context of parasitic diseases has received less attention. Here, we provide a brief overview of this field and advocate HDT as a promising strategy for antimalarial intervention based on untapped targets. HDT provides a basis from which repurposed drugs could be rapidly deployed and is likely to strongly limit the emergence of resistance. This strategy can be applied to any intracellular pathogen and is particularly well placed in situations in which rapid identification of treatments is needed, such as emerging infections and pandemics, as starkly illustrated by the current COVID-19 crisis.Ling Wei, Jack Adderley, Didier Leroy, David H.     The Freidlin-Wentzell large deviation principle is established for the distributions of stochastic evolution equations with general monotone drift and small multiplicative noise. As examples, the main results are applied to derive the large deviation principle for different types of SPDE such as stochastic reaction-diffusion equations, stochastic porous media equations and fast diffusion equations, and the stochastic p-Laplace equation in Hilbert space.     Mn clusterisation in Ga1-xMnxN

    Local structure of Mn atoms in Ga1-xMnxN has been investigated by the Mn L3 edge x-ray absorption spectrum (XAS) at total electron yield mode, which preferentially looks at atoms near the surface. A modeling defects configuration, the Mn5 micro-clusters complexed with substitutional MnGa and interstitial MnI is found for the higher Mn doping concentration.     Experimental models that consider host-pathogen interactions are relevant for improving knowledge about oral candidiasis. The aim of this study was to assess the epithelial immune responses, Candida penetration of cell monolayers, and virulence during mixed species culture infections. Single species cultures of Candida albicans and mixed cultures (C. albicans, Streptococcus mutans, and Streptococcus sanguinis) were used to infect monolayers of HaCaT and FaDu ATCC HTB-43 cells for 12 h. After infection, IL-18 and IL-34 gene expression was measured to assess epithelial cell immune responses, and lactate dehydrogenase (LDH) activity was measured as an indicator of cell damage. Microscopy determined C. albicans morphology and penetration of fungal cells through the keratinocyte monolayer. Monolayers devoid of infection served as controls. Data were analyzed by an ANOVA one-way test followed by Tukey’s post-hoc test (α = 0.05). The results found that IL-18 and IL-34 gene expression and LDH activity were significantly (p < 0.05) upregulated for both cell lines exposed to mixed species cultures compared with C. albicans alone. Candida albicans yeast and hyphae were evident in C. albicans only infections. In contrast, monolayers infected by C. albicans, S. mutans, and S. sanguinis exhibited higher microbial invasion with several hyphal aggregates detected. The presence of streptococci in C. albicans infection enhances the virulence and pathogenicity of the fungus with associated increased immune responses and tissue damage. Extrapolation of these findings to oral infection would indicate the added potential benefit of managing bacterial components of biofilms during treatment

