    Designs for generalized linear models with random block effects via information matrix approximations

    The selection of optimal designs for generalized linear mixed models is complicated by the fact that the Fisher information matrix, on which most optimality criteria depend, is computationally expensive to evaluate. Our focus is on the design of experiments for likelihood estimation of parameters in the conditional model. We provide two novel approximations that substantially reduce the computational cost of evaluating the information matrix by complete enumeration of response outcomes, or Monte Carlo approximations thereof: (i) an asymptotic approximation which is accurate when there is strong dependence between observations in the same block; (ii) an approximation via Kriging interpolators. For logistic random intercept models, we show how interpolation can be especially effective for finding pseudo-Bayesian designs that incorporate uncertainty in the values of the model parameters. The new results are used to provide the first evaluation of the efficiency, for estimating conditional models, of optimal designs from closed-form approximations to the information matrix derived from marginal models. It is found that correcting for the marginal attenuation of parameters in binary-response models yields much improved designs, typically with very high efficiencies. However, in some experiments exhibiting strong dependence, designs for marginal models may still be inefficient for conditional modelling.     Block designs for experiments with non-normal response

    Many experiments measure a response that cannot be adequately described by a linear model withnormally distributed errors and are often run in blocks of homogeneous experimental units. Wedevelop the first methods of obtaining efficient block designs for experiments with an exponentialfamily response described by a marginal model fitted via Generalized Estimating Equations. Thismethodology is appropriate when the blocking factor is a nuisance variable as, for example, occursin industrial experiments. A D-optimality criterion is developed for finding designs robust to thevalues of the marginal model parameters and applied using three strategies: unrestricted algorithmicsearch, use of minimum-support designs, and blocking of an optimal design for the correspondingGeneralized Linear Model. Designs obtained from each strategy are critically compared and shownto be much more efficient than designs that ignore the blocking structure. The designs are comparedfor a range of values of the intra-block working correlation and for exchangeable, autoregressive andnearest neighbor structures. An analysis strategy is developed for a binomial response that allows es-timation from experiments with sparse data, and its efectiveness demonstrated.     High efficiency infrared generation by difference-frequency mixing using tangential phase matching

    Measurements have been made of the energy generated in the wavelength region 3.3 to 4.0 µm, by difference-frequency mixing between a tunable dye laser and a Nd:YAG laser in lithium niobate. The highest mixing efficiencies have been observed with the beams aligned in a non-collinear geometry. It is demonstrated that this non-collinear geometry corresponds to the tangential phase-matching condition which increases the acceptance angle of a second-order nonlinear interaction over that achieved in a non-critically phase-matched geometry

    Human annoyance, acceptability and concern as responses to vibration from the construction of light rapid transit lines in residential environments

    The aim of this paper is to investigate the use of different self-reported measures for assessing the human response to environmental vibration from the construction of an urban LRT (Light Rapid Transit) system. The human response to environmental stressors such as vibration and noise is often expressed in terms of exposure–response relationships that describe annoyance as a function of the magnitude of the vibration. These relationships are often the basis of noise and vibration policy and the setting of limit values. This paper examines measures other than annoyance by expressing exposure–response relationships for vibration in terms of self-reported concern about property damage and acceptability. The exposure–response relationships for concern about property damage and for acceptability are then compared with those for annoyance. It is shown that concern about property damage occurs at vibration levels well below those where there is any risk of damage. Earlier research indicated that concern for damage is an important moderator of the annoyance induced. Acceptability, on the other hand, might be influenced by both annoyance and concern, as well as by other considerations. It is concluded that exposure–response relationships expressing acceptability as a function of vibration exposure could usefully complement existing relationships for annoyance in future policy decisions regarding environmental vibration.     The composition of the protosolar disk and the formation conditions for comets

    Conditions in the protosolar nebula have left their mark in the composition of cometary volatiles, thought to be some of the most pristine material in the solar system. Cometary compositions represent the end point of processing that began in the parent molecular cloud core and continued through the collapse of that core to form the protosun and the solar nebula, and finally during the evolution of the solar nebula itself as the cometary bodies were accreting. Disentangling the effects of the various epochs on the final composition of a comet is complicated. But comets are not the only source of information about the solar nebula. Protostellar disks around young stars similar to the protosun provide a way of investigating the evolution of disks similar to the solar nebula while they are in the process of evolving to form their own solar systems. In this way we can learn about the physical and chemical conditions under which comets formed, and about the types of dynamical processing that shaped the solar system we see today. This paper summarizes some recent contributions to our understanding of both cometary volatiles and the composition, structure and evolution of protostellar disks.Comment: To appear in Space Science Reviews.     A validation study of the CarerQol instrument in informal caregivers of people with dementia from eight European countries

    Purpose: Informal care constitutes an important part of the total care for people with dementia. Therefore, the impact of the syndrome on their caregivers as well as that of health and social care services for people with dementia should be considered. This study investigated the convergent and clinical validity of the CarerQol instrument, which measures and values the impact of providing informal care, in a multi-country sample of caregivers for people with dementia. Methods: Cross-sectional data from a sample of 451 respondents in eight European countries, collected by the Actifcare project, were evaluated. Convergent validity was analysed with Spearman's correlation coefficients and multivariate correlations between the CarerQol-7D utility score and dimension scores, and other similar quality of life measures such as CarerQol-VAS, ICECAP-O, and EQ-5D. Clinical validity was evaluated by bivariate and multivariate analyses of the degree to which the CarerQol instrument can differentiate between characteristics of caregivers, care receivers and caregiving situation. Country dummies were added to test CarerQol score differences between countries. Results: The mean CarerQol utility score was 77.6 and varied across countries from 74.3 (Italy) to 82.3 (Norway). The scores showed moderate to strong positive correlations with the CarerQol-VAS, ICECAP-O, and EQ-5D health problems score of the caregiver. Multivariate regression analysis showed that various characteristics of the caregiver, care receiver and caregiving situation were associated with caregiver outcomes, but there was no evidence of a country-level effect. Conclusion: This study demonstrates the convergent and clinical validity of the CarerQol instrument to evaluate the impact of providing informal care for people with dementia

    Neutralizing antibody vaccine for pandemic and pre-emergent coronaviruses

    Betacoronaviruses (betaCoVs) caused the severe acute respiratory syndrome (SARS) and Middle East Respiratory Syndrome (MERS) outbreaks, and the SARS-CoV-2 pandemic1–4. Vaccines that elicit protective immunity against SARS-CoV-2 and betaCoVs circulating in animals have the potential to prevent future betaCoV pandemics. Here, we show that macaque immunization with a multimeric SARS-CoV-2 receptor binding domain (RBD) nanoparticle adjuvanted with 3M-052/Alum elicited cross-neutralizing antibody (cross-nAb) responses against batCoVs, SARS-CoV-1, SARS-CoV-2, and SARS-CoV-2 variants B.1.1.7, P.1, and B.1.351. Nanoparticle vaccination resulted in a SARS-CoV-2 reciprocal geometric mean neutralization ID50 titer of 47,216, and protection against SARS-CoV-2 in macaque upper and lower respiratory tracts. Importantly, nucleoside-modified mRNA encoding a stabilized transmembrane spike or monomeric RBD also induced SARS-CoV-1 and batCoV cross-nAbs, albeit at lower titers. These results demonstrate current mRNA vaccines may provide some protection from future zoonotic betaCoV outbreaks, and provide a platform for further development of pan-betaCoV vaccines
