    Reakcija potkožnog vezivnog tkiva na materijale za definitivno punjenje kanala korena

    The aim of the study was to evaluate tissue response of two root canal sealers: Endomethasone (cink oxide eugenol-based sealers) and Ketac Endo Aplicap (glass ionomer based sealers). The sealers were freshly mixed and injected into subcutaneous connective tissue of the dorsal surface of experimental animals (Wistar Furth rats). The inflammatory reaction caused by the sealers was evaluated 7, 30 and 60 days after implantation using descriptive hisopathological analysis. The tissue sections were taken from injection sites. Each incision included skin,subcutaneous connective tissue and underlying muscular tissue. All blocks were proceeded using standard histological procedures. The tissue reactions were graded as mild,moderate or severe inflammation. After the seven days both sealers showed a moderate reaction with a decreasing trend. Both materials caused mild inflammation which diminished at the end of the experimental period. The results of this study demonstrate that Ketac Endo and Endomethasone is well tolerated by subcutaneus connective tissue of experimental animals.Cilj ovih proučavanja je bio ispitivanje biokompatibilnosti dva materijala za definitivno kanalno punjenje: Endomethasone (materijal na bazi cink oksid eugenola) i Ketac Endo Aplocap (glas jonomer cementa za kanalno punjenje). Sveže pripremljeni materijali su injicirani u potkožno vezivno tkivo leđnog dela eksperimentalnih životinja (pacovi Wistar soja). Intenzitet inflamatorne reakcije proveravan je nakon 7, 30 i 60 dana na osnovu patohistoloÅ”ke analize. Isečak je uzet sa mesta aplikacije materijala i obuhvatao je deo kože, potkožno tkivo i deo miÅ”ićnog sloja. Tkiva su podvrgnuta standardnoj histoloÅ”koj obradi. Reakcija tkiva na ubačeni materijal označena je kao blaga, srednja i jako izražena inflamacija. Posle prvog perioda od 7 dana oba materijala su izazvala intenzivnu zapaljensku reakciju tkiva sa kasnijom tendencom smanjenja inflamacije. Oba materijala su izazvala umereno zapaljenje koje je nestalo do kraja eksperimentalnog perioda. Postignuti rezultati ukazuju da su Ketac Endo i Endomethasone biokompatibilni u potkožnom vezivnom tkivu eksperimentalnih životinja

    Influence of rhamnolipids, produced by Pseudomonas aeruginosa NCAIM(P), B001380 on Cr(VI) removal capacity in liquid medium

    Pseudomonas aeruginosa NCAIM(P), B001380, a propitious bacterial strain isolated from mineral cutting oil was identified to be chromium tolerant and a producer of biosurfactant rhamnolipid (RL) with potential application in heavy metal bioremediation. Culture growth, RL production and Cr(VI) removal capacity of the strain in the presence of 50 mg L-1 (I) and 100 mg L-1 of Cr(VI) (II) were studied. Maximum of RL production were found in the late-stationary phase at 72 h for both Cr(VI)-amended cultures: I (236 mg L-1) and II (160 mg L-1), as well as the maximum of Cr(VI) removal capacity: 70 % (I) and 57 % (II). The amount of Cr in RL preparation II was 22 mg mg-1 determined by flame atomic absorption spectroscopy (FAAS). Appearance of a new band at 914 cm-1 in infrared (IR) spectrum of RL (II) indicated a significant proof for possible coordination of CrO42-ion with RL. The effect of Cr(VI) on monorhamnolipids (RL1) and dirhamnolipids (RL2) distribution and its ratio were studied by electrospray ionization mass spectrometry (ESI-MS).     The influence of chemical characteristics of precipitation on tree health in Banjica Forest (Belgrade, Serbia)

