Evaluation of the onset time and intubation conditions of rocuronium bromide in children
Publisher's copy made available with the permission of the publisher © 1997 Australian Society of AnaesthetistsWe have assessed, in children aged three to eight years, the intubating conditions after administration of rocuronium 0.6 mg/kg at 50 or 60 seconds, in groups of 15 patients. Intubating conditions were excellent in 11, good in 3 and fair in 1 patient at 50 seconds and excellent in 12 and good in 3 patients at 60 seconds. The mean onset time, for all patients, to when the first twitch of the train of four (T1), measured at the adductor pollicis, was depressed to less than 30% and 5% of control was 50 (SD 11.4) seconds and 94 (SD 31.7) seconds respectively. Depression of T1 to less than 30% of control, measured at the adductor pollicis in children, appears to indicate that intubating conditions will be clinically acceptable when using rocuronium.P.F. Structure of the PII signal transduction protein of Neisseria meningitidis at 1.85 Ȃ resolution
Copyright @ 2006 International Union of CrystallographyThe PII signal transduction proteins GlnB and GlnK are implicated in the regulation of nitrogen assimilation in Escherichia coli and other enteric bacteria. PII-like proteins are widely distributed in bacteria, archaea and plants. In contrast to other bacteria, Neisseria are limited to a single PII protein (NMB 1995), which shows a high level of sequence identity to GlnB and GlnK from Escherichia coli (73 and 62%, respectively). The structure of the PII protein from N. meningitidis (serotype B) has been solved by molecular replacement to a resolution of 1.85 Ȃ. Comparison of the structure with those of other PII proteins shows that the overall fold is tightly conserved across the whole population of related proteins, in particular the positions of the residues implicated in ATP binding. It is proposed that the Neisseria PII protein shares functions with GlnB/GlnK of enteric bacteria.This study is funded by the Medical Research Council UK and Europe (SPINE) consortium (European Commission Grant No. QLG2-CT-2002-00988)
Interpretation and Knowledge Maximization
Timothy Williamson has proposed that we should give a ‘knowledge first’ twist to David Lewis’s account of content, maintaining that for P to be the content of one’s belief is for P to be the content that would be attributed by an idealized interpreter working under certain constraints, and that the fundamental constraint on interpretation is a principle of knowledge maximization. According to this principle, an interpretation is correct to the extent that it maximizes the number of knowledgeable judgments the subject comes out as making. Here I will argue against knowledge maximization and two fallback positions suggested by Williamson’s discussion. Williamson intends the principle of knowledge maximization to form the basis of an argument against a certain sort of skepticism about judgment. In the final section I argue that the kind of general response to judgment skepticism envisaged by Williamson is neither desirable nor necessary
Crystal structure of nitrogen regulatory protein IIA from
BACKGROUND: The NMB0736 gene of Neisseria meningitidis serogroup B strain MC58 encodes the putative nitrogen regulatory protein, IIANtr (abbreviated to NM-IIANtr). The homologous protein present in Escherichia coli is implicated in the control of nitrogen assimilation. As part of a structural proteomics approach to the study of pathogenic Neisseria spp., we have selected this protein for structure determination by X-ray crystallography. RESULTS: The NM-IIANtr was over-expressed in E. coli and was shown to be partially mono-phosphorylated, as assessed by mass spectrometry of the purified protein. Crystals of un-phosphorylated protein were obtained and diffraction data collected to 2.5 A resolution. The structure of NM-IIANtr was solved by molecular replacement using the coordinates of the E. coli nitrogen regulatory protein IIAntr [PDB: 1A6J] as the starting model. The overall fold of the Neisseria enzyme shows a high degree of similarity to the IIANtr from E. coli, and the position of the phosphoryl acceptor histidine residue (H67) is conserved. The orientation of an adjacent arginine residue (R69) suggests that it may also be involved in coordinating the phosphate group. Comparison of the structure with that of E. coli IIAmtl complexed with HPr [PDB: 1J6T] indicates that NM-IIANtr binds in a similar way to the HPr-like enzyme in Neisseria. CONCLUSION: The structure of NM-IIANtr confirms its assignment as a homologue of the IIANtr proteins found in a range of other Gram-negative bacteria. We conclude that the NM- IIANtr protein functions as part of a phosphorylation cascade which, in contrast to E. coli, shares the upstream phosphotransfer protein with the sugar uptake phosphoenolpyruvate:sugar phosphotransferase system (PTS), but in common with E. coli has a distinct downstream effector mechanism
The economic and innovation contribution of universities: a regional perspective
Universities and other higher education institutions (HEIs) have come to be regarded as key sources of knowledge utilisable in the pursuit of economic growth. Although there have been numerous studies assessing the economic and innovation impact of HEIs, there has been little systematic analysis of differences in the relative contribution of HEIs across regions. This paper provides an exploration of some of these differences in the context of the UK’s regions. Significant differences are found in the wealth generated by universities according to regional location and type of institution. Universities in more competitive regions are generally more productive than those located in less competitive regions. Also, traditional universities are generally more productive than their newer counterparts, with university productivity positively related to knowledge commercialisation capabilities. Weaker regions tend to be more dependent on their universities for income and innovation, but often these universities under-perform in comparison to counterpart institutions in more competitive regions. It is argued that uncompetitive regions lack the additional knowledge infrastructure, besides universities, that are more commonly a feature of more competitive regions