    Flavor Changing Neutral Currents involving Heavy Quarks with Four Generations

    We study various FCNC involving heavy quarks in the Standard Model (SM) with a sequential fourth generation. After imposing BXsγB\to X_s\gamma, BXsl+lB\to X_sl^+l^- and ZbbˉZ\to b\bar{b} constraints, we find B(Zsbˉ+sˉb){\cal B}(Z\to s\bar{b}+\bar{s}b) can be enhanced by an order of magnitude to 10710^{-7}, while tcZ,cHt\to cZ, cH decays can reach 10610^{-6}, which are orders of magnitude higher than in SM. However,these rates are still not observable for the near future.With the era of LHC approaching, we focus on FCNC decays involving fourth generation bb^\prime and tt^\prime quarks. We calculate the rates for loop induced FCNC decays bbZ,bH,bg,bγb^\prime\to bZ, bH, bg, b\gamma, as well as t^\prime\to tZ,\tH, tg, t\gamma. If Vcb|V_{cb'}| is of order Vcb0.04|V_{cb}| \simeq 0.04, tree level bcWb^\prime\to cW decay would dominate, posing a challenge since bb-tagging is less effective. For VcbVcb|V_{cb'}| \ll |V_{cb}|, btWb'\to tW would tend to dominate, while btWb'\to t^\prime W^* could also open for heavier bb', leading to thepossibility of quadruple-WW signals via bbˉbbˉW+WW+Wb'\bar b'\to b\bar b W^+W^-W^+W^-. The FCNC bbZ,bHb'\to bZ, bH decays could still dominate if mbm_{b'} is just above 200 GeV. For the case of tt', ingeneral tbWt^\prime\to bW would be dominant, hence it behaves like a heavy top. For both bb' and tt', except for the intriguing light bb' case, FCNC decays are in the 10410210^{-4} -10^{-2} range, and are quite detectable at the LHC.For a possible future ILC, we find the associated production of FCNC e+ebsˉe^+e^-\to b\bar s, tcˉt\bar c are below sensitivity, while e+ebbˉe^+e^-\to b^\prime\bar b andttˉt^\prime\bar t can be better probed.Tevatron Run-II can still probe the lighter bb' or tt' scenario. LHC would either discover the fourth generation and measure the FCNC rates, or rule out the fourth generation conclusively.Comment: 31 pages, 15 eps figures, version to appear in JHE

    HOPX functions as a tumour suppressor in head and neck cancer.

    Head and neck squamous cell carcinoma (HNSCC) is generalized term that encompasses a diverse group of cancers that includes tumours of the oral cavity (OSCC), oropharynx (OPSCC) and nasopharynx (NPC). Genetic alterations that are common to all HNSCC types are likely to be important for squamous carcinogenesis. In this study, we have investigated the role of the homeodomain-only homeobox gene, HOPX, in the pathogenesis of HNSCC. We show that HOPX mRNA levels are reduced in OSCC and NPC cell lines and tissues and there is a general reduction of HOPX protein expression in these tumours and OPSCCs. HOPX promoter methylation was observed in a subset of HNSCCs and was associated with a worse overall survival in HPV negative tumours. RNAseq analysis of OSCC cells transfected with HOPX revealed a widespread deregulation of the transcription of genes related to epithelial homeostasis and ectopic over-expression of HOPX in OSCC and NPC cells inhibited cell proliferation, plating efficiency and migration, and enhanced sensitivity to UVA-induced apoptosis. Our results demonstrate that HOPX functions as a tumour suppressor in HNSCC and suggest a central role for HOPX in suppressing epithelial carcinogenesis

    Proximity induced metal/insulator transition in YBa2Cu3O7/La2/3Ca1/3MnO3Y Ba_2 Cu_3 O_7 / La_{2/3} Ca_{1/3} Mn O_3 superlattices

    The far-infrared dielectric response of superlattices (SL) composed of superconducting YBa2_{2}Cu3_{3}O7_{7} (YBCO) and ferromagnetic La0.67_{0.67}% Ca0.33_{0.33}MnO3_{3} (LCMO) has been investigated by ellipsometry. A drastic decrease of the free carrier response is observed which involves an unusually large length scale of dcrit^{crit}\approx 20 nm in YBCO and dcrit^{crit}\approx 10 nm in LCMO. A corresponding suppression of metallicity is not observed in SLs where LCMO is replaced by the paramagnetic metal LaNiO3_{3}. Our data suggest that either a long range charge transfer from the YBCO to the LCMO layers or alternatively a strong coupling of the charge carriers to the different and competitive kind of magnetic correlations in the LCMO and YBCO layers are at the heart of the observed metal/insulator transition. The low free carrier response observed in the far-infrared dielectric response of the magnetic superconductor RuSr2_{2}GdCu2_{2}O8_{8} is possibly related to this effect

    Three-dimensional conceptual model for service-oriented simulation

    In this letter, we propose a novel three-dimensional conceptual model for an emerging service-oriented simulation paradigm. The model can be used as a guideline or an analytic means to find the potential and possible future directions of the current simulation frameworks. In particular, the model inspects the crossover between the disciplines of modeling and simulation, service-orientation, and software/systems engineering.