    The most represented tree species in the Banjica Forest are Acer negundo, Quercus robur, Acer pseudoplatanus, Populus nigra, Fraxinus pennsylvanica, Fraxinus ornus and Robinia pseudoacacia. According to the ICP Forests combined assessment (degree of defoliation and decolorization), endangered species are Populus nigra (64.3% of heavily damaged trees), Quercus robur (45.5%), Fraxinus pennsylvanica (37.0%) and Acer negundo (26.6%), while the situation is much better for Acer pseudoplatanus and Fraxinus ornus. For Robinia pseudoacacia, 83% of trees are without decolorization, however, defoliation is established. In the period from April to October 2009, the average pH of rainwater was 5.46, and 5.18 in the period from November 2009 to March 2010. The concentration of SO42- in the period from April to October 2009 amounted to an average of 24.21 mg/l, and 28.87 mg/l in the period from November 2009 to March 2010. The concentration of SO42- and pH values is a possible explanation for the condition of the trees. [Acknowledgments.     Polyunsaturated fatty acids (PUFAs) are necessary for overall health. Two PUFAs families, n-6 and n-3 fatty acids, are physiologically and metabolically distinct. The proportion of PUFAs in serum and erythrocyte phospholipids, which depends on endogenous metabolism controlled by genetic polymorphisms and dietary intake, is an important determinant of both health and disease. Both n-3 and n-6 PUFAs are processed to powerful promoters of eicosanoids synthesis at the cyclooxygenase and lipoxygenase level. Evidence from observational and intervention studies suggest that n-3 PUFAs are cardio-protective, perhaps through their anti-inflammatory, anti-arrhythmic, lipid-lowĀ­ering and antihypertensive effects. In contrast, dietary n-6 PUFAs have pro-inflammatory effects. Low n-3 and elevated n-6 PUFAs levels were found in patients with cancer on different sites. The present review focuses on current knowledge related to PUFAs intake and status in health and disease, with reference to the Serbian population.Polinezasićene masne kiseline (PMK) su neophodne za normalno funkcionisanje organizma. Dve PMK familije, n-6 i n-3 masne kiseline se fizioloÅ”ki i metabolički razlikuju. Udeo PMK u fosfolipidima seruma i eritrocita je važan pokazatelj zdravlja i bolesti, i zavisi od endogenog metabolizma, koji je kontrolisan genetskim polimorfizmom, i unosa hrane. I n-6 i n-3 PMK su prekursori za sintezu eikozanoida na ciklooksigenaznom i lipooksigenaznom nivou. Opservacione i interventne studije ukazuju da n-3 PMK imaju kardioprotektivni efekat, delujući anti-inflamatorno, anti-aritmogeno, hipolipidemično i antihipertenzivno. Nasuprot tome, smatra se da n-6 PMK imaju pro-inflamatorno dejstvo. Nizak nivo n-3 i poviÅ”en udeo n-6 PMK je pokazan kod pacijenata sa različitim tipovima maligniteta.     Potential preservation of dental pulp stem cells

    Dental pulp stem cells (DPSCs) as postnatal stem cells have recently been described. They are clonogenic cells, capable for self-renewal with high proliferative potential. Their multilineage potential and plasticity enables their differentiation into different kind of cells, such as osteoblasts, chondrocytes, adipocytes, muscle cells, neural cells, odontoblasts, cementoblasts and ameloblasts. DPSCs are an important human stem cells source, especially in patients who lost their chance for umbilical cord blood isolation and preservation. As these cells became useful for tissue engineering and cell therapy, proper mode of their preservation also became important. The most important points in the cryopreservation and recovery procedure are: growth phase of harvested cells, number of cells, the proper cryopreservative concentration and serum concentration. The cryopreservation process includes the following general components: harvesting of the cells, addition of cryopreservative, the freezing procedure, the thawing procedure and assessment of the viability prior to transplantation. There is no single and perfect cryopreservation method. Further investigations should be regarding capability of DPSCs and their differentiated cells to recover and restart proliferation, differentiation and new tissue production for therapeutic use after cryopreservation

    Cadmium specific proteomic responses of a highly resistant Pseudomonas aeruginosa san ai

    Pseudomonas aeruginosa san ai is a promising candidate for bioremediation of cadmium pollution, as it resists a high concentration of up to 7.2 mM of cadmium. Leaving biomass of P. aeruginosa san ai exposed to cadmium has a large biosorption potential, implying its capacity to extract heavy metal from contaminated medium. In the present study, we investigated tolerance and accumulation of cadmium on protein level by shotgun proteomics approach based on liquid chromatography and tandem mass spectrometry coupled with bioinformatics to identify proteins. Size exclusion chromatography was used for protein prefractionation to preserve native forms of metalloproteins and protein complexes. Using this approach a total of 60 proteins were observed as up-regulated in cadmium-amended culture. Almost a third of the total numbers of up-regulated were metalloproteins. Particularly interesting are denitrification proteins which are over expressed but not active, suggesting their protective role in conditions of heavy metal exposure. P. aeruginosa san ai developed a complex mechanism to adapt to cadmium, based on: extracellular biosorption, bioaccumulation, the formation of biofilm, controlled siderophore production, enhanced respiration and modified protein profile. An increased abundance of proteins involved in: cell energy metabolism, including denitrification proteins; amino acid metabolism; cell motility and posttranslational modifications, primarily based on thiol-disulfide exchange, were observed. Enhanced oxygen consumption of biomass in cadmium-amended culture versus control was found. Our results signify that P. aeruginosa san ai is naturally well equipped to overcome and survive high doses of cadmium and, as such, has a great potential for application in bioremediation of cadmium polluted sites.Supplementary material: [http://cherry.chem.bg.ac.rs/handle/123456789/3179