Squirrelpox virus: assessing prevalence, transmission and environmental degradation
Red squirrels (Sciurus vulgaris) declined in Great Britain and Ireland during the last century, due to habitat loss and the introduction of grey squirrels (Sciurus carolinensis), which competitively exclude the red squirrel and act as a reservoir for squirrelpox virus (SQPV). The disease is generally fatal to red squirrels and their ecological replacement by grey squirrels is up to 25 times faster where the virus is present. We aimed to determine: (1) the seropositivity and prevalence of SQPV DNA in the invasive and native species at a regional scale; (2) possible SQPV transmission routes; and, (3) virus degradation rates under differing environmental conditions. Grey (n = 208) and red (n = 40) squirrel blood and tissues were sampled. Enzyme-linked immunosorbent assay (ELISA) and quantitative real-time polymerase chain reaction (qPCR) techniques established seropositivity and viral DNA presence, respectively. Overall 8% of squirrels sampled (both species combined) had evidence of SQPV DNA in their tissues and 22% were in possession of antibodies. SQPV prevalence in sampled red squirrels was 2.5%. Viral loads were typically low in grey squirrels by comparison to red squirrels. There was a trend for a greater number of positive samples in spring and summer than in winter. Possible transmission routes were identified through the presence of viral DNA in faeces (red squirrels only), urine and ectoparasites (both species). Virus degradation analyses suggested that, after 30 days of exposure to six combinations of environments, there were more intact virus particles in scabs kept in warm (25°C) and dry conditions than in cooler (5 and 15°C) or wet conditions. We conclude that SQPV is present at low prevalence in invasive grey squirrel populations with a lower prevalence in native red squirrels. Virus transmission could occur through urine especially during warm dry summer conditions but, more notably, via ectoparasites, which are shared by both species
Radial velocity eclipse mapping of exoplanets
Planetary rotation rates and obliquities provide information regarding the
history of planet formation, but have not yet been measured for evolved
extrasolar planets. Here we investigate the theoretical and observational
perspective of the Rossiter-McLauglin effect during secondary eclipse (RMse)
ingress and egress for transiting exoplanets. Near secondary eclipse, when the
planet passes behind the parent star, the star sequentially obscures light from
the approaching and receding parts of the rotating planetary surface. The
temporal block of light emerging from the approaching (blue-shifted) or
receding (red-shifted) parts of the planet causes a temporal distortion in the
planet's spectral line profiles resulting in an anomaly in the planet's radial
velocity curve. We demonstrate that the shape and the ratio of the
ingress-to-egress radial velocity amplitudes depends on the planetary
rotational rate, axial tilt and impact factor (i.e. sky-projected planet
spin-orbital alignment). In addition, line asymmetries originating from
different layers in the atmosphere of the planet could provide information
regarding zonal atmospheric winds and constraints on the hot spot shape for
giant irradiated exoplanets. The effect is expected to be most-pronounced at
near-infrared wavelengths, where the planet-to-star contrasts are large. We
create synthetic near-infrared, high-dispersion spectroscopic data and
demonstrate how the sky-projected spin axis orientation and equatorial velocity
of the planet can be estimated. We conclude that the RMse effect could be a

The Relative Importance of Different Trophic Pathways for Secondary Exposure to Anticoagulant Rodenticides
Secondary exposure of predators to anticoagulant rodenticides, and in particular second generation anticoagulant rodenticides (SGARs), is a global phenomenon. The widespread and large-scale nature of this exposure has attracted considerable concern, although the consequences in terms of likelihood of poisoning of individuals and resultant impacts on populations are not well characterised. Secondary exposure of predators may as rise from once or more of: (i) eating contaminated commensal rodents subject to control (target species are typically rats and house mice); (ii) consumption of contaminated non-target small mammals (such as Peromyscus, Microtus, and Apodemus species) that encounter and feed on what are rodent-attractive baits; (iii) consumption of non-rodent vertebrate and invertebrate prey that may also incidentally encounter and eat baits. We hypothesised that predators feeding primarily on target species may be most at risk of exposure to SGARs while those predominantly taking non-mammalian prey may be at least risk. We tested this hypothesis by comparing exposure, determined from the presence and magnitude of SGAR liver residues, in red kites (Milvus milvus), which feeds extensively on rats, in barn owls (Tyto alba), kestrels (Falco tinnunculus), and tawny owls (Strix aluco) that feed widely on non-target small mammals, and in sparrowhawks (Accipiter nisus) that feed predominantly on small birds. We found that the scale and magnitude of exposure was broadly consistent with our hypothesis, and that controlling for age in the analysis could be important as older birds can accumulate residues with age. However, exposure in kestrels was typically greater than that in barn owls and tawny owls, despite what is thought to be a general similarity among the species in their diets. We discuss the relative importance of trophic pathways relative to other factors that may drive secondary exposure in predators, and confirm that species that feed on rats or other target species may be at most risk of exposure and poisoning
